A simple isocratic HPLC technique was developed for the quantitative analysis of organic acids in dairy products. An Ammex HPX- 87 column at 65°C, 0.0090N H2SO4 mobile phase and UV detection at 220 and 275 nm were utilized. Orotic, citric, pyruvic, lactic, uric, formic, acetic, propionic, butyric, and hippuric acids were quantitated for whole milk, skim powder, cultured buttermilk, sour cream, cottage cheese, yogurt, sharp Cheddar cheese, and blue cheese. Over 90% recoveries of acids added to whole milk were observed for ail acids except butyrid; the average recovery for butyric was 86%.

Lactate Acid Inhibition of Salmonella Typhimurium in Yogurt

A simple and efficient process for the production of a ruminant feed supplement, rich in crude protein (defined as total N X 6.25), by bacterial fermentation of cheese whey has been developed. The lactose in unpasteurized whey is fermented to lactate acid by Lactobacillus bulgaricus at a temperature of 43 degrees C and pH 5.5. The lactic acid produced is continually neutralized with ammonia to form ammonium lactate. The fermented product is concentrated by evaporation to a solids content of about 70% and adjusted to pH 6.8 with additional ammonia. The concentrated product contains about 55% crude protein. Approximately 6 to 8% of the crude protein is derived from bacterial cells. 17% from whey proteins, and 75 to 77% from ammonium lactate. The efficiency of conversion of lactose to lactic acid usually exceeds 95%. The fermentation time is greatly reduced upon the addition of 0.2% yeast extract or 0.1% corn steep liquor as a source of growth factors. Whey containing lactose at concentrations up to 7% can be fermented efficiently, but at higher concentrations lactose is fermented incompletely. The process has been scaled up to a pilot plant level, and 40 tons of concentrated product were produced fro animal feeding trials, without ever encountering putrefactive spoilage.

Bacterial fermentation of cheese whey for production of a ruminant feed supplement rich in curde protein.

In experimental small-scale manufacture of Cheddar cheese a study was made of the movement of lactose, lactic acid, calcium and phosphorus between curd and whey, and of the effect of high acidity developed in the curd before whey separation, on the pH and physical properties of the cheese. It was found that while the curd particles remain in contact with the whey, the lactose fermented in the curd is replaced by lactose diffusing from the whey. Lactic acid produced in the curd diffuses into the whey rather slowly.When a high lactic acid level is attained rapidly the curd does not cheddar well, the pH of the cheese is low, the flavour is sour and bitter, the body is crumbly and the cheese is bleached. When a high lactic acid level is reached slowly, the curd also does not cheddar properly but the pH of the cheese may be within normal limits up to 3 weeks, at which age its body is coherent and its colour is normal. However, on maturing, the cheese becomes acid, the body crumbly and liquid separation occurs even at 2 months of age. This points to a risk in grading cheese at an early age.It is suggested that the abnormal behaviour in cheddaring of ‘high-acid’ curds is connected with an excessive loss of calcium. When high acidity is developed over an extended time in the whey, the curd retains more lactose but loses more phosphorus so that its buffering capacity is reduced. Cheese made from such curd becomes highly acid at maturity and this leads to the development of a bitter flavour.It is, therefore, concluded that both the rate of development of acidity, and the time the curd is in the whey, are critical factors in controlling the quality of Cheddar cheese.

Criteria for Judging a True Rumen Organism and a Description of Five Rumen Bacteria

A method is described for the colorimetric determination of lactic acid in milk and unsweetened milk products. The protein and lactose is removed after precipitation with copper sulphate and calcium hydroxide at 45° C, the lactic acid in an aliquot of the filtrate is oxidized to acetaldehyde in sulphuric acid in the presence of copper sulphate, a purple colour is developed with p–hydroxydiphenyl, and the colour intensity is measured in a photoelectric absorptiometer or other suitable instrument.Recoveries of from 96 to 103%, averaging 100%, were obtained when 20–400 mg. lactic acid/100 ml. were added to fresh milk.The proposed method has an advantage over existing methods in the ease and speed with which large numbers of determinations can be carried out simultaneously whilst retaining the essential accuracy.
