The production of metal-organic frameworks (MOFs) in the form of colloids has brought a paradigm shift in the design of new functional porous materials. Along with their intrinsic interest as porous solids, and contrary to their bulk powder counterparts, colloidal MOF particles can additionally be dispersed, shaped, functionalized, transformed and assembled in a controlled manner, conferring them further properties and applications. In this regard, zeolitic imidazolate framework-8 (ZIF-8) has become a pioneering MOF constituent of colloidal science. Today, the understanding of the role of synthetic parameters, learned after one decade of research, enables the production of monodisperse colloidal ZIF-8 particles with tunable dimensions and morphologies, offering the opportunity to develop new functional materials and composites with novel and promising functionalities. This tutorial review provides a useful guide to prepare ZIF-8 in its colloidal form, covering the published studies on the synthesis of homogeneous ZIF-8 particles with controlled size and shape. In addition, we present the most relevant advances in the development of colloidal ZIF-8 hybrid single-particles, reflecting the great potential and rapid development of this interdisciplinary research field. Finally, we highlight how formulation of ZIF-8 as colloids has led to the emergence of novel physicochemical phenomena that are useful for practical applications. This review aims at promoting the development of MOFs as colloids, taking ZIF-8 as a pioneering and successful case that clearly shows the benefits of bridging MOF chemistry and colloidal science.

Metal-organic frameworks: A novel platform for combined advanced therapies

Understanding the stability of zeolitic imidazolate framework-8 (ZIF-8) under physiological conditions is critical in biotechnology and biomedicine for biosensing, biocatalysis, and drug delivery. The use of ZIF-8 has shown that this metal organic framework (MOF) and its derived bio-composites can degrade in presence of buffer solutions. Here we present an in-depth analysis of the structural and chemical changes of pure ZIF-8 particles exposed to phosphate buffered saline (PBS) media. Two different particle sizes (2 μm and 250 nm) were selected and the decomposition operated by 10 mM PBS (aka 1X) was studied using powder X-ray diffraction (PXRD), Fourier transformed infrared spectroscopy (FTIR), time resolved atomic force microscopy (AFM), in situ small angle X-ray scattering (SAXS), and 31P NMR.

Degradation of ZIF-8 in phosphate buffered saline media

Covalent organic frameworks (COFs) are an emerging class of organic crystalline polymers with well-defined molecular geometry and tunable porosity. COFs are formed via reversible condensation of lightweight molecular building blocks, which dictate its geometry in two or three dimensions. Among COFs, 2D COFs have garnered special attention due to their unique structure composed of two-dimensionally extended organic sheets stacked in layers generating periodic columnar π-arrays, functional pore space, and their ease of synthesis. These unique features in combination with their low density, high crystallinity, large surface area, and biodegradability have made them an excellent candidate for a plethora of applications ranging from energy to biomedical sciences. In this article, the evolution of 2D COFs is briefly discussed in terms of different types of chemical linkages, synthetic strategies of bulk and nanoscale 2D COFs, and their tunability from a biomedical perspective. Next, the biomedical applications of 2D COFs specifically for drug delivery, phototherapy, biosensing, bioimaging, biocatalysis, and antibacterial activity are summarized. In addition, current challenges and emerging approaches in designing 2D COFs for advanced biomedical applications are discussed.
