Direct microscopic quantification of respiring (i.e., viable) bacteria was performed for drinking water samples and biofilms grown on different opaque substrata. Water samples or biofilms developed in flowing drinking water were incubated with the vital redox dye 5-cyano-2,3-ditolyl tetrazolium chloride (CTC) and R2A medium. One hour of incubation with 0.5 mM CTC was sufficient to obtain intracellular reduction of CTC to the insoluble fluorescent formazan (CTF) product, which was indicative of cellular respiratory (i.e., electron transport) activity. This result was obtained with both planktonic and biofilm-associated cells. Planktonic bacteria were captured on 0.2-microns-pore-size polycarbonate membrane filters and examined by epifluorescence microscopy. Respiring cells containing CTF deposits were readily detected and quantified as red-fluorescing objects on a dark background. The number of CTC-reducing bacteria was consistently greater than the number of aerobic CFU determined on R2A medium. Approximately 1 to 10% of the total planktonic population (determined by counterstaining with 4,6-diamidino-2-phenylindole) were respirometrically active. The proportion of respiring bacteria in biofilms composed of drinking water microflora was greater, ranging from about 5 to 35%, depending on the substratum. Respiring cells were distributed more or less evenly in biofilms, as demonstrated by counterstaining with 4,6-diamidino-2-phenylindole. The amount of CTF deposited in single cells of Pseudomonas putida that formed monospecies biofilms was quantified by digital image analysis and used to indicate cumulative respiratory activity. These data indicated significant cell-to-cell variation in respiratory activity and reduced electron transport following a brief period of nutrient starvation.(ABSTRACT TRUNCATED AT 250 WORDS)

The death and lysis of microorganisms in environmental processes

The results of studies on intramolecular interconversions in systems of norbornadiene quadricyclane and their derivatives are classified and discussed. The mechanisms of the forward photoreaction and reverse thermal process in relation to the nature of the substituents and carbocycles, type of sensitiser and catalyst, and properties of the medium are discussed in detail and classified. An analysis is made of the influence of these factors on the spectroscopic, kinetic, and thermodynamic properties of this system as an accumulator and converter of light energy. Methods of improving its characteristics — light absorption, the quantum yield of the photoreaction, and stability of the photoproduct — have been established. Norbornadiene-quadricyclane — an effective molecular system for the storage of solar energy

The aim of this study was to establish the use of the fluorescent probes carboxyfluorescein (cF) and propidium iodide (PI) for rapid assessment of viability, using Lactococcus lactis subsp. lactis ML3 exposed to different stress treatments. The cF labeling indicated the reproductive capacity of mixtures of nontreated cells and cells killed at 70 degrees C very well. However, after treatment up to 60 degrees C the fraction of cF-labeled cells remained high, whereas the survival decreased for cells treated at above 50 degrees C and was completely lost for those treated at 60 degrees C. In an extended series of experiments, cell suspensions were exposed to heating, freezing, low pH, or bile salts, after which the colony counts, acidification capacity, glycolytic activity, PI exclusion, cF labeling, and cF efflux were measured and compared. The acidification capacity corresponded with the number of CFU. The glycolytic activity, which is an indicator of vitality, was more sensitive to the stress conditions than the reproduction, acidification, and fluorescence parameters. The cF labeling depended on membrane integrity, as was confirmed by PI exclusion. The fraction of cF-labeled cells was not a general indicator of reproduction or acidification, nor was PI exclusion or cF labeling capacity (the internal cF concentration). When the cells were labeled by cF, a subsequent lactose-energized efflux assay was needed for decisive viability assessment. This novel assay proved to be a good and rapid indicator of the reproduction and acidification capacities of stressed L. lactis and has potential for physiological research and dairy applications related to lactic acid bacteria.

Rapid Fluorescence Assessment of the Viability of Stressed Lactococcus lactis

The present invention concerns a spectrometric technique to determine microorganism detection and identification by taking advantage of the inherent extracellular enzymes present in living organisms, as opposed to dead, non-enzyme producing organisms. These enzymes are harnessed in the in vivo reactions with a non-fluorescent dye containing a select organic functional group that is known to be cleaved or hydrolyzed by the certain enzyme. The dye is tailored such that one of the products fluoresces, so that by employing a conventional spectrofluorimeter, the rate of fluorescence can be determined. By subjecting a plurality of samples having different cellular concentrations of viable microorganisms to the same non-fluorescent dye, or by subjecting the same bacterial sample to a number of different non-fluorescent dyes, a pattern of fluorescent rates emerge. By employing the pattern recognition set to standard microorganism fluorescent response curves, microorganism detection and identification can be determined. Thus, the present invention concerns a process for determining microorganism detection, identification and concentration.
