In plant tissue culture research, there is a constant need to search for novel substances that could result in better or more efficient growth in vitro. A relatively unknown compound, phloroglucinol (1,3,5-trihydroxybenzene), which is a degradation product of phloridzin, has growth-promoting properties. Phloroglucinol increases shoot formation and somatic embryogenesis in several horticultural and grain crops. When added to rooting media together with auxin, phloroglucinol further stimulates rooting, most likely because phloroglucinol and its homologues act as auxin synergists or auxin protectors. Of particular interest is the ability of phloroglucinol—a precursor in the lignin biosynthesis pathway—to effectively control hyperhydricity through the process of lignification, thus maximizing the multiplication rate of woody species and other species that are difficult to propagate. Phloroglucinol has also been used to improve the recovery of cryopreserved Dendrobium protocorms, increasing the potential of cryopreservation for application in ornamental biotechnology. Phloroglucinol demonstrates both cytokinin-like and auxin-like activity, much like thidiazuron, and thus has considerable potential for application in a wide range of plant tissue culture studies.

Phloroglucinol in plant tissue culture

Transgenic grape plants were regenerated from somatic embryos derived from leaves of in vitro-grown plants of 'Thompson Seedless' grape ( Vitis vinifera L.) plants. Somatic embryos were either exposed directly to engineered Agrobacterium tumefaciens or they were bombarded twice with 1-μm gold particles and then exposed to A. tumefaciens. Somatic embryos were transformed with either the lytic peptide Shiva-1 gene or the tomato ringspot virus (TomRSV) coat protein (CP) gene. After cocultivation, secondary embryos proliferated on Emershad/Ramming proliferation (ERP) medium for 6 weeks before selection on ERP medium containing 40 μg·mL -1 kanamycin (kan). Transgenic embryos were identified after 3 to 5 months under selection and allowed to germinate and develop into rooted plants on woody plant medium containing 1 μM 6-benzylaminopurine, 1.5% sucrose, 0.3% activated charcoal, and 0.75% agar. Integration of the foreign genes into these grapevines was verified by growth in the presence of kanamycin (kan), positive β- glucuronidase (GUS) and polymerase chain-reaction (PCR) assays, and Southern analysis.

Constructs with self-complementary sequences separated by an intron produce ''hairpin'' RNA (intron- hairpin-RNA (ihpRNA)) structures that efficiently elicit posttranscriptional gene silencing (PTGS). In the current study, the authors use this technology to confer resistance to plum pox virus (PPV) in herbaceous and woody perennial plants by silencing the PPV-coat protein (CP) gene. The authors confirmed the high capacity of ihpRNA constructs for inducing RNA silencing in Nicotiana benthamiana Domin., as more than 75% of the transformants displayed PTGS as evaluated by specific small interfering RNA (siRNA) production. The authors demonstrated that ihpRNA constructs provided PPV resistance, and they found a correlation between the length of the PPV sequence introduced in the ihpRNA constructs and the frequency of transgenic-resistant plants. Plants transformed with the full-length sequence produced a higher percentage of resistant lines. The authors further demonstrated for the first time that ihpRNA technology is applicable to a woody perennial species. A transgenic plum (Prunus domestica L.) PPV-CP ihpRNA line showed gene silencing characteristics (hypermethylation of the transgene sequence and specific siRNA production) and resistance to PPV infection 16 months after inoculation.

Plum Pox Virus Coat Protein Gene Intron-hairpin-RNA (ihpRNA) Constructs Provide Resistance to Plum Pox Virus in Nicotiana benthamiana and Prunus domestica

The effect of meta-Topolin (mT), an aromatic natural cytokinin, on micropropagation and adventitious shoot regeneration was evaluated on Prunus rootstocks, Torinel (Prunus domestica L.) and Ferdor (Prunus insititia  ×  domestica). In vitro shoots were grown for three subcultures on a multiplication medium containing 2.1, 4.2 or 6.3 μM of mT or 2.1 µM N
 6-benzyladenine (BA). Then, apical leaves were excised and transferred on a medium supplied with BA, thidiazuron (TDZ) or zeatin for adventitious regeneration. Shoots multiplied on 2.1 μM mT or BA, were also induced to root with α-naphthalene acetic acid and acclimatized. meta-Topolin did not improve shoot proliferation, respect to BA, however, positively influences growth and quality of shoots. Ferdor from mT showed higher rooting percentage (92 %), root number and length, respect to the control, while a similar response was observed in Torinel with both cytokinins. Acclimatisation was higher than 90 % for both genotypes and, after 5 months, the highest length of roots was found in plants from mT. Adventitious regeneration was obtained only in leaves from shoots previously grown on mT. The highest regeneration responses, 65 and 42 %, respectively for Ferdor and Torinel, were obtained in the regeneration medium supplied with TDZ.

Effect of meta-Topolin on micropropagation and adventitious shoot regeneration in Prunus rootstocks

An improved Agrobacterium-mediated transformation protocol for plum (Prunus domestica L.) hypocotyl slices was developed based on the addition of 2,4-d to the co-cultivation medium. This method increased transformation efficiency up to 10 × (42%) over previous reports with an average efficiency of 25% of hypocotyl slices producing transgenic plants. Timing of each step in the protocol was optimized producing self-rooted transgenic plants in the greenhouse in approximately 6 months. In order to test the system for its utility in functional genomic studies, we developed two hairpin constructs using a fragment of the peach (P. persica) Phytoene desaturase (PDS) gene. When A. tumefaciens with these constructs was used for targeted post-transcriptional gene silencing (PTGS), approximately 50% of the transformed plums were knockout PDS gene plants. The easy and efficient plum transformation protocol that we report here can be readily used for functional genomics studies in Prunus specifically, and Rosaceae and other woody species in general.

High transformation efficiency in plum (Prunus domestica L.): a new tool for functional genomics studies in Prunus spp.

We describe here an improved system for routinely developing transgenic plum plants (Prunus domestica L.) through the use of Agrobacterium tumefaciens. The production of non-transformed "escapes" has been virtually eliminated, and rates of plant establishment in the greenhouse have been dramatically improved. The system is based on the regeneration of shoots from hypocotyls extracted from mature seed. The shoot regeneration medium is Murashige and Skoog (MS) salts and vitamins supplemented with 7.5 microM thidiazuron and 0.25 microM indole-butyric acid. Transferring the explants after co-cultivation to shoot regeneration medium containing 80 mg l(-1) of kanamycin and 300 mg l(-1) of Timentin reduced the total number of regenerated shoots without affecting the transformation rate. Transformation rates using the described system averaged 1.2% of the hypocotyl slices producing transgenic plants, with a range of 0-4.2%. The transgenic shoots rooted at a rate of 90% on half-strength MS salts and vitamins supplemented with 5 microM alpha-naphthaleneacetic acid and 0.01 microM kinetin. Plantlets were transferred to a greenhouse directly from culture tubes with a 90% average survival.
