TL;DR: In-situ hybridization to interphase nuclei and chromosomes of Arabidopsis thaliana shows that there are four sites of rDNA in a diploid nucleus, and the strength of hybridization indicates that copy number is similar at both pairs of sites.

TL;DR: The number of major rDNA loci (the genes coding for 18S-5.8S-26S rRNA) was investigated in the economically important Brassica species and their wild relatives by in situ hybridization of an rDNA probe to metaphase chromosomes and interphase nuclei, indicating that the number of loci has been reduced during evolution.

Molecular Cytogenetics of the Genus Arabidopsis: In situ Localization of rDNA Sites, Chromosome Numbers and Diversity in Centromeric Heterochromatin

TL;DR: In situ hybridization is used to determine the number of sites of rDNA in species in the genus Arabidopsis, finding that a highly repeated para-centromeric sequence from A. thaliana, pAL1, is absent in the other three species, and the A.thaliana genome is not present or the centromeric DNA has evolved substantially in the polyploid species A. pumila and A. griffithiana.
