We compared growth and the content of sugar, protein, and photosynthetic pigments, as well as chlorophyll fluorescence parameters in 15- and 27-day-old Chinese cabbage (Brassica chinensis L.) plants grown under a high-pressure sodium (HPS) lamps or a light source built on the basis of red (650 nm) and blue (470 nm) light-emitting diodes (LEDs) with a red to blue photon ratio of 7: 1. One group of plants was grown at a photosynthetic photon flux (PPF) level of 391 ± 24 μ mol/(m2 s) (normal level); the other, at a PPF level of 107 ± 9 μ mol/(m2 s) (low light). Plants of the third group were firstly grown at the low light and then (on the 12th day) transferred to the normal level. When grown at the normal PPF level, the plants grown under LEDs didn’t differ from plants grown under HPS lamps in shoot fresh weight, but they showed a lower root fresh and dry weights and the lower content of total sugar and sugar reserves in the leaves. No differences in the pigment content and photosystem II quantum yield were found; however, a higher Chl a/b ratio in plants grown under LEDs indicates a different proportion of functional complexes in thylakoid membranes. The response to low light conditions was mostly the same in plants grown under HPS lamps and LEDs; however, LED plants showed a lower growth rate and a higher nonphotochemical fluorescence quenching. In the case of the altered PPF level during growth, the plant photosynthetic apparatus adapted to new conditions of illumination within three days. Plants grown under HPS lamps at a constant normal PPF level and those transferred to the normal PPF level on the 12th day, on the 27th day didn’t differ in shoot fresh weight, but in plants grown under LEDs, the differences were considerable. Our results show that LED-based light sources can be used for plant growing. At the same time, some specific properties of plant photosynthesis and growth under these conditions of illumination were found.

Growth and photosynthesis of Chinese cabbage plants grown under light-emitting diode-based light source

Marigold (Tagetes erecta) flowers are primarily used in industry for their high pigment content. Flower color development implies that chloroplast–chromoplast transition is associated with carotenoid biosynthesis. We report the recovery of undifferentiated pigmented marigold cells, various callus tissues, and their analysis by transmission electron microscopy in order to observe accumulating pigment and development of subcellular structures. Callus was generated from leaf explants and after several rounds of recurrent selection. Green-, yellow-, and brown-colored callus were obtained that showed distinct carotenoid profiles. For green material, violaxanthin, lutein, zeaxanthin, and β-carotene were produced, while yellow callus generated mainly lutein, as did the brown callus. Chloroplast–chromoplast transition was followed by measuring plastid size and shape in undifferentiated marigold cells by digital image analysis. Cellular alterations were evident in brown callus. Chloroplasts were the main structure in green callus, while yellow callus clearly showed the formation of plastoglobules, structures that are correlated with chloroplast–chromoplast transition. The high number of plastoglobules observed in yellow callus is possibly directly related to pigment synthesis and accumulation.

Plastid analysis of pigmented undifferentiated cells of marigold Tagetes erecta L. by transmission electron microscopy

The heat tolerance of wheat (Triticum aestivum L.) and radish (Raphanus sativus L. var. minor) cenoses exposed to elevated and damaging air temperatures (35°C for 20 h, 45°C for 7 h) under photoculture conditions at various levels of photosynthetically active radiation (PAR) was assessed by measuring characteristics of the slow induction curve of chlorophyll fluorescence at 682 and 734 nm and the CO2 exchange rate. Irrespective of the illumination level, the exposure of the cenoses to 35°C did not induce irreversible changes in the plant photosynthetic apparatus. The lowest extent of damage to wheat and radish cenoses exposed to 45°C was observed at 150 W/m2 of PAR, whereas the highest damage of the plants was observed at an illumination level that was close to the compensation point of the cenose photosynthesis (50–70 W/m2 of PAR at air temperature of 24°C). Viability index proved to be the most sensitive characteristic, compared to other characteristics, which were determined by measuring the slow phase of fluorescence induction at 682 and 734 nm. In the cenoses studied, the pattern of changes in the viability index in response to a stress factor was close to the changes in the photosynthetic rate.

Characteristics of Slow Induction Curve of Chlorophyll Fluorescence and CO2 Exchange for the Assessment of Plant Heat Tolerance at Various Levels of Light Intensity

Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600–700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.
