Soil amendment with biochar is evaluated globally as a means to improve soil fertility and to mitigate climate change. However, the effects of biochar on soil biota have received much less attention than its effects on soil chemical properties. A review of the literature reveals a significant number of early studies on biochar-type materials as soil amendments either for managing pathogens, as inoculant carriers or for manipulative experiments to sorb signaling compounds or toxins. However, no studies exist in the soil biologyliterature that recognize the observed largevariations ofbiochar physico-chemical properties. This shortcoming has hampered insight into mechanisms by which biochar influences soil microorganisms, fauna and plant roots. Additional factors limiting meaningful interpretation of many datasets are the clearly demonstrated sorption properties that interfere with standard extraction procedures for soil microbial biomass or enzyme assays, and the confounding effects of varying amounts of minerals. In most studies, microbial biomass has been found to increase as a result of biochar additions, with significant changes in microbial community composition and enzyme activities that may explain biogeochemical effects of biochar on element cycles, plant pathogens, and crop growth. Yet, very little is known about the mechanisms through which biochar affects microbial abundance and community composition. The effects of biochar on soil fauna are even less understood than its effects on microorganisms, apart from several notable studies on earthworms. It is clear, however, that sorption phenomena, pH and physical properties of biochars such as pore structure, surface area and mineral matter play important roles in determining how different biochars affect soil biota. Observations on microbial dynamics lead to the conclusion of a possible improved resource use due to co-location of various resources in and around biochars. Sorption and therebyinactivation of growth-inhibiting substances likelyplaysa rolefor increased abundance of soil biota. No evidence exists so far for direct negative effects of biochars on plant roots. Occasionally observed decreases in abundance of mycorrhizal fungi are likely caused by concomitant increases in nutrient availability,reducing theneedfor symbionts.Inthe shortterm,therelease ofavarietyoforganic molecules from fresh biochar may in some cases be responsible for increases or decreases in abundance and activity of soil biota. A road map for future biochar research must include a systematic appreciation of different biochar-types and basic manipulative experiments that unambiguously identify the interactions between biochar and soil biota.

Inoculants of plant growth-promoting bacteria for use in agriculture

Inoculation of legume seed is an efficient and convenient way of introducing effective rhizobia to soil and subsequently the rhizosphere of legumes. However, its full potential is yet to be realised. Following widespread crop failures, the manufacture of high quality inoculants revolutionised legume technology in Australia in the 1960s. Many improvements to inoculants and the advent of an inoculant control service ensured that quality was optimised and maintained. Minimum standards for the number of rhizobia per seed were set after consideration of several factors including seed size and loss of viability during inoculation. Despite manufacturers' recommendations for storage and application of inoculants, there is a distinct lack of control over the inoculation process; hence the full potential of high quality products may not always be achieved. The efficacy of inoculation varies depending on several factors, all of which affect the number of viable rhizobia available for infection of legume roots. Increased numbers of viable rhizobia per seed by application of inoculant above the commercially recommended rate, results in a continued linear increase in nodulation and yield. Several studies have reported yield increases of up to 25%. However, applying higher quantities of inoculant is uneconomical and technically difficult. Alternatively, higher numbers of viable rhizobia per seed may be achieved by improving survival during seed inoculation. Despite recognition of the factors affecting survival of rhizobia on seed and a substantial demand for commercially pre-inoculated legume seed, poor survival is still a major concern. Desiccation, temperature and seed coat toxicity all influence survival of rhizobia on seed. Their adverse effects may be ameliorated by selecting tolerant rhizobial strains and legume seed cultivars with low toxicity or artificially, by the use of additives in the seed coating. The accumulation of the desiccant protectant trehalose in strains of rhizobia, may result in better survival under desiccation stress. Similarly, the accumulation of exopolysaccharide (EPS) may act as a barrier reducing excessive water loss. Polymeric adhesives such as gum arabic, methyl cellulose and polyvinyl pyrollidone (PVP) have improved survival. However, studies of additives used in inoculation have been ad hoc and little of their mode of action is understood. A better understanding of the mechanisms involved in the protection of rhizobia from adverse conditions will assist in defining the optimum conditions for seed inoculation and storage to ensure a higher quality product for farmers at the time of sowing.

Legume seed inoculation technology—a review

Inputs of biologically fixed nitrogen derived from the symbiotic relationship between legumes and their root-nodule bacteria into terrestrial ecosystems amount to at least 70 million metric tons per year. It is obvious that this enormous quantity will need to be augmented as the world's population increases and as the natural resources that supply fertilizer nitrogen diminish. This objective will be achieved through the development of superior legume varieties, improvement in agronomic practice, and increased efficiency of the nitrogen fixation process itself by better management of the symbiotic relationship between plant and bacteria. This paper considers ways and means by which populations of root-nodule bacteria, established and introduced, can be manipulated ecologically, agronomically, edaphically and genetically to improve legume productivity and, as a consequence, soil fertility.

Manipulation of rhizobia microflora for improving legume productivity and soil fertility: a critical assessment

Inoculant production and formulation

The effects of inoculant carrier, temperature, and storage period on the survival of Rhizobium strains were determined by plate count and most-probable-number analyses. Preliminary experiments showed that survival of rhizobia was affected by each of these factors and their interactions. Results of further studies indicated that six strains of rhizobia survived better at high temperatures when lyophilized and suspended in an oil carrier as compared to finely ground peat. The oil base inocula contained ca. 105 viable rhizobia per g after 56 days of incubation at 60°C, whereas peat base inocula contained ≤10 rhizobia per g. These results suggest that an oil carrier will protect rhizobia from rapid death at usually lethal high temperatures.

Effects of Carrier and Temperature on Survival of Rhizobium spp. in Legume Inocula: Development of an Improved Type of Inoculant.

Patterns of intrinsic resistance and susceptibility to different levels of antibiotics were determined for strains of both fast- and slow-growing rhizobia. These patterns were stable to plant passage when they were used to identify Rhizobium strains in nodule suspensions or nodule isolates. The method of identification by intrinsic resistance and susceptibility patterns was reliable for identifying strains in field nodules when strains were first isolated from the nodules to provide a standard inoculum size and then typed on antibiotic-containing media. Other patterns of resistance were encountered during identification of field isolates; these patterns may have resulted from acquired resistance to certain antibiotics or from mixed infections of the nodules. The occurrence of resistance patterns identical to those of inoculant strains among native strains was directly related to the size of the soil population. High strain recovery was associated directly with high rates of inoculation.
