These systems may involve separating sample components by partitioning them into droplets or other partitions, amplifying or otherwise reacting the components within the droplets, detecting the amplified components, or characteristics thereof, and/or analyzing the resulting data, among others.

The invention provides a method, apparatus and system for separating blood and other types of cellular components, and can be combined with holographic optical trapping manipulation or other forms of optical tweezing. One of the exemplary methods includes providing a first flow having a plurality of blood components; providing a second flow; contacting the first flow with the second flow to provide a first separation region; and differentially sedimenting a first blood cellular component of the plurality of blood components into the second flow while concurrently maintaining a second blood cellular component of the plurality of blood components in the first flow. The second flow having the first blood cellular component is then differentially removed from the first flow having the second blood cellular component. Holographic optical traps may also be utilized in conjunction with the various flows to move selected components from one flow to another, as part of or in addition to a separation stage.

Provided herein are methods, compositions, and kits for assays, many of which involve amplification reactions such as digital PCR or droplet digital PCR. The assays may be used for such applications as sequencing, copy number variation analysis, and others. An apparatus for acoustic concentration of particles in a fluid flow includes a substantially acoustically transparent membrane and a vibration generator that define a fluid flow path therebetween. A multichannel system for classifying particles according to one or more characteristics of the particles includes a plurality of flow cytometry units. Each flow cytometry unit is operable to classify particles in a mixture of particles by interrogating a stream of fluid containing particles using a beam of electromagnetic radiation. Each flow cytometry unit has a sensor operable to generate a time-varying output signal indicative of at least one characteristic of the particles in the stream of fluid as the stream of fluid is interrogated by the beam of electromagnetic radiation. The flow cytometry units share an integrated platform and perhaps even a common processor for receiving and processing information from the units. The common processor is programmed to receive the time-varying output signals from the flow cytometry units substantially continuously and to process the output signals.

System for analyzing particles using multiple flow cytometry units

A multi-channel apparatus for classifying particles according to one or more particle characteristics. The apparatus comprises a plurality of flow cytometry units, each of which is operable to classify particles in a mixture of particles by interrogating a stream of fluid containing the particles with a beam of electromagnetic radiation. The flow cytometry units share an integrated platform comprising at least one of the following: (1) a common supply of particles; (2) a common housing; (3) a common processor for controlling operation of the units; (4) a common processor for receiving and processing information from the units; and (5) a common fluid delivery system. The integrated platform can include a common source of electromagnetic radiation. A method of the invention comprises using a plurality of flow cytometry units sharing the integrated platform to perform a flow kilometric operation, such as analyzing or sorting particles.

Systems for Efficient Staining and Sorting of Populations of Cells

A system and method for sorting a mixture of stained particles including a digital signal processor for analyzing and classifying the digital information generated from the particles and providing a sorting signal to a sorting system as a function of the analyzed and classified digital information.

Digital sampling apparatus and methods for sorting particles

A method of sorting stained particles in a flow cytometer by generating a mathematical model based upon extracted features of waveform pulses produced in response to the stained particles. Waveform pulses can then be analyzed to determine the probability the stained particles belong in either a first or a second population and a decision boundary can be established. Sorting decisions can be made based upon the extracted features of the waveform pulses relative to the decision boundary in combination with other desirability factors for particles or droplets containing the particles.
