Glutathione peroxidase activity in selenium-deficient rat liver☆

The antioxidant activity and free-radical and active oxygen-scavenging activity of burdock extracts were investigated. Of the solvents used for extraction, water yielded the greatest amount of extract that exhibited the strongest antioxidant activity. Water extracts of burdock (WEB) and hot water extracts of burdock (HWEB) exhibited comparable and marked activity on inhibition of linoleic acid peroxidation, indicating that heat treatment did not alter the antioxidant activity of WEB. WEB and HWEB produced significantly lower (P<0.05) malondialdehyde (MDA) in both linoleic acid and liposome model systems than did the control. Moreover, mixtures of tocopherol (Toc), WEB, and HWEB exhibited a remarkable synergistic antioxidant effect in a liposome system; WEB and HWEB thus potentiated the action of Toc. Furthermore, WEB and HWEB displayed a marked inhibitory effect on lipid peroxidation of rat liver homogenate in vitro. WEB and HWEB exhibited an 80% scavenging effect on α,α-diphenyl-β-picrylhydrazyl radical and marked reducing power, indicating that WEB and HWEB act as primary antioxidants. Both extracts at a dose of 1.0 mg exhibited a 60.4–65.0% scavenging effect on superoxide and an 80.5% scavenging effect on hydrogen peroxide. They also showed a marked scavenging effect on the hydroxyl radical. These results revealed that WEB and HWEB are also active as oxygen scavengers and as secondary antioxidants. Based on these results, termination of free-radical reactions and quenching of reactive oxygen species in burdock extracts are suggested to be, in part, responsible for the antioxidant activity of burdock extracts.

Experiments were conducted with male rats to quantitate the relation ship between dietary selenium (Se) intake and the amount of the enzyme glutathione peroxidase (GSH-Px) in erythrocytes and liver. Weanling male rats were fed torula yeast-based diets with 0, 0.05, 0.1, 0.5, 1.0, or 5.0 ppm Se supplemented as sodium selenite. Liver GSH-Px fell to undetectable levels (<1% of that found in the weanling rats) within 24 days in the O ppm Se group; feeding 0.1 ppm Se, or greater, caused liver GSH-Px to increase above that found in the weanling rats. The erythrocyte GSH-Px response to lack of dietary Se was somewhat smaller in magnitude and more gradual; however, only 21% of initial erythrocyte GSH-Px activity remained in the unsupplemented group after 66 days. Increased dietary Se resulted in corresponding increases of erythrocyte GSH-Px activity. Resupplementing with 0.1, 0.5, or 5.0 ppm Se elevated the depressed erythrocyte GSH-Px levels of the deficient rats. Increased dietary Se provided for both faster elevation, and higher maximal GSH-Px activity which in all cases was achieved 60 to 90 days after resupplementation. The results suggest that tissue GSH-Px can be used as an indicator of animal Se status, but other factors such as age, sex, and dietary vitamin E may have to be considered. Lack of GSH-Px in livers of Se-deficient rats may explain the liver necrosis observed when the diet is also deficient in vitamin E and sulfur-containing amino acids. J. Nutr. 104: 580-587,

Effect of dietary selenium on erythrocyte and liver glutathione peroxidase in the rat

Ethane evolution was monitored from vitamin E and selenium (Se)-deficient rats to determine if lipid peroxidation occurs in vivo when these rats develop fatal organ lesions. Weanling rats were fed a vitamin E and Se-deficient, or supplemented, diet for 40 to 90 days. Each was then prefasted for 4 hours and fasting was continued for 24 to 40 hours while ethane was collected. Approximately 50% of the doubly-deficient rats died as a result of fasting. Pathological signs included hematuria, lung hemorrhage, and liver necrosis. Ethane evolution increased exponentially 10 to 20 hours before death and then declined 2 hours before death. Rats that survived (at least 5 days after ethane collection) evolved 7.4+/-1.3 nmoles ethane/100 g body weight/24 hours compared to 100+/-6 for rats that died. Supplementation of the basal diet with vitamin E (200 IU/kg), Se (0.2 ppm, as Na2SeO3), or both, completely prevented mortality and reduced ethane evolution values to 0.4+/-0.2, 3.1+/-0.4, or 0.2+/-0.2, respectively. These experiments indicate that lipid peroxidation occurs in vivo as a result of vitamin E and Se deficiency, and the peroxidation process greatly accelerates during the terminal phase of the fatal disease.

Lipid peroxidation in vivo during vitamin E and selenium deficiency in the rat as monitored by ethane evolution.

Dietary vitamin E, selenium (Se), and methionine were tested for their ability to inhibit carbon tetrachloride (CCL4)-induced lipid peroxidation. Peroxidation, in vivo, was monitored by the evolution of ethane, an autoxidation product of omega-3-unsaturated fatty acids. Weanling rats were fed a basal diet low in vitamin E, Se, and sulfur-containing amino acids, or diets individually supplemented with these factors. After 3 to 7 weeks, the rats were injected with CCL4 (ip) and ethane was collected for 9 hours. Cumulative ethane evolution was increased by CCl4 in all groups. Vitamin E, Se, and methionine reduced ethane evolution from CCl4-treated rats by 82%, 74%, and 60%, respectively. The toxicity of CCl4 was decreased in correlation with ethane evolution. Thus, methionine and Se, probably by maintaining intracellular glutathione and glutathione peroxidase, protected against CCl4-induced lipid peroxidation, as did vitamin E. Substitution of cod liver oil, which is rich in omega-3-unsaturated fat, for lard in the basal diet increased CCl4-induced ethane evolution six-fold. Relative inhibition by the dietary supplements was not changed. Thus, the feeding of cod liver oil greatly increased ethane production which facilitated the detection and measurement of lipid peroxidation in vivo.
