Soil salinity restricts plant growth and productivity. Na + rep- resents the major ion causing toxicity because it competes with K + for binding sites at the plasma membrane. Inocula- tion with arbuscular mycorrhizal fungi (AMF) can alleviate salt stress in the host plant through several mechanisms. These may include ion selection during the fungal uptake of nutrients from the soil or during transfer to the host plant. AM benefits could be enhanced when native AMF isolates are used.Thus, we investigated whether native AMF isolated from an area with problems of salinity and desertification can help maize plants to overcome the negative effects of salinity stress better than non-AM plants or plants inoculated with non-native AMF. Results showed that plants inoculated with two out the three native AMF had the highest shoot dry biomass at all salinity levels. Plants inoculated with the three native AMF showed significant increase of K + and reduced Na + accumulation as compared to non-mycorrhizal plants, concomitantly with higher K + /Na + ratios in their tissues. For the first time, these effects have been correlated with regula- tion of ZmAKT2, ZmSOS1 and ZmSKOR genes expression in the roots of maize, contributing to K + and Na + homeostasis

Arbuscular mycorrhizal fungi native from a Mediterranean saline area enhance maize tolerance to salinity through improved ion homeostasis

It is well known that salinization (high-pH) has been considered as a major environmental threat to agricultural systems. The aim of this study was to investigate the differences between salt stress and alkali stress in metabolic profiles and nutrient accumulation of wheat; these parameters were also evaluated to determine the physiological adaptive mechanisms by which wheat tolerates alkali stress. The harmful effect of alkali stress on the growth and photosynthesis of wheat were stronger than those of salt stress. High-pH of alkali stress induced the most of phosphate and metal ions to precipitate; as a result, the availability of nutrients significantly declined. Under alkali stress, Ca sharply increased in roots, however, it decreased under salt stress. In addition, we detected the 75 metabolites that were different among the treatments according to GC-MS analysis, including organic acids, amino acids, sugars/polyols and others. The metabolic data showed salt stress and alkali stress caused different metabolic shifts; alkali stress has a stronger injurious effect on the distribution and accumulation of metabolites than salt stress. These outcomes correspond to specific detrimental effects of a highly pH environment. Ca had a significant positive correlation with alkali tolerates, and increasing Ca concentration can immediately trigger SOS Na exclusion system and reduce the Na injury. Salt stress caused metabolic shifts toward gluconeogenesis with increased sugars to avoid osmotic stress; energy in roots and active synthesis in leaves were needed by wheat to develop salt tolerance. Alkali stress (at high pH) significantly inhibited photosynthetic rate; thus, sugar production was reduced, N metabolism was limited, amino acid production was reduced, and glycolysis was inhibited.

Comparative metabolic responses and adaptive strategies of wheat (Triticum aestivum) to salt and alkali stress.

An in-depth understanding of the effects of drought stress on plant metabolism is necessary to improve the drought tolerance of wheat and to utilize genetic resources for the development of drought stress-tolerant wheat varieties. In this study, the profiles of 58 key metabolites produced by wheat seedlings in response to drought stress were investigated to determine various physiological processes related to drought tolerance between drought-tolerant and drought-sensitive wheat genotypes. Results showed that the wheat metabolome was dominated by sugars, organic acids and amino acids; the wheat metabolome played important roles to enhance the drought tolerance of shoots. Under drought stress, JD17 exhibited higher growth indices and higher photosynthesis ability than JD8. A high level of compatible solutes and energy in shoots were essential for wheat to develop drought tolerance. Drought also caused system alterations in widespread metabolic networks involving transamination, tricarboxylic acid cycle, glycolysis, glutamate-mediated proline biosynthesis, shikimate-mediated secondary metabolisms and γ-aminobutyric acid metabolisms. Long-term drought stress resulted in the drought-tolerant wheat genotype JD17, which induced metabolic shifts in the tricarboxylic acid cycle and glycolysis with the depletion of the γ-aminobutyric acid shut process. In JD17, the prolonged drought stress induced a progressive accumulation of osmolytes, including proline, sucrose, fructose, mannose and malic acid. This research extended our understanding of the mechanisms involved in wheat seedling drought tolerance; this study also demonstrated that gas chromatography-mass spectrometry metabolomics could be an effective approach to understand the drought effects on plant biochemistry.

Metabolic responses to drought stress in the tissues of drought-tolerant and drought-sensitive wheat genotype seedlings

Production of Jatropha curcas as a biodiesel feedstock on marginal lands is growing rapidly. Biomass production on these lands is limited. Hence, the objective of this study was to evaluate the effect of arbuscular mycorrhiza (AM) fungi and salinity (0.1, 0.2, 0.3, 0.4, and 0.5% NaCl) on (1) seedling growth, leaf relative water content (RWC), lipid peroxidation, solute accumulation (proline and sugars), and photosynthetic pigments (Chl a and b) of Jatropha; (2) mycorrhizal colonization (%) and mycorrhizal dependency (MD) of Jatropha; and (3) glomalin content (Bradford reactive soil protein) in soil. Increased soil salinity significantly (P < 0.05) decreased AM root colonization (r
 2 = 0.98) of AM-inoculated plants and decreased survival (r
 2 = 0.93) and growth (shoot length, r

Under salt stress, AM-inoculated Jatropha plants had greater dry weight of shoots and roots, better leaf water status, less leaf membrane damage (low lipid peroxidation activity), higher solute (proline and sugars), and higher leaf chlorophyll concentrations than non-AM-inoculated plants. The mycorrhizal dependency (MD) of Jatropha increased from 12.13 to 20.84% with salinity (0–0.4% NaCl). Root AM colonization (%) and glomalin content in soil were negatively correlated with salinity (P < 0.05, r = −0.95). We conclude that inoculation with AM fungi lessens the deleterious effect of salt stress on seedling growth parameters under salt levels up to 0.5% NaCl (electrical conductivity of 7.2 dS m−1). Inoculation of Jatropha seedlings with AM fungi can promote the establishment of Jatropha under NaCl-induced stress.

Influence of Arbuscular Mycorrhizal (AM) Fungi and Salinity on Seedling Growth, Solute Accumulation, and Mycorrhizal Dependency of Jatropha curcas L.

GsSRK, a G-type lectin S-receptor-like serine/threonine protein kinase, is a positive regulator of plant tolerance to salt stress.

We compared the effects of salt-stresses (SS, 1: 1 molar ratio of NaCl to Na2SO4) and alkali-stresses (AS, 1: 1 molar ratio of NaHCO3 to Na2CO3) on the growth, photosynthesis, solute accumulation, and ion balance of barley seedlings, to elucidate the mechanism of AS (high-pH) damage to plants and the physiological adaptive mechanism of plants to AS. The effects of SS on the water content, root system activity, membrane permeability, and the content of photosynthetic pigments were much less than those of AS. However, AS damaged root function, photosynthetic pigments, and the membrane system, led to the severe reductions in water content, root system activity, content of photosynthetic pigments, and net photosynthetic rate, and a sharp increase in electrolyte leakage rate. Moreover, with salinity higher than 60 mM, Na+ content increased slowly under SS and sharply under AS. This indicates that high-pH caused by AS might interfere with control of Na+ uptake in roots and increase intracellular Na+ to a toxic level, which may be the main cause of some damage emerging under higher AS. Under SS, barley accumulated organic acids, Cl−, SO42−, and NO3− to balance the massive influx of cations, the contribution of inorganic ions to ion balance was greater than that of organic acids. However, AS might inhibit absorptions of NO3− and Cl−, enhance organic acid synthesis, and SO42− absorption to maintain intracellular ion balance and stable pH.

Comparative effects of salt-stress and alkali-stress on the growth, photosynthesis, solute accumulation, and ion balance of barley plants

This study compared the effects of salt (NaCl) stress on growth, photosynthesis and solute accumulation in seedlings of the three poplar (Populus bonatii) cultivars Populus × BaiLin-2 (BL-2), Populus × BaiLin-3 (BL-3), and Populus × Xjiajiali (XJJL). The results showed that BL-2 and BL-3 could not survive at a salinity level of 200 mM but XJJL grew well. The effect of moderate salt stress on leaf extension of the three cultivars was only slight. At a high level of salinity, however, NaCl clearly inhibited leaf extension of BL-2 and BL-3, whereas it did not affect that of XJJL, and the net photosynthetic rate (P
 N) in XJJL was much higher than those of BL-2 and BL-3. The lower P
 N of BL-2 and BL-3 might be associated with the high concentration of Na+ and/or Cl− accumulated in the leaves, which could be toxic in photosynthesis system. In summary, the greater salt-tolerance of XJJL compared with that of BL-2 and BL-3 might be explained by the higher P
 N and photosynthetic area, the lower Na+/K− ratio and Cl− in the leaf, and the greater accumulation of soluble sugars and SO4
 2−.
