TL;DR: In this article, both enantiomers of rolipram (1 ) have been prepared in large quantity from a common intermediate rac- 3-(3'-cyclopentyloxy-4'methoxy)phenyl-4-nitro butyric acid (6), which was resolved byway of the two readily separable diastereoisomeric amides obtained with (S )-(−)-phenylethylamine.

TL;DR: Bromonitrile-Noxide adds to the enone completely regio-and stereoselectively to form the bromoisoxazoline 10b/c. The latter can be transformed into the nitrile 11, previously hydrolyzed 14 to the target 16α-carbomethoxyprednisolone 4a a potentially locally active antiinflammatory steroid.

N-arylrolipram derivatives as potent and selective PDE4 inhibitors

TL;DR: Derivatization of rolipram led to the identification of 3-[4-(3-cyclopentyloxy-4-methoxyphenyl)-2-oxo-pyrrolidin-1-yl]-5-( 3- methoxybenzyloxy)-benzoic acid N',N'-dimethylhydrazide (4), a potent and selective inhibitor of PDE4.
