Visible-light-mediated photocatalytic generation of carbon-centred radicals from alkyl, alkenyl and aryl iodides, which then undergo subsequent hydrogen-atom abstraction or reductive cyclizations, is reported. The protocol is characterized by the use of inexpensive reagents, mild conditions, exceptional functional group tolerance, and good to high yields.

Engaging unactivated alkyl, alkenyl and aryl iodides in visible-light-mediated free radical reactions

In spite of the considerable progress made in recent years toward the understanding of the chemistry and biological function of the cobalt-containing B(12) group of compounds, much of the information still is more descriptive than definitive in nature. In general terms, it is known that the free vitamin forms can function as methyl group carriers and that the 5'-deoxyadenosyl or coenzyme forms serve as hydrogen carriers; but the mechanism of these processes is not understood in detail. More systematic studies of the pure chemistry of these complex molecules containing a carbon-cobalt covalent bond are needed before the biochemist can interpret many of his observations on the enzyme-catalyzed reactions. Even in relatively simple solutions it is difficult to ascertain the state of oxidation of several of the vitamin forms, and these problems are compounded when the reactive thiol compounds and complex proteins of the biological systems also are present. For example, both vitamin B(12r) (the Co(2+) form) and corresponding analogs are known to disproportionate in solution to B(12s) (Co(1+)) and B(12a) (Co(3+)) under a variety of mild conditions (12, 57). This means that in the biological systems it is exceedingly difficult to ascertain the chemical nature of many B(12) intermediates and reaction products. The role of the protein moiety of the various B(12)-linked enzymes in the catalytic processes is little known as is, also, the mode of binding of the B(12) derivative to the protein. These types of questions perhaps can be answered eventually by the crystallographers, whose art is becoming increasingly sophisticated. Note added after preparation of manuscript. In contrast to the values given in Table 4 for the molecular weights of the two dissimilar protein moieties of glycerol dehydrase, a recent report (57a), gives a value of 188,000 for the molecular weight of a stable, catalytically inactive complex of 1 mole of hydroxocobalamin and 1 mole of the apoenzyme complex of glycerol dehydrase. The latter is presumed to contain one equivalent of each of the two dissimilar protein subunits. The original estimate of 240,000 as the molecular weight of the unstable sulfhydryl protein moiety (39) was undoubtedly made on partially aggregated material.

Vitamin B 12.

Cultures of Clostridium formicoaceticum and C. thermoaceticum growing on fructose and glucose, respectively, were shown to rapidly oxidize CO to CO(2). Rates up to 0.4 mumol min(-1) mg of wet cells(-1) were observed. Carbon monoxide oxidation by cell suspensions was found (i) to be dependent on pyruvate, (ii) to be inhibited by alkyl halides and arsenate, and (iii) to stimulate CO(2) reduction to acetate. Cell extracts catalyzed the oxidation of carbon monoxide with methyl viologen at specific rates up to 10 mumol min(-1) mg of protein(-1) (35 degrees C, pH 7.2). Nicotinamide adenine dinucleotide, nicotinamide adenine dinucleotide phosphate and ferredoxin from C. pasteurianum were ineffective as electron acceptors. The catalytic mechanism of carbon monoxide oxidation was "ping-pong," indicating that the enzyme catalyzing carbon monoxide oxidation can be present in an oxidized and a reduced form. The oxidized form was shown to react reversibly with cyanide, and the reduced form was shown to react reversibly with alkyl halides: cyanide inactivated the enzyme only in the absence of carbon monoxide, and alkyl halides inactivated it only in the presence of carbon monoxide. Extracts inactivated by alkyl halides were reactivated by photolysis. The findings are interpreted to indicate that carbon monoxide oxidation in the two bacteria is catalyzed by a corrinoid enzyme and that in vivo the reaction is coupled with the reduction of CO(2) to acetate. Cultures of C. acidi-urici and C. cylindrosporum growing on hypoxanthine were found not to oxidize CO, indicating that clostridia mediating a corrinoid-independent total synthesis of acetate from CO(2) do not possess a CO-oxidizing system.

Photostability studies of drugs and drug products are an integral part of the product development process in the pharmaceutical industry. These studies are carried out to ensure quality, efficacy, and safety of the formulated products during manufacture, storage, and use. This review deals with the concept of photostability and related aspects and the literature available in the field. It highlights the role of the photochemistry in the photostability studies, describes the functional groups important for the photoreactivity of drugs, explains photophysical processes, and deals with the kinetics of photochemical reactions. The various modes of photodegradation of drugs with examples of selected compounds are presented. The biological consequences of the effect of light on the drug degradation are described. The photostability testing of drugs and drug products and the requirements under ICH guideline are discussed. Some information on the packaging requirements for the formulated products is provided. The various methods used for the photostabilization of solid and liquid dosage forms are also discussed.
