‘‘Gentlemen don’t need mistletoe’’ said the Christmas whiskey billboard I saw recently in Florida, a garish reminder of how much mistletoe lore is embedded in western society. Mistletoe The Genus Viscum, one in the series of medicinal and aromatic plants for industry and academic researchers, centers on the mistletoe in this advertisement, the common European mistletoe,Viscum album. There are helpful discussions on African, Asian, and Argentine mistletoes and their uses but the corpus of the book deals with V. album. Viscum album, considered sacred by the Druids, is no doubt the most widely used parasitic angiosperm for various health concoctions. As a result, much has been learned about its biochemistry and pharmaceutical potential. A great deal of the work on the culture and utilization ofV. album has been done by investigators at Institute Hiscia Center for Cancer Research in Switzerland where votaries of Rudolf Steiner’s distinct form of homeopathy have used mistletoe extracts (‘‘Iscador’’) for many years in cancer treatment. Mistletoe collected from less common hosts are considered to have greater efficacy in preparation of Iscador. As a result, there is a helpful chapter on culturing V. album which is not as difficult as might be imagined for an obligate parasite. Most of the chapters deal with the medicinal aspects of mistletoe including toxicology, clinical aspects, chemistry, and biochemistry emphasizing the lectins which are one of the more desirable compounds produced by the parasite. I found the treatment by Bu ̈ssing (Biological and pharmacological properties of Viscum album L.) helpful because it reviews the link between folk and modern medicine. However, several relevant papers were omitted which are included in a recent excellent review of mistletoes (Watson 2001). Like many books with a diversity of authors, the quality of chapters varies. Obviously produced as a reference volume, Mistletoe lacks an extensive index. Especially aggravating is the lack of an author’s index, limiting the value of the book for anyone looking for specific papers. These days, $70 for a hardbound monograph is reasonable. Color images are well-produced but some of the black and white figures are blurry in my copy. This volume will be of value to ethnobotanists, anyone interested in alternative medicines, and students of mistletoes and parasitic plants. LITERATURE CITED

Genetic Data Analysis II. Methods for Discrete Population Genentic Data

The basic notions of statistical mechanics (microstates, multiplicities) are quite simple, but understanding how the second law arises from these ideas requires working with cumbersomely large numbers. To avoid getting bogged down in mathematics, one can compute multiplicities numerically for a simple model system such as an Einstein solid -- a collection of identical quantum harmonic oscillators. A computer spreadsheet program or comparable software can compute the required combinatoric functions for systems containing a few hundred oscillators and units of energy. When two such systems can exchange energy, one immediately sees that some configurations are overwhelmingly more probable than others. Graphs of entropy vs. energy for the two systems can be used to motivate the theoretical definition of temperature, $T= (\partial S/\partial U)^{-1}$, thus bridging the gap between the classical and statistical approaches to entropy. Further spreadsheet exercises can be used to compute the heat capacity of an Einstein solid, study the Boltzmann distribution, and explore the properties of a two-state paramagnetic system.

A different approach to introducing statistical mechanics

Humans have domesticated diverse species from across the plant kingdom, yet much of our foundational knowledge of domestication has come from studies investigating relatively few of the most important annual food crops. Here, we examine the impacts of domestication on genetic diversity in a tropical perennial fruit species, mango (Mangifera indica). We used restriction site associated DNA sequencing to generate genomic single nucleotide polymorphism (SNP) data from 106 mango cultivars from seven geographical regions along with 52 samples of closely related species and unidentified cultivars to identify centers of mango genetic diversity and examine how post-domestication dispersal shaped the geographical distribution of diversity. We identify two gene pools of cultivated mango, representing Indian and Southeast Asian germplasm. We found no significant genetic bottleneck associated with the introduction of mango into new regions of the world. By contrast, we show that mango populations in introduced regions have elevated levels of diversity. Our results suggest that mango has a more complex history of domestication than previously supposed, perhaps including multiple domestication events, hybridization and regional selection. Our work has direct implications for mango breeding and genebank management, and also builds on recent efforts to understand how woody perennial crops respond to domestication.

Population genomic analysis of mango (Mangifera indica) suggests a complex history of domestication

Local mangos (Mangifera indica L.) are highly valued for home consumption in rural Kenya and are regarded by the local population to be comparatively drought tolerant and less susceptible to pests and diseases than the improved varieties. These are characteristics which make them interesting for improvement and breeding purposes. To date, research on Kenyan mangos has mainly focused on introduced and commercial varieties, whereas information on local varieties and landraces is lacking. We present the first comprehensive morphological and molecular characterisation of local mango landraces from Eastern and Central Kenya. Thirty-eight local mango trees were sampled and characterized by 75 selected qualitative (44) and quantitative (31) morphological descriptors selected from the descriptor list developed for mango by Bioversity International (former IPGRI). Hierarchical cluster analysis was performed using first all variables and finally only 10 selected key descriptors. Additionally, dried mango leaves from the same 38 trees were used for molecular classification with 19 simple sequence repeat markers. Genetic relatedness between the mango samples was visualized using a dendrogram based on Nei’s genetic distance and Neighbor Joining methods. Morphological characterisation resulted in six distinct clusters, and molecular analysis in eight clusters, which partly supported the morphological classification. Four of the eight molecular clusters were consistent and molecular results confirmed morphological classification in these cases. Identification of local mango landraces using morphological traits can be considered as satisfying under field conditions, e.g. for rootstock identification in nurseries, but environmental conditions may influence the results. Molecular marker analysis is more expensive, but independent from environmental influences and thus suitable for identification of landraces in field genebanks and for breeding purposes. Outcomes of the present study can form the basis for urgently needed future conservation efforts, including circa situ conservation on farms and the development of ‘conservation through use’ strategies for local mango landraces in Kenya.

Identification of mango (Mangifera indica L.) landraces from Eastern and Central Kenya using a morphological and molecular approach

Mango is one of the most important fruits of tropical ecological region of the world, well known for its nutritive value, aroma and taste. Its world production is >45MT worth >200 billion US dollars. Genomic resources are required for improvement in productivity and management of mango germplasm. There is no web-based genomic resources available for mango. Hence rapid and cost-effective high throughput putative marker discovery is required to develop such resources. RAD-based marker discovery can cater this urgent need till whole genome sequence of mango becomes available. Using a panel of 84 mango varieties, a total of 28.6 Gb data was generated by ddRAD-Seq approach on Illumina HiSeq 2000 platform. A total of 1.25 million SNPs were discovered. Phylogenetic tree using 749 common SNPs across these varieties revealed three major lineages which was compared with geographical locations. A web genomic resources MiSNPDb, available at http://webtom.cabgrid.res.in/mangosnps/ is based on 3-tier architecture, developed using PHP, MySQL and Javascript. This web genomic resources can be of immense use in the development of high density linkage map, QTL discovery, varietal differentiation, traceability, genome finishing and SNP chip development for future GWAS in genomic selection program. We report here world's first web-based genomic resources for genetic improvement and germplasm management of mango.

MiSNPDb: a web-based genomic resources of tropical ecology fruit mango (Mangifera indica L.) for phylogeography and varietal differentiation.

Allelic patterns and genetic distances were examined in a collection of 103 foreign and Brazilian mango (Mangifera indica) accessions in order to develop a reference database to support cultivar protection and breeding programs. An UPGMA dendrogram was generated using Jaccard's coefficients from a distance matrix based on 50 alleles of 12 microsatellite loci. The base pair number was estimated by the method of inverse mobility. The cophenetic correlation was 0.8. The accessions had a coefficient of similarity from 30 to 100%, which reflects high genetic variability. Three groups were observed in the UPGMA dendrogram; the first group was formed predominantly by foreign accessions, the second group was formed by Brazilian accessions, and the Dashehari accession was isolated from the others. The 50 microsatellite alleles did not separate all 103 accessions, indicating that there are duplicates in this mango collection. These 12 microsatellites need to be validated in order to establish a reliable set to identify mango cultivars.

Genetic diversity estimates based on morphological and molecular data can provide different information on the relationship between cultivars of a species. This study aimed to develop new microsatellite markers as additional tools in genetic studies on mangoes (Mangifera indica L.), and to analyze the genetic variability of 20 mango cultivars based on morphological descriptors and microsatellite markers. We aimed to better understand the cultivars enhanced breeding histories and to support crossbreeding planning. Positive clones were selected from a DNA library enriched for microsatellite regions for sequencing and primer design. Four plants of each of the 20 accessions were used for observations, based on 48 morphological descriptors. Twenty accessions were analyzed using 27 microsatellite markers, of which 16 were developed during this study. The clusters, based on the morphological descriptors by Ward-MLM strategy and the microsatellite markers, suggested that Brazilian mango cultivars have extensive genetic diversity and are related to cultivars with different provenances, demonstrating their different enhanced breeding histories.
