During cell division, mitotic spindles are assembled by microtubule-based motor proteins. The bipolar organization of spindles is essential for proper segregation of chromosomes, and requires plus-end-directed homotetrameric motor proteins of the widely conserved kinesin-5 (BimC) family. Hypotheses for bipolar spindle formation include the 'push-pull mitotic muscle' model, in which kinesin-5 and opposing motor proteins act between overlapping microtubules. However, the precise roles of kinesin-5 during this process are unknown. Here we show that the vertebrate kinesin-5 Eg5 drives the sliding of microtubules depending on their relative orientation. We found in controlled in vitro assays that Eg5 has the remarkable capability of simultaneously moving at approximately 20 nm s(-1) towards the plus-ends of each of the two microtubules it crosslinks. For anti-parallel microtubules, this results in relative sliding at approximately 40 nm s(-1), comparable to spindle pole separation rates in vivo. Furthermore, we found that Eg5 can tether microtubule plus-ends, suggesting an additional microtubule-binding mode for Eg5. Our results demonstrate how members of the kinesin-5 family are likely to function in mitosis, pushing apart interpolar microtubules as well as recruiting microtubules into bundles that are subsequently polarized by relative sliding.

The mitotic spindle uses microtubule-based motor proteins to assemble itself and to segregate sister chromatids. It is becoming clear that motors invoke several distinct mechanisms to generate the forces that drive mitosis. Moreover, in carrying out its function, the spindle appears to pass through a series of transient steady-state structures, each established by a delicate balance of forces generated by multiple complementary and antagonistic motors. Transitions from one steady state to the next can occur when a change in the activity of a subset of mitotic motors tips the balance.

Microtubule motors in mitosis

In this article, we review the dynamic nature of the filaments (microtubules) that make up the labile fibers of the mitotic spindle and asters, we discuss the roles that assembly and disassembly of microtubules play in mitosis, and we consider how such assembling and disassembling polymer filaments can generate forces that are utilized by the living cell in mitosis and related movements.

Force Generation by Microtubule Assembly/Disassembly in Mitosis and Related Movements

The first cleavage furrow in eggs of Arbacia (sea urchin) is accompanied by a uniform ring of aligned microfilaments, called the contractile ring. Individual contractile ring filaments measure 35–60 A and occasionally appear "hollow." The contractile ring exists from about 20 sec after anaphase to the end of furrowing activity, i.e., 6–7 min at 20°C. It is closely associated with the plasma membrane at all times, and is probably assembled there. It is about 8 µ wide and 0.2 µ thick throughout cleavage. Its volume decreases, however, suggesting a contraction-related disassembly of contractile ring filaments, rather than a sliding-filament mechanism in the strict sense. Cytochalasin B (>10-6 M) arrests cleavage within 60 sec, by which time contractile ring filaments are no longer visible ultrastructurally. The furrow may be seen to recede within this time. Karyokinesis is unaffected. Simultaneous disruption of furrowing activity and of the contractile ring largely confirms the vital role of the contractile ring as the organelle of cell cleavage.

Autonomous oscillations found in gene expression and metabolic, cardiac and neuronal systems have attracted significant attention both because of their obvious biological roles and their intriguing dynamics. In addition, de novo designed oscillators have been demonstrated, using components that are not part of the natural oscillators. Such oscillators are useful in testing the design principles and in exploring potential applications not limited by natural cellular behaviour. To achieve transcriptional and metabolic integration characteristic of natural oscillators, here we designed and constructed a synthetic circuit in Escherichia coli K12, using glycolytic flux to generate oscillation through the signalling metabolite acetyl phosphate. If two metabolite pools are interconverted by two enzymes that are placed under the transcriptional control of acetyl phosphate, the system oscillates when the glycolytic rate exceeds a critical value. We used bifurcation analysis to identify the boundaries of oscillation, and verified these experimentally. This work demonstrates the possibility of using metabolic flux as a control factor in system-wide oscillation, as well as the predictability of a de novo gene-metabolic circuit designed using nonlinear dynamic analysis.

Chromosome motion during mitosis can be explained by combining the facts available about the equilibrium between the mitotic spindle and its subunits with the postulate that the recently described cross-bridges between spindle microtubules are capable of sliding adjacent tubules over one another.
