Publisher Summary This chapter describes sterile insect techniques (SIT). It is a biologically based method for the control of key insect pests. Wild female insects inseminated by released, radiation-sterilized males do not reproduce, and repeated releases of the sterilized insects lead to a reduction in pest population numbers. Effective control using sterile insects is achieved when they are used systematically as part of area-wide integrated pest management (AW-IPM) programs. SIT is species-specific, nonpolluting, and resistance-free. Since the original concept was developed in the United States in the 1940s, SIT has been used successfully for screwworm flies, tsetse flies, fruit flies, and moths. Technical progress in behavioral ecology, mass rearing, strain improvement, global information, positioning and monitoring systems, and aerial release, combined with economies of scale and a growing demand for pest-free and low-pesticide agricultural products in local and international trade, have increased the use of SIT in AW-IPM programs. These programs, by decreasing insecticide use, have also facilitated the use of biological control agents against secondary insect pests.

Sterile Insect Technique

Repeated DNA makes up a large fraction of a typical mammalian genome, and some repetitive elements are able to move within the genome (transposons and retrotransposons). DNA transposons move from one genomic location to another by a cut-and-paste mechanism. They are powerful forces of genetic change and have played a significant role in the evolution of many genomes. As genetic tools, DNA transposons can be used to introduce a piece of foreign DNA into a genome. Indeed, they have been used for transgenesis and insertional mutagenesis in different organisms, since these elements are not generally dependent on host factors to mediate their mobility. Thus, DNA transposons are useful tools to analyze the regulatory genome, study embryonic development, identify genes and pathways implicated in disease or pathogenesis of pathogens, and even contribute to gene therapy. In this review, we will describe the nature of these elements and discuss recent advances in this field of research, as well as our evolving knowledge of the DNA transposons most widely used in these studies.

DNA Transposons: Nature and Applications in Genomics

With world trade in agricultural commodities increasing, the introduction of exotic insects into new areas where they become pests will increase. The development and application of quarantine treatments or other mitigation approaches to prevent pest introduction in traded commodities raise many research and regulatory issues. The probit 9 standard for quarantine treatment efficacy has given way to risk-based alternatives. Varietal testing may have merit for some treatments or commodities but not for others. Development of generic treatments to control broad groups of insects or insects in all commodities can expedite new trade in agricultural products. Area-wide pest management programs lower pest levels before harvest and improve the quarantine security provided by any postharvest treatments. Systems approaches capitalize on cumulative pest mortality from multiple control components to achieve quarantine security in an exported commodity. Certain quarantine treatment technologies such as irradiation are not universally accepted, which is slowing their adoption. Standardized phytosanitary measures and research protocols are needed to improve the flow of information when countries propose to trade in a regulated commodity.

Current trends in quarantine entomology

The Sterile Insect Technique is a species-specific and environmentally friendly method of pest control involving mass release of sterilized insects that reduce the wild population through infertile matings. Insects carrying a female-specific autocidal genetic system offer an attractive alternative to conventional sterilization methods while also eliminating females from the release population. We exploited sex-specific alternative splicing in insects to engineer female-specific autocidal genetic systems in the Mediterranean fruit fly, Ceratitis capitata. These rely on the insertion of cassette exons from the C. capitata transformer gene into a heterologous tetracycline-repressible transactivator such that the transactivator transcript is disrupted in male splice variants but not in the female-specific one. As the key components of these systems function across a broad phylogenetic range, this strategy addresses the paucity of sex-specific expression systems (e.g., early-acting, female-specific promoters) in insects other than Drosophila melanogaster. The approach may have wide applicability for regulating gene expression in other organisms, particularly for combinatorial control with appropriate promoters.

Genetic Sexing Strains in Mediterranean Fruit Fly, an Example for Other Species Amenable to Large-Scale Rearing for the Sterile Insect Technique

The sterile insect technique (SIT) is currently being used for the control of many agricultural pests, including some lepidopteran species. The SIT relies on the rearing and release of large numbers of genetically sterile insects into a wild population. The holokinetic chromosomes of Lepidoptera respond differently to radiation than do species where there is a localized centromere. This difference has enabled a variation of the SIT to be developed for Lepidoptera where a substerilizing dose of radiation is given to the insects before their release with the result that a certain level of sterility is inherited by the F1 offspring. The development of genetic sexing strains for fruit flies, enabling the release of males only, has resulted in enormous economic benefits in the mass rearing and has increased the efficiency of the field operations severalfold. This article outlines Mendelian approaches that are currently available to separate large numbers of males and females efficiently for different lepidopteran species and describes their difficulties and constraints. Successful transgenesis in several lepidopteran species opens up new possibilities to develop genetic sexing strains. The proposal to develop genetic sexing strains described in this article takes advantage of the fact that in Lepidoptera, the female is the heterogametic sex, with most species having a WZ sex chromosome pair, whereas the males are ZZ. This means that if a conditional lethal gene can be inserted into the W chromosome, then all females should die after the application of the restrictive condition. The assumptions made to accommodate this model are discussed, and the advantages to be gained for control programs are elucidated.
