Seed banking is now widely used for the ex situ conservation of wild plant species. Many seed banks that conserve wild species broadly follow international genebank guidelines for seed collection, processing, storage, and management. However, over the last 10-20 years, problems and knowledge gaps have been identified, which have led to more focused seed conservation research on diverse species. For example, there is now greater ecogeographic understanding of seed storage behaviour and of the relative longevity of orthodox seeds, and we are therefore able to predict which species should be conserved using cryostorage techniques; seed development studies have identified when seeds should be harvested for maximal tolerance of desiccation and longevity in storage, as well as highlighting how seed development can vary between species; and there is now a wealth of literature on the dormancy-breaking and germination requirements of wild species which, as well as enabling better management of accessions, will also mean that their use in restoration, species reintroduction, or for evaluation for other applications is possible. Future research may be focused, for example, on nursery and plant production systems for wild plant species that maximize genetic diversity, so that introduced seeds and plant materials have the resilience to cope with future environmental stresses.

Advances in seed conservation of wild plant species: a review of recent research

Plasma membrane (PM) recovery from the impaired dry state is essential for seed germination, but its underlying mechanism remains unclear. In this study, we found that ZmANN33 and ZmANN35, two annexin genes in maize, encode proteins that participate in PM recovery during seed germination. The expression of both genes was up-regulated during seed germination and strongly repressed by chilling (either 15 or 5 °C) as compared with the normal temperature (25 °C). In addition, the increased membrane damage caused by chilling imbibition was correlated with suppressed expression of ZmANN33 and ZmANN35, while rapid recovery of their expression levels accompanied the rescue of the damaged membrane. Arabidopsis seedlings ectopically expressing ZmANN33 or ZmANN35 had longer seedling length than wild-type (WT) plants during the recovery period after 3 d of chilling stress, indicating the positive roles of these two gene products in the plant’s recovery from chilling injury. Moreover, these transgenic seedlings had lower lipid peroxidation and higher peroxidase activities than WT during the recovery period. Consistently, root cells of these transgenic seedlings had more intact PM after chilling stress, supporting the proposition that ZmANN33 and ZmANN35 contribute to the maintenance of PM integrity. The enhanced PM integrity is likely due to the accelerated exocytotic process after chilling stress. We also showed that both ZmANN33 and ZmANN35 localized in the cytosol near the plasma membrane. Thus, we conclude that ZmANN33 and ZmANN35 play essential roles in membrane recovery during maize seed germination.

Nutrient seed priming is a strategy to increase the seed reserves of mineral nutrients as primary source for mineral nutrition during seedling development and early growth. The present study investigates the effects of zinc (Zn) and manganese (Mn) seed priming on growth and nutritional status of soybean under conditions of Zn and Mn limitation. Nutrient seed priming increased the natural seed reserves for Zn by, approximately, sixfold and by fivefold for Mn; however, 40–60% of the primed nutrients were adsorbed to the seed coat. Zinc seed priming was able to maintain plant growth for 5 weeks in the same way as Zn supply via the nutrient solution. It is concluded that nutrient seed priming offers perspectives to improve seed quality of soybean for early seedling development under limited nutrient supply or availability and needs further investigation on performance under various stress conditions.

Accumulation and distribution of Zn and Mn in soybean seeds after nutrient seed priming and its contribution to plant growth under Zn- and Mn-deficient conditions

A laboratory experiment was conducted to determine the effect of ultrasound (US) exposure time on germination behavior of sesame seeds. All tests were carried out at 20 kHz in a water bath ultrasonic device varying two factors, treatment duration (10, 20 and 30 min) and germination temperature (15, 20 and 25 oC). Parallel tests were run in which seeds were soaked in water without sonication in order to eliminate the effect of water from US test results. US treatments enhanced seeds water uptake. At mild exposure time it improved sesame seed germination performance and seedling growth at suboptimal temperatures as indicated by higher germination percentage and germination rate. US applying for 20 min had relatively high superoxide dismutase activity; however, had not significant differences with control and US duration for 10 min. The catalase activity was strongly increased by applying the US for a 10 and 20 min. Among the treatments, application of US vibration for 10 and 20 min reduced both of malondialdehyde and H 2 O 2 contents, however high US duration (30 min) increased both of the traits. In general, ultrasonic priming technique can be useful for early planting the sesame seeds, and lead to higher yields.

Sonication of seeds increase germination performance of sesame under low temperature stress

Etching of the seed cuticle by cold plasma shortens imbibitional leakage in Linum usitatissimum L.

Low and high moisture kernels and embryos were imbibed by soaking in distilled water at 5°C and 25°C. Leaching of seeds and embryos was measured by ion conductivity of imbibition water after fixed time intervals. Imbibed kernels were placed on moist filter paper after 1 h imbibition and embryos on a solid culture medium after 15 min imbibition and germinated at 25°C. Primary root length was measured 3 days after germination.The leakage from seeds or embryos was correlated with the velocity of water uptake but did not show its highest values when low moisture seeds or embryos were imbibed at chilling temperature. Furthermore germination was apparently not affected by imbibitional chilling of either low moisture content seeds or embryos. In one of the three genotypes tested (B73) seedlings from low moisture content seeds imbibed at 5°C showed a reduction of the primary root growth. However, this reduction was also seen in seedlings from low moisture B73 seeds imbided at 25°C and resulting in higher level of leaching. Thus, injury by imbibition of low moisture seeds appears independent of chilling temperature and not due to loss of semipermeability of membrane lipids. Our results suggest that, in the case of embryos, water inrush into the cells reaches levels injurious for membrane integrity independently of the two moisture contents and temperature considered.
