1. The in vivo alkaline comet (single cell gel electrophoresis) assay (hereafter called simply the comet assay) is used for the detection of DNA strand breaks in cells or nuclei isolated from multiple tissues of animals, usually rodents, that have been exposed to potentially genotoxic material(s). The comet assay has been reviewed and recommendations have been published by various expert groups (1) (2) (3) (4) (5) (6) (7) (8) (9) (10). This Test Guideline is part of a series of Test Guidelines on genetic toxicology. A document presented as an Introduction to the Test Guidelines on genotoxicity (11) can also be referred to and provides succinct and useful guidance to users of these Test Guidelines.

Antibody blockade of TNF-α reduces inflammation and scarring in experimental crescentic glomerulonephritis

Graded doses of Nature Cure Bitters (NCB) were administered daily (100, 200 and 400 mg/kg p.o) to rats for 28 days and the effects on body weight, organ weight, clinical signs, gross pathology, haematology, histology and serum biochemical parameters were evaluated. The relative weights of the heart, liver and testes of treated rats were unaffected in contrast to a significant increase in the relative weights of the lungs, kidneys and spleen. The packed cell volume and haemoglobin concentrations were significantly reduced whereas total leucocyte counts and glucose levels were remarkably increased. A significant decrease in alkaline phosphatase occurred in all the groups but alanine aminotransferase and albumin levels were significantly elevated. NCB elicited hypo-cholesterolaemic effects in addition to lowering urea, uric acid, BUN and total protein concentrations. Histological findings did not reveal any treatment-related effects. The calculated therapeutic index was >37.5. These preliminary results suggest that NCB was not likely to produce severe toxicological effects on organ weights, haematological and biochemical indices when given at normal therapeutic doses.

This chapter discusses the care and maintenance that should be provided to laboratory animals. The animal house is an important adjunct of the laboratory. The essential features of any animal house are light and airy rooms with adequate heating and drainage, separate food stores, and a sterilizing room. The design of the animal house depends on the facilities available, but the minimum essentials are a quarantine room, a normal stock room, and an infected animal room. Mice, rats, guinea pigs, and rabbits are the more common animals used for experimental purposes. While these are hardy animals in their natural surroundings, when housed together, they are susceptible to epidemic infections. Cleanliness and regular feeding are two very important factors in maintaining a trouble-free stock.

The care and maintenance of laboratory animals

This chapter provides an overview of cerebrospinal fluid. Cerebrospinal fluid envelops the brain and spinal cord, acting as a buffer against mechanical shock. Specimens of spinal fluid are obtained by introducing a needle between adjacent vertebrae, usually in the lumbar region, with the patient's back flexed to separate the bones. The chapter discusses the techniques for estimation of total protein. There are two main methods: (1) colorimetric (Biuret) and (2) turbidometric. The resulting turbidity is proportional to the amount of protein present. The opacity of the solution is compared with standard turbidity tubes of the same calibre. The chapter also describes the technique for estimation of glucose, chlorides, globulin, sugar, and total protein. The sugar present in cerebrospinal fluid is glucose. Levels of urea in blood and cerebrospinal fluid are the same that is 15–40 mg per 100 ml. The range of normal values is wide but is generally considered to be 50–100 mg per 100 ml. The estimation may, therefore, be carried out on whichever fluid is more readily available.
