Sustainability is a key principle in natural resource management, and it involves operational efficiency, minimisation of environmental impact and socio-economic considerations; all of which are interdependent. It has become increasingly obvious that continued reliance on fossil fuel energy resources is unsustainable, owing to both depleting world reserves and the green house gas emissions associated with their use. Therefore, there are vigorous research initiatives aimed at developing alternative renewable and potentially carbon neutral solid, liquid and gaseous biofuels as alternative energy resources. However, alternate energy resources akin to first generation biofuels derived from terrestrial crops such as sugarcane, sugar beet, maize and rapeseed place an enormous strain on world food markets, contribute to water shortages and precipitate the destruction of the world's forests. Second generation biofuels derived from lignocellulosic agriculture and forest residues and from non-food crop feedstocks address some of the above problems; however there is concern over competing land use or required land use changes. Therefore, based on current knowledge and technology projections, third generation biofuels specifically derived from microalgae are considered to be a technically viable alternative energy resource that is devoid of the major drawbacks associated with first and second generation biofuels. Microalgae are photosynthetic microorganisms with simple growing requirements (light, sugars, CO 2 , N, P, and K) that can produce lipids, proteins and carbohydrates in large amounts over short periods of time. These products can be processed into both biofuels and valuable co-products. This study reviewed the technologies underpinning microalgae-to-biofuels systems, focusing on the biomass production, harvesting, conversion technologies, and the extraction of useful co-products. It also reviewed the synergistic coupling of microalgae propagation with carbon sequestration and wastewater treatment potential for mitigation of environmental impacts associated with energy conversion and utilisation. It was found that, whereas there are outstanding issues related to photosynthetic efficiencies and biomass output, microalgae-derived biofuels could progressively substitute a significant proportion of the fossil fuels required to meet the growing energy demand.

Aquatic phototrophs: efficient alternatives to land-based crops for biofuels

C-phycocyanin (C-PC) is a blue pigment in cyanobacteria, rhodophytes and cryptophytes with fluorescent and antioxidative properties. C-PC is presently extracted from open pond cultures of the cyanobacterium Arthrospira platensis although these cultures are not very productive and open for contaminating organisms. C-PC is considered a healthy ingredient in cyanobacterial-based foods and health foods while its colouring, fluorescent or antioxidant properties are utilised only to a minor extent. However, recent research and developments in C-PC synthesis and functionality have expanded the potential applications of C-PC in biotechnology, diagnostics, foods and medicine: The productivity of C-PC has been increased in heterotrophic, high cell density cultures of the rhodophyte Galdieria sulphuraria that are grown under well-controlled and axenic conditions. C-PC purification protocols based on various chromatographic principles or novel two-phase aqueous extraction methods have expanded in numbers and improved in performance. The functionality of C-PC as a fluorescent dye has been improved by chemical stabilisation of C-PC complexes, while protein engineering has also introduced increased stability and novel biospecific binding sites into C-PC fusion proteins. Finally, our understanding of the physiological functions of C-PC in humans has been improved by a mechanistic hypothesis that links the chemical properties of the phycocyanobilin chromophores of C-PC to the natural antioxidant, bilirubin, and may explain the observed health benefits of C-PC intake. This review outlines how C-PC is produced and utilised and discusses the novel C-PC synthesis procedures and applications.

Production of phycocyanin : a pigment with applications in biology, biotechnology, foods and medicine

Cultivation of microalgae using natural and man-made open-ponds istechnologically simple, but not necessary cheap due to the high downstream processing cost. Products of microalgae cultured in open-pondscould only be marketed as value-added health food supplements, specialityfeed and reagents for research. The need to achieve higher productivityand to maintain monoculture of algae led to the development of enclosedtubular and flat plate photobioreactors. Despite higher biomassconcentration and better control of culture parameters, data accumulatedin the past 25 years have shown that the illuminated areal, volumetricproductivity and cost of production in these enclosed photobioreactors arenot better than those achievable in open-pond cultures. The technicaldifficulty in sterilizing these photobioreactors has hindered their applicationfor the production of high value pharmaceutical products. The alternativeof cultivating microalgae in heterotrophic mode in sterilizable fermentorshas achieved some commercial success. The maximum specific growth ratesof heterotrophic algal cultures are in general slower than those measured inphotosynthetic cultures. The biomass productivity of heterotrophic algalcultures has yet to achieve a level that is comparable to industrialproduction of yeast and other heterotrophic microrganisms. Mixotrophiccultivation of microalage takes advantage of their ability to utilise organicenergy and carbon substrates and perform photosynthesis concurrently. Moreover, production of some algal metabolites is light regulated. Futuredesign of sterilizable bioreactors for mixotrophic cultivation of microalgaemay have to consider the organic substrate the main source of energy andlight the supplemental source of energy, a change in mindset.

Microalgal mass culture systems and methods: Their limitation and potential

In her News Feature “Biotech’s green gold”, Emily Waltz details the ‘hype’ being propagated around emerging microalgal biofuel technologies, which often exceeds the physical and thermodynamic constraints that ultimately define their economic viability. Our calculations counter such excessive claims and demonstrate that 22 MJ m−2 d−1 solar radiation supports practical yield maxima of ∼60 to 100 kl oil ha−1 y−1 (∼6,600 to 10,800 gal ac−1 y−1) and an absolute theoretical ceiling of ∼94 to 155 kl oil ha−1 y−1, assuming a maximum photosynthetic conversion efficiency of 10%. To evaluate claims and provide an accurate analysis of the potential of microalgal biofuel systems, we have conducted industrial feasibility studies and sensitivity analyses based on peer-reviewed data and industrial expertise. Given that microalgal biofuel research is still young and its development still in flux, we anticipate that the stringent assessment of the technology's economic potential presented below will assist R&D investment and policy development in the area going forward.

An economic and technical evaluation of microalgal biofuels

This work represents an attempt to assess the relative contribution of the factors limiting productivity ofSpirulina platensis in open raceways throughout the year. Temperature of the culture during daylight exerted the predominant effect on productivity and elevating the temperature resulted in a significant rise in productivity even in summer. Photoinhibition had a decisive role in summer in determining productivity ofSpirulina in open raceways in that growth almost ceased after mid-day. Contamination by other microorganisms, particularlyS. minor andChlorella sp. was estimated to reduce the net biomass yield by at least 15 to 20%, but measures to curtail the establishment of these species in the raceway have been devised. The effect of harvest time during the day on the yield of dry mass was examined: no conclusion could be drawn except in mid-summer, when evening harvesting resulted in a significant increase in the output rate of dry weight over morning harvesting.
