I have developed "tennis elbow" from lugging this book around the past four weeks, but it is worth the pain, the effort, and the aspirin. It is also worth the (relatively speaking) bargain price. Including appendixes, this book contains 894 pages of text. The entire panorama of the neural sciences is surveyed and examined, and it is comprehensive in its scope, from genomes to social behaviors. The editors explicitly state that the book is designed as "an introductory text for students of biology, behavior, and medicine," but it is hard to imagine any audience, interested in any fragment of neuroscience at any level of sophistication, that would not enjoy this book. Galvanic vestibular stimulation (GVS) is a simple, safe, and specific way to elicit vestibular reflexes. Yet, despite a long history, it has only recently found popularity as a research tool and is rarely used clinically. The obstacle to advancing and exploiting GVS is that we cannot interpret the evoked responses with certainty because we do not understand how the stimulus acts as an input to the system. This paper examines the electrophysiology and anatomy of the vestibular organs and the effects of GVS on human balance control and develops a model that explains the observed balance responses. These responses are large and highly organized over all body segments and adapt to postural and balance requirements. To achieve this, neurons in the vestibular nuclei receive convergent signals from all vestibular receptors and somatosensory and cortical inputs. GVS sway responses are affected by other sources of information about balance but can appear as the sum of otolithic and semicircular canal responses. Electrophysiological studies showing similar activation of primary afferents from the otolith organs and canals and their convergence in the vestibular nuclei support this. On the basis of the morphology of the cristae and the alignment of the semicircular canals in the skull, rotational vectors calculated for every mode of GVS agree with the observed sway. However, vector summation of signals from all utricular afferents does not explain the observed sway. Thus we propose the hypothesis that the otolithic component of the balance response originates from only the pars medialis of the utricular macula.

Direct modulation of the secretory machinery underlies PKA-dependent synaptic facilitation in hippocampal neurons.

We investigated the probability of survival of mouse spinal neurons in monolayer cultures after transection lesions of dendrites made within 400 μm of the perikarya. Based on a total of 650 lesioned neurons, the following observations were made. First, neuronal survival is a function of lesion distance from the perikaryon and of process diameter at the lesion site. For an average lesion diameter of 3 μm, dendrite transections at 50 μm, 100 μm, and 150 μm were associated with survival probabilities of 30%, 53%, and 70%, respectively. Second, the fate of the injured cells was definitely established 24 hours after injury and very likely was determined as early as 2 hours. Third, early stages of deterioration leading to cell death were associated with cytoplasmic phase brightness on light microscopy, correlating with an appearance of numerous, small, electron-lucent vacuoles and swollen mitochondria on electron microscopy. Neuronal survival or death after dendrite transection close to the perikaryon: correlation with electrophysiologic, morphologic, and ultrastructural changes.

The functional plasticity of the nervous system may result in part from the direct modulation of the effectiveness of the release machinery of synaptic terminals. To date, direct modulation of secretion in neurons has proven difficult to study because of the lack of a suitable tool to probe the release machinery independently of calcium influx. We report that the polyvalent cation ruthenium red (RR) directly evokes rapid and reversible calcium-independent quantal secretion in hippocampal neurons by binding to external sites on the presynaptic terminal membrane. This binding can be displaced by heparin and is not associated with ultrastructural damage to the synaptic terminals. The use of RR-evoked release as a tool has allowed us to detect a direct modulation of the secretory apparatus after activation of A1 adenosine receptors on hippocampal neurons.

Calcium-independent activation of the secretory apparatus by ruthenium red in hippocampal neurons: a new tool to assess modulation of presynaptic function

Cultured mouse spinal neurons were fixed at three different intervals after dendrite amputation: within the first 15 min, at 2 h and at 24 h. Dendrites were amputated at lesion distance of either 50 μm (31% probability of cell survival) or 100 μm (53% probability of cell survival) from the edge of their perikarya. When fixed within 15 min, operated neurons showed a two-phase gradient of ultrastructural damage which spread from the transection site towards the perikaryon. At 2 h after dendrite amputation all neurons operated close to their perikarya were categorized as either viable, moribund or dead, based on their appearance with phase contrast microscopy. These categories of response to physical trauma corresponded to distinctly different ultrastructural changes. Moribund neurons were filled with membrane-bound vesicles which were derived from swollen mitochondria and grossly dilated cisternae of the smooth endoplasmic reticulum. The cytoplasm of dead neurons contained large clear areas and many condensed, dark mitochondria. Both moribund and dead neurons lacked cytoskeletal elements. All of these ultrastructural changes are hypothesized to be the result of an increase in the intracellular concentrations of free calcium. Although evidence of residual mitochondrial swelling was present in some surviving neurons at 24 h, the ultrastructure of others was comparable to that of control cells. Some surviving neurons had terminal swellings at the ends of the severed neurites which were very similar to retraction balls of transected axons after CNS trauma.
