Despite the many advancements in the pharmaceutical and medical fields and the development of numerous antimicrobial drugs aimed to suppress and destroy pathogenic microorganisms, infectious diseases still represent a major health threat affecting millions of lives daily. In addition to the limitations of antimicrobial drugs associated with low transportation rate, water solubility, oral bioavailability and stability, inefficient drug targeting, considerable toxicity, and limited patient compliance, the major cause for their inefficiency is the antimicrobial resistance of microorganisms. In this context, the risk of a pre-antibiotic era is a real possibility. For this reason, the research focus has shifted toward the discovery and development of novel and alternative antimicrobial agents that could overcome the challenges associated with conventional drugs. Nanotechnology is a possible alternative, as there is significant evidence of the broad-spectrum antimicrobial activity of nanomaterials and nanoparticles in particular. Moreover, owing to their considerable advantages regarding their efficient cargo dissolving, entrapment, encapsulation, or surface attachment, the possibility of forming antimicrobial groups for specific targeting and destruction, biocompatibility and biodegradability, low toxicity, and synergistic therapy, polymeric nanoparticles have received considerable attention as potential antimicrobial drug delivery agents. In this context, the aim of this paper is to provide an up-to-date overview of the most recent studies investigating polymeric nanoparticles designed for antimicrobial therapies, describing both their targeting strategies and their effects.

Polymeric Nanoparticles for Antimicrobial Therapies: An Up-To-Date Overview.

The interest in nano-sized materials to develop novel products has increased exponentially in the last decade, together with the search for green methods for their synthesis. An alternative to contribute to a more sustainable approach is the use of microbial-derived molecules to assist nanomaterial synthesis. In this sense, biosurfactants (BSs) have emerged as eco-friendly substitutes in nano-sized materials preparation. The inherent amphiphilic and self-assembly character of BSs associated with their low eco-toxicity, biodegradability, biocompatibility, structural diversity, biological activity, and production from renewable resources are potential advantages over chemically-derived surfactants. In nanotechnology, these versatile molecules play multiple roles. In nanoparticle (NP) synthesis, they act as capping and reducing agents and they also provide self-assembly structures to encapsulation, functionalization, or templates and act as emulsifiers in nanoemulsions. Moreover, BSs can also play as active compounds owing to their intrinsic biological properties. This review presents the recent trends in the development of BS-based nanostructures and their biomedical and environmental applications. Fundamental aspects regarding their antimicrobial and anticancer activities are also discussed.

Microbial surfactants in nanotechnology: recent trends and applications.

Biosurfactants have been investigated as potential alternatives for synthetic surfactants in several areas, for example, in environmental and pharmaceutical fields. In that regard, extensive research has been carried out with sophorolipids and rhamnolipids that also present various biological properties with therapeutic significance. These biosurfactants are obtained as complex mixtures of slightly different molecules, and thus when studying these microbial glycolipids, the ability to identify and purify the produced compounds is of extreme importance. This study aimed to develop improved methodologies for the identification, separation, and purification of sophorolipids and rhamnolipids. Therefore, an ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was modified to ensure faster characterization of both sophorolipids and rhamnolipids, enabling the identification and fragmentation pattern description of 10 and 13 congeners, respectively. The separation and purification of these biosurfactants was achieved with novel reversed-phase solid-phase extraction methods guaranteeing the isolation of different glycolipids, including those considered for their significant biological activity (e.g. antimicrobial, anticancer). It was possible to isolate sophorolipids and rhamnolipids with purity of 94% and 99%, respectively. The methods presented herein can be easily implemented and are expected to make purification of these biosurfactants easier, facilitating the study of their individual properties in further works.
