The Twentieth Century Reanalysis (20CR) project is an international effort to produce a comprehensive global atmospheric circulation dataset spanning the twentieth century, assimilating only surface pressure reports and using observed monthly sea-surface temperature and sea-ice distributions as boundary conditions. It is chiefly motivated by a need to provide an observational dataset with quantified uncertainties for validations of climate model simulations of the twentieth century on all time-scales, with emphasis on the statistics of daily weather. It uses an Ensemble Kalman Filter data assimilation method with background ‘first guess’ fields supplied by an ensemble of forecasts from a global numerical weather prediction model. This directly yields a global analysis every 6 hours as the most likely state of the atmosphere, and also an uncertainty estimate of that analysis.

The 20CR dataset provides the first estimates of global tropospheric variability, and of the dataset's time-varying quality, from 1871 to the present at 6-hourly temporal and 2° spatial resolutions. Intercomparisons with independent radiosonde data indicate that the reanalyses are generally of high quality. The quality in the extratropical Northern Hemisphere throughout the century is similar to that of current three-day operational NWP forecasts. Intercomparisons over the second half-century of these surface-based reanalyses with other reanalyses that also make use of upper-air and satellite data are equally encouraging.

It is anticipated that the 20CR dataset will be a valuable resource to the climate research community for both model validations and diagnostic studies. Some surprising results are already evident. For instance, the long-term trends of indices representing the North Atlantic Oscillation, the tropical Pacific Walker Circulation, and the Pacific–North American pattern are weak or non-existent over the full period of record. The long-term trends of zonally averaged precipitation minus evaporation also differ in character from those in climate model simulations of the twentieth century. Certain insulators have exotic metallic states on their surfaces. These states are formed by topological effects that also render the electrons travelling on such surfaces insensitive to scattering by impurities. Such topological insulators may provide new routes to generating novel phases and particles, possibly finding uses in technological applications in spintronics and quantum computing.

The birth of topological insulators

Substantial progress has been made toward understanding the genetic architecture, cellular substrates, brain circuits and endophenotypic profiles of neuropsychiatric disorders, including autism spectrum disorders (ASD), schizophrenia and Alzheimer's disease. Recent evidence implicates spiny synapses as important substrates of pathogenesis in these disorders. Although synaptic perturbations are not the only alterations relevant for these diseases, understanding the molecular underpinnings of spine pathology may provide insight into their etiologies and may reveal new drug targets. Here we discuss recent neuropathological, genetic, molecular and animal model studies that implicate structural alterations at spiny synapses in the pathogenesis of major neurological disorders, focusing on ASD, schizophrenia and Alzheimer's disease as representatives of these categories across different ages of onset. We stress the importance of reverse translation, collaborative and multidisciplinary approaches, and the study of the spatio-temporal roles of disease molecules in the context of synaptic regulatory pathways and neuronal circuits that underlie disease endophenotypes.

Dendritic spine pathology in neuropsychiatric disorders

The first cancer-targeted microRNA (miRNA) drug - MRX34, a liposome-based miR-34 mimic - entered Phase I clinical trials in patients with advanced hepatocellular carcinoma in April 2013, and miRNA therapeutics are attracting special attention from both academia and biotechnology companies. Although miRNAs are the most studied non-coding RNAs (ncRNAs) to date, the importance of long non-coding RNAs (lncRNAs) is increasingly being recognized. Here, we summarize the roles of miRNAs and lncRNAs in cancer, with a focus on the recently identified novel mechanisms of action, and discuss the current strategies in designing ncRNA-targeting therapeutics, as well as the associated challenges.

MicroRNAs and other non-coding RNAs as targets for anticancer drug development

Lithium metal is one of the most attractive anode materials for electrochemical energy storage. However, the growth of Li dendrites during electrochemical deposition, which leads to a low Coulombic efficiency and safety concerns, has long hindered the application of rechargeable Li-metal batteries. Here we show that a 3D current collector with a submicron skeleton and high electroactive surface area can significantly improve the electrochemical deposition behaviour of Li. Li anode is accommodated in the 3D structure without uncontrollable Li dendrites. With the growth of Li dendrites being effectively suppressed, the Li anode in the 3D current collector can run for 600 h without short circuit and exhibits low voltage hysteresis. The exceptional electrochemical performance of the Li-metal anode in the 3D current collector highlights the importance of rational design of current collectors and reveals a new avenue for developing Li anodes with a long lifespan.

The various contributions to this book record the remarkable progress that has been made in the understanding and management of pancreatic diseases over the last decade. The improvement in understanding has led in some cases to better management with improved outcome for the patient, whereas in other areas the way is now clear towards a better prospect for the future.
