Plants are sessile organisms. Consequently they have to adapt constantly to fluctuations in the environment. Some of these changes involve essential factors such as nutrients, light, and water. Plants have evolved independent systems to sense nutrients such as phosphate and nitrogen. However, many of the environmental factors may reach levels which represent stress for the plant. The fluctuations can range between moderate and unfavorable, and the factors can be of biotic or abiotic origin. Among the biotic factors influencing plant life are pathogens and herbivores. In case of bacteria and fungi, symbiotic interactions such as nitrogen-fixating nodules and mycorrhiza, respectively, may be established. In case of insects, a tritrophic interaction of herbivores, carnivores, and plants may occur mutualistically or parasitically. Among the numerous abiotic factors are low temperature, frost, heat, high light conditions, ultraviolet light, darkness, oxidation stress, hypoxia, wind, touch, nutrient imbalance, salt stress, osmotic adjustment, water deficit, and desiccation. In the last decade jasmonates were recognized as being signals in plant responses to most of these biotic and abiotic factors. Signaling via jasmonates was found to occur intracellularly, intercellularly, and systemically as well as interorganismically. Jasmonates are a group of ubiquitously occurring plant growth regulators originally found as the major constituents in the etheric oil of jasmine, and were first suggested to play a role in senescence due to a strong senescence-promoting effect. Subsequently, numerous developmental processes were described in which jasmonates exhibited hormone-like properties. Recent knowledge is reviewed here on jasmonates and their precursors, the octadecanoids. After discussing occurrence and biosynthesis, emphasis is placed upon the signal transduction pathways in plant stress responses in which jasmonates act as a signal. Finally, examples are described on the role of jasmonates in developmental processes.

Jasmonates and octadecanoids: signals in plant stress responses and development.

12-oxo-phytodienoic acid and several phytoprostanes are cyclopentenone oxylipins that are formed via the enzymatic jasmonate pathway and a nonenzymatic, free radical-catalyzed pathway, respectively. Both types of cyclopentenone oxylipins induce the expression of genes related to detoxification, stress responses, and secondary metabolism, a profile clearly distinct from that of the cyclopentanone jasmonic acid. Microarray analyses revealed that 60% of the induction by phytoprostanes and 30% of the induction by 12-oxo-phytodienoic acid was dependent on the TGA transcription factors TGA2, TGA5, and TGA6. Moreover, treatment with phytoprostanes and 12-oxo-phytodienoic acid inhibited cell division and root growth, a property also shared by jasmonic acid. Besides being potent signals, cyclopentenones and other lipid peroxidation products are reactive electrophiles that can covalently bind to and damage proteins. To this end, we show that at least two of the induced detoxification enzymes efficiently metabolize cyclopentenones in vitro. Accumulation of two of these metabolites was detectable during Pseudomonas infection. The cyclopentenone oxylipin gene induction profile resembles the defense response induced by a variety of lipophilic xenobiotics. Hence, oxidized lipids may activate chemosensory mechanisms of a general broad-spectrum detoxification network involving TGA transcription factors.

General Detoxification and Stress Responses Are Mediated by Oxidized Lipids through TGA Transcription Factors in Arabidopsis

The biosynthesis of jasmonic acid: a physiological role for plant lipoxygenase.

The natural occurrence of jasmonic acid and its methyl ester in plants has been studied using different methods such as GC, GC-MS, HPLC, radioimmunoassay, and bioassay. Jasmonic acid was detected in several Leguminosae plants and a number of species belonging to nine other Angiospermae families. Highest amounts occurred in fruit parts, especially the immature pericarp, but it was found also in flowers and vegetative plant parts, e.g. leaves, stems, and germs. Young apple fruits contain both jasmonic acid and methyl jasmonate, and in Douglas fir, the only Gymnospermae species studied, only the methyl ester could be detected. Jasmonic acid is discussed as an endogenous plant growth regulator widely distributed in higher plants.

Occurrence of the plant growth regulator jasmonic acid in plants

Jasmonates are plant signal molecules that are derived from lipids through the action of lipoxygenase. Jasmonates regulate gene expression during plant development and in response to water deficit, wounding, and pathogen elicitors. The signal transduction chain that mediates jasmonate action was investigated by isolating and studying two methyl jasmonate (MeJA)-insensitive mutants of Arabidopsis thaliana. The recessive mutants, jin1 and jin4, are nonallelic and neither corresponds to coi1, a previously identified MeJA-insensitive mutant. Both mutants showed reduced sensitivity to MeJA-mediated root growth inhibition as well as reduced MeJA induction of AtVsp in leaves. Expression of AtVsp in flowers was not altered in the mutants. Furthermore, MeJA modulation of the jasmonate-responsive lipoxygenase and phenylalanine ammonia lyase genes was not altered in the mutants. jin4 plants exhibited increased sensitivity to abscisic acid in seed germination assays, whereas jin1 plants showed wild-type sensitivity. Neither mutant showed altered sensitivity to ethylene in hypocotyl growth inhibition assays. jin1 and jin4 identify genes that modulate the response of AtVsp to MeJA in leaves of A. thaliana.
