It is of great importance to develop multifunctional bioactive scaffolds, which combine angiogenesis capacity, osteostimulation, and antibacterial properties for regenerating lost bone tissues. In order to achieve this aim, we prepared copper (Cu)-containing mesoporous bioactive glass (Cu-MBG) scaffolds with interconnective large pores (several hundred micrometer) and well-ordered mesopore channels (around 5 nm). Both Cu-MBG scaffolds and their ionic extracts could stimulate hypoxia-inducible factor (HIF)-1a and vascular endothelial growth factor(VEGF) expression in human bone marrow stromal cells(hBMSCs). In addition, both Cu-MBG scaffolds and their ionic extracts significantly promoted the osteogenic differentiation of hBMSCs by improving their bone-related gene expression (alkaline phosphatase (ALP), osteopontin(OPN) and osteocalcin (OCN)). Furthermore, Cu-MBG scaffolds could maintain a sustained release of ibuprofen and significantly inhibited the viability of bacteria. This study indicates that the incorporation of Cu2þ ions into MBG scaffolds significantly enhances hypoxia-like tissue reaction leading to the coupling of angiogenesis and osteogenesis. Cu2þ ions play an important role to offer the multifunctional properties of MBG scaffold system. This study has demonstrated that it is possible to develop multifunctional scaffolds by combining enhanced angiogenesis potential, osteostimulation, and antibacterial properties for the treatment of large bone defects.

Copper-containing mesoporous bioactive glass scaffolds with multifunctional properties of angiogenesis capacity, osteostimulation and antibacterial activity

Cancer is one of the top life-threatening dangers to the human survival, accounting for over 10 million deaths per year. Bioactive glasses have developed dramatically since their discovery 50 years ago, with applications that include therapeutics as well as diagnostics. A new system within the bioactive glass family, mesoporous bioactive glasses (MBGs), has evolved into a multifunctional platform, thanks to MBGs easy-to-functionalize nature and tailorable textural properties-surface area, pore size, and pore volume. Although MBGs have yet to meet their potential in tumor treatment and imaging in practice, recently research has shed light on the distinguished MBGs capabilities as promising theranostic systems for cancer imaging and therapy. This review presents research progress in the field of MBG applications in cancer diagnosis and therapy, including synthesis of MBGs, mechanistic overview of MBGs application in tumor diagnosis and drug monitoring, applications of MBGs in cancer therapy ( particularly, targeted delivery and stimuli-responsive nanoplatforms), and immunological profile of MBG-based nanodevices in reference to the development of novel cancer therapeutics.

Mesoporous Bioactive Glasses in Cancer Diagnosis and Therapy: Stimuli-Responsive, Toxicity, Immunogenicity, and Clinical Translation

Synthesis and characterization of novel mesoporous strontium-modified bioactive glass nanospheres for bone tissue engineering applications

Sol-gel chemistry offers a flexible, widely accepted methodology that enables the creation of a new generation of bioactive glass (BG). In the current study, a sol-gel method was used to synthesize ZrO2 incorporated 56SiO2–34CaO–10P2O5 mol% bioactive glass. The highly crystalline structure was composed of small zirconium oxide nanoparticles (ZrO2) of less than 200 nm in size. It was successfully fabricated using a hydrothermal method. Polyurethane foam (PU) was selected to fabricate a highly porous BG-ZrO2 scaffold using a foam replica technique. The physicochemical, morphological properties of the BG-ZrO2 compositions were evaluated using X-ray diffraction (XRD), Fourier transforms infrared (FTIR), thermo-gravimetric analysis (TGA), transmission electron microscope (TEM) and scanning electron microscope (SEM) with energy dispersive spectroscopy (EDS). In-vitro degradation analysis of the BG-ZrO2 scaffolds was performed after immersion of the samples in simulated body fluid (SBF). The incorporation of ZrO2 nanoparticles into the bioactive glass matrix enhances both the mechanical strength and thermal stability. Since the novel formed BG-ZrO2 scaffolds possesses respectable antibacterial properties against some bacterial strains, this renders it an ideal tissue engineering substitute, capable of reducing failure rates in implants.
