During ontogeny, plants interact with a wide variety of microorganisms. The association with mutualistic microbes results in benefits for the plant. By contrast, pathogens may cause a remarkable impairment of plant growth and development. Both types of plant-microbe interactions provoke notable changes in the polyamine (PA) metabolism of the host and/or the microbe, being each interaction a complex and dynamic process. It has been well documented that the levels of free and conjugated PAs undergo profound changes in plant tissues during the interaction with microorganisms. In general, this is correlated with a precise and coordinated regulation of PA biosynthetic and catabolic enzymes. Interestingly, some evidence suggests that the relative importance of these metabolic pathways may depend on the nature of the microorganism, a concept that stems from the fact that these amines mediate the activation of plant defense mechanisms. This effect is mediated mostly through PA oxidation, even though part of the response is activated by non-oxidized PAs. In the last years, a great deal of effort has been devoted to profile plant gene expression following microorganism recognition. In addition, the phenotypes of transgenic and mutant plants in PA metabolism genes have been assessed. In this review, we integrated the current knowledge on this field and analyze the possible roles of these amines during the interaction of plants with microbes.

Physiological and molecular implications of plant polyamine metabolism during biotic interactions

The fungal pathogen Fusarium graminearum causes Fusarium Head Blight (FHB) disease on wheat which can lead to trichothecene mycotoxin (e.g. deoxynivalenol, DON) contamination of grain, harmful to mammalian health. DON is produced at low levels under standard culture conditions when compared to plant infection but specific polyamines (e.g. putrescine and agmatine) and amino acids (e.g. arginine and ornithine) are potent inducers of DON by F. graminearum in axenic culture. Currently, host factors that promote mycotoxin synthesis during FHB are unknown, but plant derived polyamines could contribute to DON induction in infected heads. However, the temporal and spatial accumulation of polyamines and amino acids in relation to that of DON has not been studied. Following inoculation of susceptible wheat heads by F. graminearum, DON accumulation was detected at two days after inoculation. The accumulation of putrescine was detected as early as one day following inoculation while arginine and cadaverine were also produced at three and four days post-inoculation. Transcripts of ornithine decarboxylase (ODC) and arginine decarboxylase (ADC), two key biosynthetic enzymes for putrescine biosynthesis, were also strongly induced in heads at two days after inoculation. These results indicated that elicitation of the polyamine biosynthetic pathway is an early response to FHB. Transcripts for genes encoding enzymes acting upstream in the polyamine biosynthetic pathway as well as those of ODC and ADC, and putrescine levels were also induced in the rachis, a flower organ supporting DON production and an important route for pathogen colonisation during FHB. A survey of 24 wheat genotypes with varying responses to FHB showed putrescine induction is a general response to inoculation and no correlation was observed between the accumulation of putrescine and infection or DON accumulation. The activation of the polyamine biosynthetic pathway and putrescine in infected heads prior to detectable DON accumulation is consistent with a model where the pathogen exploits the generic host stress response of polyamine synthesis as a cue for production of trichothecene mycotoxins during FHB disease. However, it is likely that this mechanism is complicated by other factors contributing to resistance and susceptibility in diverse wheat genetic backgrounds.

Methyl jasmonate alters arginine catabolism and improves postharvest chilling tolerance in cherry tomato fruit

Exogenous treatment with jasmonates (JA) has been shown to reduce the levels of polyamines in many plants. But the role of endogenous JA on polyamine biosynthesis or other cellular metabolites has thus far remained uninvestigated. We developed transgenic tomato (Solanum lycopersicum L.) having severely reduced methyl JA levels by silencing a fruit ripening-associated lipoxygenase (LOX), SlLoxB, using a truncated LOX gene under the control of the constitutive CaMV35S promoter. The LOX suppressed and MeJA-deficient fruits had lowered polyamine levels. Thus, these transgenic fruits were used as a plant model to evaluate the effects of reduced endogenous MeJA on cellular metabolites in ripening tomato fruits using NMR spectroscopy. During on-shelf ripening, transgenic fruits were significantly reduced in the content of 19 out of 30 metabolites examined, including Ile, Val, Ala, Thr, Asn Tyr, Glu, Gln, His, Phe, Trp, GABA, citrate, succinate, myo-inositol, unidentified compound B, nucleic acid compound Nucl1, choline, and trigonelline as compared to the wild-type azygous counterparts. A significant increase in β-glucose levels in transgenic fruits was observed at the pink stage. The transgenic fruits were equivalent to the wild type in lycopene level and chlorophyll degradation rates. Taken together, these results show that intracellular MeJA significantly regulates overall primary metabolism, especially aminome (amino acids and polyamines) of ripening fruits.

Leaf rust of wheat (Triticum aestivum L.), incited by Puccinia recondita ex Desm. f. sp. tritici Eriks, is one of the most important wheat diseases in Egypt. Methyl jasmonate (MJ) is a potential plant elicitor which induces a wide range of chemical and anatomical defense reactions in conifers and might be used to increase systemic resistance against biotic damage. In the greenhouse, different concentrations of MJ (10, 20 and 30 mM) were applied as seed soaking plus foliar spray or only as foliar spray to control leaf rust and induction of secondary compound production in leaves of wheat plants. Foliar spray was applied after 30 and 50 days of sowing. Results indicated that all concentrations and treatments reduced the severity of rust disease caused by P. recondita f. sp. tritici in wheat leaves during 45 days of inoculations. Aflatoxins are extremely potent natural carcinogens and a major food safety concern because of potential contamination of food commodities. Plant resistance to aflatoxin contamination is not commercially available and fungicides are not effective. The use of antagonists microorganisms against mycotoxin fungi is a promising approach to manage aflatoxin contamination via biological control. The objective of this study was to evaluate some effective biological control include Rhodotorula glutinis, Penicillium oxalicum and Trichoderma harzianum for inhibition aflatoxin B1 produce by Aspergillus flavus isolated from Walnuts and Pistachio grown in Sinai governorate. The filtrates of all isolates produced varying degrees of inhibition against A. flavus. Nevertheless, the highest percentage of inhibition of toxigenic A. flavus by the culture filtrate of R. glutinis at 150 mg/kg . The ability of biological control agents strains to prevent aflatoxin production have been confirmed by HPLC analysis. Certain yeast strain was found to confine the spreading and sporulation and reduce the amount of aflatoxin production. Sporulation was limited due to the inhibitory effect of yeast. There was a noticeable decrease in fungal dry mass and aflatoxin production. Treated walnuts and pistachio and seeds with biocontrol isolates were effective in controlling fungal infection and increased seeds germination .
