Antifouling technology—past, present and future steps towards efficient and environmentally friendly antifouling coatings

This introductory review on plasma health care is intended to provide the interested reader with a summary of the current status of this emerging field, its scope, and its broad interdisciplinary approach, ranging from plasma physics, chemistry and technology, to microbiology, biochemistry, biophysics, medicine and hygiene. Apart from the basic plasma processes and the restrictions and requirements set by international health standards, the review focuses on plasma interaction with prokaryotic cells (bacteria), eukaryotic cells (mammalian cells), cell membranes, DNA etc. In so doing, some of the unfamiliar terminology—an unavoidable by-product of interdisciplinary research—is covered and explained. Plasma health care may provide a fast and efficient new path for effective hospital (and other public buildings) hygiene— helping to prevent and contain diseases that are continuously gaining ground as resistance of pathogens to antibiotics grows. The delivery of medically active 'substances' at the molecular or ionic level is another exciting topic of research through effects on cell walls (permeabilization), cell excitation (paracrine action) and the introduction of reactive species into cell cytoplasm. Electric fields, charging of surfaces, current flows etc can also affect tissue in a controlled way. The field is young and hopes are high. It is fitting to cover the beginnings in New Journal of Physics, since it is the physics (and non- equilibrium chemistry) of room temperature atmospheric pressure plasmas that have made this development of plasma health care possible.

Plasma medicine: an introductory review

The application of strong electric fields in water and organic liquids has been studied for several years, because of its importance in electrical transmission processes and its practical applications in biology, chemistry, and electrochemistry. More recently, liquid-phase electrical discharge reactors have been investigated, and are being developed, for many environmental applications, including drinking water and wastewater treatment, as well as, potentially, for environmentally benign chemical processes. This paper reviews the current status of research on the application of high-voltage electrical discharges for promoting chemical reactions in the aqueous phase, with particular emphasis on applications to water cleaning.

Electrohydraulic Discharge and Nonthermal Plasma for Water Treatment

Pulse power technology using high intensity (up to 300 kV/cm) nanosecond pulsed electric fields (nsPEF) has been applied for decontamination and amelioration of biofouling, but until now effects on human cells have not been investigated. To analyze structural and functional changes in human cells and solid tumors following exposure to nsPEF, we utilized flow cytometry and immunofluorescence microscopy. We provide further support for the hypothesis that as the pulse duration is decreased, there is a lower incidence of electric field interactions at the plasma membrane and a higher incidence of interactions with intracellular structures. The nsPEF effects are pulse duration/electric field intensity-dependent and energy density- or temperature-independent. We also show that nsPEF induces programmed cell. death (apoptosis) in cultured cells as indicated by cell shrinkage, Annexin-V-FITC binding to phosphatidylserine on intact cells, and caspase activation. Mouse fibrosarcoma tumors exposed to nsPEF exhibit fragmented DNA and reduced tumor growth in a mouse model. These studies show that nsPEF effects are distinctly different than electroporation pulses and provide the first evidence for the potential application of nsPEF to induce apoptosis and inhibit tumor growth.

Nanosecond pulsed electric field (nsPEF) effects on cells and tissues: apoptosis induction and tumor growth inhibition

The medical, food processing, and heating, ventilating, and air conditioning industries are searching for improved pasteurization, disinfection, and sterilization technologies. Candidate techniques must deal with and overcome such problems as thermal sensitivity and destruction by heat, formation of toxic by-products, costs, and inefficiency in performance. We report the results of a plasma source, the One Atmosphere Uniform Glow Discharge Plasma (OAUGDP), which operates at atmospheric pressure in air and produces antimicrobial active species at room temperature, OAUGDP exposures have reduced log numbers of Gram negative and Gram positive bacteria, bacterial endospores, yeast, and bacterial viruses on a variety of surfaces. The nature of the surface influenced the degree of lethality, with microorganisms on polypropylene being most sensitive, followed by glass, and cells embedded in agar. Experimental results showed at least a 5 log/sub 10/ CFU reduction in bacteria within a range of 50-90 s of exposure. After 10-25 s of exposure, macromolecular leakage and bacterial fragmentation were observed. Vulnerability of cell membranes to reactive oxygen species (ROC) is hypothesized. Results from several novel OAUGDP configurations are presented, including a remote exposure reactor (RER) which uses transported active species to sterilize material located more than 20 cm from the plasma generation site, and a second planar electrode configuration developed for air filter sterilization. Applications of these technologies to the healthcare industry, the food industry, and decontaminating surfaces compromised by biological warfare agents are discussed.

An overview of research using the one atmosphere uniform glow discharge plasma (OAUGDP) for sterilization of surfaces and materials

The effect of pulsed electric fields with amplitudes in the range of 100 V/cm-100 kV/cm on bacteria and aquatic nuisance species has been explored. The pulse duration was so short that heating of the biological matter could be neglected. The electrical energy required for lysing of bacteria, or stunning of aquatic species, decreases when the pulse duration is reduced. For lysing of Eschericia coli, this tendency has been proven to hold for pulsewidths as short as 60 ns. For macroorganisms, however, it was found that for pulsewidths of less than 5 /spl mu/s, the tendency is reversed: the energy required to affect the macroorganisms increases again. This minimum in energy, or maximum in efficiency, respectively, can be understood by taking the time required for electrical charging of the cell membrane into account. Applications of the pulsed electric field technique (PEFT) are in biofouling prevention, debacterialization of liquids, and in the field of medicine. A series of field tests on biofouling prevention in a cooling system with untreated water as coolant has demonstrated the economic feasibility of the electro-technology.

The effect of pulsed electric fields on biological cells: experiments and applications

The processes of persistent photoconductivity followed by photoquenching have been demonstrated at megawatt power levels in copper-compensated, silicon-doped, semi-insulating gallium arsenide. These processes allow a photoconductive switch to be developed that can be closed by the application of one laser pulse (/spl lambda/=1.06 /spl mu/m) and opened by the application of a second laser pulse with a wavelength equal to twice that of the first laser (/spl lambda/=2.13 /spl mu/m). This switch is called the bistable optically controlled semiconductor switch (BOSS). The opening phase of the BOSS requires a sufficient concentration of recombination centers (RC) in the material for opening to occur in the subnanosecond regime. These RC's are generated in the bulk GaAs material by fast-neutron irradiation (/spl sim/1 MeV). Neutron-irradiated BOSS devices have been opened against a rising average electric field of about 36 kV/cm (18 kV) in a time less than 1 ns while operating at a repetition rate, within a two-pulse burst, of about 1 GHz. The ability to modify the frequency content of the electrical pulses, by varying the time separation, is demonstrated. Results demonstrating the operation of two BOSS devices imbedded in a frequency-agile RF source configuration are also discussed.
