Leaf-macronutrient content and yield in grafted melon plants. A model to evaluate the influence of rootstock genotype

The role of soil fauna in crucial ecosystem services such as nutrient cycling remains poorly quantified, mainly because of the overly reductionistic approach adopted in most experimental studies. Given that increasing nitrogen inputs in various ecosystems influence the structure and functioning of soil microbes and the activity of fauna, we aimed to quantify the role of the entire soil nematode community in nutrient mineralization in an experimental set-up emulating nutrient-rich field conditions and accounting for crucial interactions amongst the soil microbial communities and plants. To this end, we reconstructed a complex soil foodweb in mesocosms that comprised largely undisturbed native microflora and the entire nematode community added into defaunated soil, planted with Lolium perenne as a model plant, and amended with fresh grass-clover residues. We determined N and P availability and plant uptake, plant biomass and abundance and structure of the microbial and nematode communities during a three-month incubation. The presence of nematodes significantly increased plant biomass production (+9%), net N (+25%) and net P (+23%) availability compared to their absence, demonstrating that nematodes link below- and above-ground processes, primarily through increasing nutrient availability. The experimental set-up presented allows to realistically quantify the crucial ecosystem services provided by the soil biota.

Nematodes enhance plant growth and nutrient uptake under C and N-rich conditions.

Chloride (Cl(-)) is a micronutrient that accumulates to macronutrient levels since it is normally available in nature and actively taken up by higher plants. Besides a role as an unspecific cell osmoticum, no clear biological roles have been explicitly associated with Cl(-) when accumulated to macronutrient concentrations. To address this question, the glycophyte tobacco (Nicotiana tabacum L. var. Habana) has been treated with a basal nutrient solution supplemented with one of three salt combinations containing the same cationic balance: Cl(-)-based (CL), nitrate-based (N), and sulphate+phosphate-based (SP) treatments. Under non-saline conditions (up to 5 mM Cl(-)) and no water limitation, Cl(-) specifically stimulated higher leaf cell size and led to a moderate increase of plant fresh and dry biomass mainly due to higher shoot expansion. When applied in the 1-5 mM range, Cl(-) played specific roles in regulating leaf osmotic potential and turgor, allowing plants to improve leaf water balance parameters. In addition, Cl(-) also altered water relations at the whole-plant level through reduction of plant transpiration. This was a consequence of a lower stomatal conductance, which resulted in lower water loss and greater photosynthetic and integrated water-use efficiency. In contrast to Cl(-), these effects were not observed for essential anionic macronutrients such as nitrate, sulphate, and phosphate. We propose that the abundant uptake and accumulation of Cl(-) responds to adaptive functions improving water homeostasis in higher plants.

Extraction and analysis of nitrogen, phosphorus and carbon fractions in plant material

The effects of N and P deficiency, isolated or in combination, on leaf gas exchange and fast chlorophyll (Chl) fluorescence emission were studied in common bean cv. Negrito. 10-d-old plants grown in aerated nutrient solution were supplied with high N (HN, 7.5 mol m−3) or low N (LN, 0.5 mol m−3), and also with high P (HP, 0.5 mol m−3) or low P (LP, 0.005 mol m−3). Regardless of the external P supply, in LN plants the initial fluorescence (F0) increased 12 % in parallel to a quenching of about 14 % in maximum fluorescence (Fm). As a consequence, the variable to maximum fluorescence ratio (Fv/Fm) decreased by about 7 %, and the variable to initial fluorescence ratio (Fv/F0) was lowered by 25 % in relation to control plants. In LP plants, Fv/Fm remained unchanged whilst Fv/F0 decreased slightly as a result of 5 % decline in Fm. Under N deficiency, the net photosynthetic rate (PN) halved at 6 d after imposition of treatment and so remained afterwards. As compared to LN plants, PN declined in LP plants latter and to a less extent. From 12 d of P deprivation onwards. PN fell down progressively to display rates similar to those of LN plants only at the end of the experiment. The greater PN in LP plants was not reflected in larger biomass accumulation in relation to LN beans. In general, P and N limitation affected photosynthesis parameters and growth without showing any synergistic or additive effect between deficiency of both nutrients.
