Transforming growth factor-β (TGF-β) is generally acknowledged to be the cytokine with the broadest range of activities in repair of injured tissue, based both on the variety of cell types that produce and/or respond to it and on the spectrum of its cellular responses (Roberts and Sporn, 1990). TGF-β is released from degranulating platelets and secreted by all of the major cell types participating in the repair process, including lymphocytes, macrophages, endothelial cells, smooth muscle cells, epithelial cells, and fibroblasts (see Fig. 1). A unique feature of this molecule is that its autoinduction results in sustained expression at the site of a wound and extends the effectiveness of both the initial burst of endogenous TGF-β released upon injury and exogenous TGF-β that might be applied to a wound. The ability of TGF-β to improve and/or accelerate tissue repair has been studied extensively in a variety of animal models of both normal and impaired healing. A limited number of clinical trials are in progress, but it is anticipated that many new applications for TGF-β will ultimately be found, once problems with appropriate timing and formulation can be solved.

Transforming Growth Factor-β

Macrophages play a key role in both normal and pathological processes involving immune and inflammatory responses, to a large extent through their capacity to secrete a wide range of biologically active molecules. To identify some of these as yet not characterized molecules, we have used a subtraction cloning approach designed to identify genes expressed in association with macrophage activation. One of these genes, designated macrophage inhibitory cytokine 1 (MIC-1), encodes a protein that bears the structural characteristics of a transforming growth factor β (TGF-β) superfamily cytokine. Although it belongs to this superfamily, it has no strong homology to existing families, indicating that it is a divergent member that may represent the first of a new family within this grouping. Expression of MIC-1 mRNA in monocytoid cells is up-regulated by a variety of stimuli associated with activation, including interleukin 1β, tumor necrosis factor α (TNF-α), interleukin 2, and macrophage colony-stimulating factor but not interferon γ, or lipopolysaccharide (LPS). Its expression is also increased by TGF-β. Expression of MIC-1 in CHO cells results in the proteolytic cleavage of the propeptide and secretion of a cysteine-rich dimeric protein of Mr 25 kDa. Purified recombinant MIC-1 is able to inhibit lipopolysaccharide -induced macrophage TNF-α production, suggesting that MIC-1 acts in macrophages as an autocrine regulatory molecule. Its production in response to secreted proinflammatory cytokines and TGF-β may serve to limit the later phases of macrophage activation.

MIC-1, a novel macrophage inhibitory cytokine, is a divergent member of the TGF-β superfamily

Following the initial purification and characterization of transforming growth factor-β (TGF-β) as a homodimeric, 25-kDa peptide (Frolik et al. 1983; Assoian et al. 1983; Roberts et al. 1983a), there has been an exponential increase in knowledge relating to this molecule. The cloning of TGF-β1 and the resultant elucidation of its precursor structure (Derynck et al. 1985) have led to the identification of at least four other forms of TGF-β and the definition of a larger gene family comprising many other structurally related, but functionally distinct, regulatory proteins. The original narrow definition of TGF-β, in terms of induction of a transformed phenotype in mesenchymal cells (Moses et al. 1981; Roberts et al. 1981, 1983 b), has now been supplanted by the knowledge that this peptide affects many functions in nearly all cells. Immunohistochemical and in situ hybridization studies have identified in vivo sites of TGF-β action; its broad spectrum of cellular targets as well as its multifunctional actions suggest that it has a pivotal control function in many physiological and pathological processes. Elucidation of physiological mechanisms of activation of the latent forms of the TGF-βs and of regulation of expression of the TGF-βs and their receptors are important problems that must still be solved for better understanding of the action of the TGF-βs. In this chapter we summarize current knowledge of the chemistry and complex biology of the growing family of TGF-βs.

The Transforming Growth Factor-βs

The transforming growth factor-β (TGF-β) is the prototype of the TGF-β family of growth and differentiation factors, which is encoded by 33 genes in mammals and comprises homo- and heterodimers. This review introduces the reader to the TGF-β family with its complexity of names and biological activities. It also introduces TGF-β as the best-studied factor among the TGF-β family proteins, with its diversity of roles in the control of cell proliferation and differentiation, wound healing and immune system, and its key roles in pathology, for example, skeletal diseases, fibrosis, and cancer.

TGF-β and the TGF-β Family: Context-Dependent Roles in Cell and Tissue Physiology

We have investigated the ability of exogenous transforming growth factor-beta (TGF-beta) to induce osteogenesis and chondrogenesis, critical events in both bone formation and fracture healing. Daily injections of TGF-beta 1 or 2 into the subperiosteal region of newborn rat femurs resulted in localized intramembranous bone formation and chondrogenesis. After cessation of the injections, endochondral ossification occurred, resulting in replacement of cartilage with bone. Gene expression of type II collagen and immunolocalization of types I and II collagen were detected within the TGF-beta-induced cartilage and bone. Moreover, injection of TGF-beta 2 stimulated synthesis of TGF-beta 1 in chondrocytes and osteoblasts within the newly induced bone and cartilage, suggesting positive autoregulation of TGF-beta. TGF-beta 2 was more active in vivo than TGF-beta 1, stimulating formation of a mass that was on the average 375% larger at a comparable dose (p less than 0.001). With either TGF-beta isoform, the dose of the growth factor determined which type of tissue formed, so that the ratio of cartilage formation to intramembranous bone formation decreased as the dose was lowered. For TGF-beta 1, reducing the daily dose from 200 to 20 ng decreased the cartilage/intramembranous bone formation ratio from 3.57 to zero (p less than 0.001). With TGF-beta 2, the same dose change decreased the ratio from 3.71 to 0.28 (p less than 0.001). These data demonstrate that mesenchymal precursor cells in the periosteum are stimulated by TGF-beta to proliferate and differentiate, as occurs in embryologic bone formation and early fracture healing.

Transforming growth factor-beta and the initiation of chondrogenesis and osteogenesis in the rat femur.

Human glioblastoma cells secrete a peptide, termed glioblastoma-derived T cell suppressor factor (G-TsF), which has suppressive effects on interleukin-2-dependent T cell growth. As shown here, complementary DNA for G-TsF reveals that G-TsF shares 71% amino acid homology with transforming growth factor-beta (TGF-beta). In analogy to TGF-beta it is apparently synthesized as the carboxy-terminal end of a precursor polypeptide which undergoes proteolytic cleavage to yield the 112 amino-acid-long mature form of G-TsF. Comparison of the amino-terminal sequence of G-TsF with that of porcine TGF-beta 2 and bovine cartilage-inducing factor B shows complete homology, which indicates that we have cloned the human analogue of these factors. It is tempting to consider a role for G-TsF in tumor growth where it may enhance tumor cell proliferation in an autocrine way and/or reduce immunosurveillance of tumor development.
