Infection structures of phytopathogenic fungi are modified hyphae specialized for the invasion of plant tissues. Initial events are adhesion to the cuticle and directed growth of the germ tube on the plant surface. At the site of penetration, appressoria are often formed that may have melanized walls and develop high turgor pressure to support the penetration process. The penetration hypha accumulates components of the cytoskeleton in the tip and secretes a variety of cell wall‐degrading enzymes in a highly regulated fashion in order to penetrate the cuticle and the plant cell wall. This article reviews recent papers on the cytology, physiology, and molecular biology of the penetration process.

Morphogenesis and mechanisms of penetration by plant pathogenic fungi

There is emerging evidence that the proteolytic machinery of plants plays important roles in defense against pathogens. The oomycete pathogen Phytophthora infestans, the agent of the devastating late blight disease of tomato (Lycopersicon esculentum) and potato (Solanum tuberosum), has evolved an arsenal of protease inhibitors to overcome the action of host proteases. Previously, we described a family of 14 Kazal-like extracellular serine protease inhibitors from P. infestans. Among these, EPI1 and EPI10 bind and inhibit the pathogenesis-related (PR) P69B subtilisin-like serine protease of tomato. Here, we describe EPIC1 to EPIC4, a new family of P. infestans secreted proteins with similarity to cystatin-like protease inhibitor domains. Among these, the epiC1 and epiC2 genes lacked orthologs in Phytophthora sojae and Phytophthora ramorum, were relatively fast-evolving within P. infestans, and were up-regulated during infection of tomato, suggesting a role during P. infestans-host interactions. Biochemical functional analyses revealed that EPIC2B interacts with and inhibits a novel papain-like extracellular cysteine protease, termed Phytophthora Inhibited Protease 1 (PIP1). Characterization of PIP1 revealed that it is a PR protein closely related to Rcr3, a tomato apoplastic cysteine protease that functions in fungal resistance. Altogether, this and earlier studies suggest that interplay between host proteases of diverse catalytic families and pathogen inhibitors is a general defense-counterdefense process in plant-pathogen interactions.

A Phytophthora infestans Cystatin-Like Protein Targets a Novel Tomato Papain-Like Apoplastic Protease

'Compatibility' describes the complementary relationship between a plant species and an adapted pathogen species that underlies susceptibility and which ultimately results in disease. Owing to elaborate surveillance systems and defence mechanisms on the plant side and a common lack of adaptation of many microbial pathogens, resistance is the rule and compatibility the exception for most plant-microbe combinations. While there has been major scientific interest in 'resistance' in the past decade, which has revealed many of its underlying molecular components, the analysis of 'compatibility', although intimately intertwined with 'resistance', has not been pursued with a similar intensity. Various recent studies, however, provide a first glimpse of the pivotal players and potential molecular mechanisms essential for compatibility in both the plant and parasite partners. In this review we highlight these findings with a particular emphasis on obligate biotrophic and hemibiotrophic fungal and oomycete pathogens and discuss novel strategies that might help to uncover further the molecular principles underlying compatibility to these highly specialized pathogens.

Purification and Characterization of Chitin Deacetylase from Colletotrichum lindemuthianum(

Cell-free extracts of UV-irradiated rice (Oryza sativa L.) leaves have a much greater capacity for the synthesis from geranylgeranyl pyrophosphate of diterpene hydrocarbons, including the putative precursors of rice phytoalexins, than extracts of unstressed leaves (KA Wickham, CA West [1992] Arch Biochem Biophys 293: 320-332). An elicitor bioassay was developed on the basis of these observations in which 6-day-old rice cell suspension cultures were incubated for 40 hours with the substance to be tested, and an enzyme extract of the treated cells was assayed for its diterpene hydrocarbon synthesis activity as a measure of the response to elicitor. Four types of cell wall polysaccharides and oligosaccharide fragments that have elicitor activity for other plants were tested. Of these, polymeric chitin was the most active; a suspension concentration of approximately 7 micrograms per milliliter gave 50% of the maximum response in the bioassay. Chitosan and a branched β-1,3-glucan fraction from Phytophthora megasperma f. sp. glycinea cell walls were only weakly active, and a mixture of oligogalacturonides was only slightly active. A crude mycelial cell wall preparation from the rice pathogen, Fusarium moniliforme, gave a response comparable to that of chitin, and this activity was sensitive to predigestion of the cell wall material with chitinase before the elicitor assay. N-Acetylglucosamine, chitobiose, chitotriose, and chitotetrose were inactive as elicitors, whereas a mixture of chitin fragments solubilized from insoluble chitin by partial acid hydrolysis was highly active. Constitutive chitinase activity was detected in the culture filtrate and enzyme extract of cells from a 6-day-old rice cell culture; the amount of chitinase activity increased markedly in both the culture filtrate and cell extracts after treatment of the culture with chitin. We propose on the basis of these results that soluble chitin fragments released from fungal cell walls through the action of constitutive rice chitinases serve as biotic elicitors of defense-related responses in rice.
