Alginate-based microcapsules for immunoisolation of pancreatic islets

The enormous variety of polysaccharides that can be extracted from marine plants and animal organisms or produced by marine bacteria means that the field of marine polysaccharides is constantly evolving. Recent advances in biological techniques allow high levels of polysaccharides of interest to be produced in vitro. Biotechnology is a powerful tool to obtain polysaccharides from a variety of micro-organisms, by controlling the growth conditions in a bioreactor while tailoring the production of biologically active compounds. Following an overview of the current knowledge on marine polysaccharides, with special attention to potential pharmaceutical applications and to more recent progress on the discovering of new polysaccharides with biological appealing characteristics, this review will focus on possible strategies for chemical or physical modification aimed to tailor the final properties of interest.

The first experimental finding of supercooling in a polysaccharide is reported. Electron micrographs of schizophyllan, obtained from samples for which the native linear triple helix had been denatured in DMSO by high temperature or at high pH and subsequently renatured revealed circular and linear morphologies as well as larger, more complex clusters.

Supercoiling in circular triple-helical polysaccharides

Double-stranded xanthan was depolymerized by partial acid hydrolysis to produce samples where M w ranged from 6×10 6 to 7×10 4 . These were characterized with respect to chemical composition in the side chains, molecular weight, molecular weight distribution, intrinsic viscosity, and conformation (optical rotation). Additional information about molecular size and conformation was obtained by electron microscopy. For each sample a fraction consisting of depolymerized, double-stranded species could be obtained. Their conformational properties were largely analogous to undegraded xanthan, despite the reduced M w and the changes in the side chains

Depolymerization of double-stranded xanthan by acid hydrolysis: characterization of partially degraded double strands and single-stranded oligomers released from the ordered structures

The depolymerization of double-stranded xanthan and triple-stranded scleroglucan is simulated using a Monte Carlo method. Chain scissions are introduced randomly in each strand to simulate chemical degradation. The simulations show that degradation of such polymers deviates significantly from that of single-stranded polymers. There is initially an apparently enheanced stability where scission of the glycosidic linkages is effectively masked because of interstrand stabilization. This period is followed by a power law region, M w ∼ t -ν
