Methyl parathion (MP), an organophosphate pesticide licensed only for agricultural uses, was sprayed illegally for pest control in Mississippi and Ohio residences. To evaluate the association between MP exposure and neurobehavioral development, we assessed children 6 years or younger at the time of the spraying and local comparison groups of unexposed children using the Pediatric Environmental Neurobehavioral Test Battery (PENTB). The PENTB is composed of informant-based procedures (parent interview and questionnaires) and performance-based procedures (neurobehavioral tests for children 4 years or older) that evaluate cognitive, motor, sensory, and affect domains essential to neurobehavioral assessment. Children were classified as exposed or unexposed on the basis of urinary para-nitrophenol levels and environmental wipe samples for MP. Exposed children had more difficulties with tasks involving short-term memory and attention. Additionally, parents of exposed children reported that their children had more behavioral and motor skill problems than did parents of unexposed children. However, these effects were not consistently seen at both sites. There were no differences between exposed and unexposed children in tests for general intelligence, the integration of visual and motor skills, and multistep processing. Our findings suggest that MP might be associated with subtle changes to short-term memory and attention and contribute to problems with motor skills and some behaviors, but the results of the study are not conclusive.

Long-term neurobehavioral health effects of methyl parathion exposure in children in Mississippi and Ohio.

We developed an intelligent polymerized crystalline colloidal array (IPCCA) photonic crystal sensing material which reversibly senses the organophosphate compound methyl paraoxon at micromolar concentrations in aqueous solutions. A periodic array of colloidal particles is embedded in a poly-2-hydroxyethylacrylate hydrogel. The particle lattice spacing is such that the array Bragg-diffracts visible light. We utilize a bimodular sensing approach in which the enzyme organophosphorus hydrolase (OPH) catalyzes the hydrolysis of methyl paraoxon at basic pH, producing p-nitrophenolate, dimethylphosphate, and two protons. The protons lower the pH and create a steady-state pH gradient. Protonation of the phenolates attached to the hydrogel makes the free energy of mixing of the hydrogel less favorable, which causes the hydrogel to shrink. The IPCCA’s lattice constant decreases, which blueshifts the diffracted light. The magnitude of the steady-state diffraction blueshift is proportional to the concentration of methyl paraoxon. The current detection limit is 0.2 μmol methyl paraoxon per liter.

Photonic crystal sensor for organophosphate nerve agents utilizing the organophosphorus hydrolase enzyme

To evaluate the association between MP exposure and neurobehavioral development, children 6 years of age or younger at the time of the spraying and local comparison groups of unexposed children were assessed using the pediatric environmental neurobehavioral test battery (PENTB). The PENTB is composed of informant-based procedures (parent interview and questionnaires) and performance-based procedures (neurobehavioral tests for children 4 years of age or older) that evaluate each of the four broad domains (cognitive, motor, sensory, and affect) essential to neurobehavioral assessment. Children were classified as exposed or unexposed using urinary p-nitrophenol (PNP) levels and environmental wipe samples for MP. Exposure was defined as a urinary PNP level of greater than or equal to 100 ppb for the child or any other individual living in the household. Environmental wipe sample levels of greater than or equal to 150 g MP/100 cm2 and greater than or equal to 132.9 g MP/100 cm2 were used to define MP exposure for children living in Mississippi and Ohio, respectively. The PENTB was first administered in summer 1999 (year 1). The PENTB was readministered in summer 2000 (year 2) to children who participated in year 1 of the study. A description of the methods used in the study are presented. Results of data analyses for both years of the study will be presented in a separate publication.

Methods of assessing neurobehavioral development in children exposed to methyl parathion in Mississippi and Ohio.

A predictive system was developed and tested in a series of exercises with the objective of evaluating the preparedness and effectiveness of multi-agency response to food terrorism attacks. A computerized simulation model, Risk Reduction Effectiveness and Capabilities Assessment Program (RRECAP) was developed to identify the key factors that influence the outcomes of an attack and quantify the relative reduction of such outcomes caused by each factor. The model was funded by a grant from the U.S. Department of Agriculture. The exercises showed that RRECAP was an inexpensive and effective way for agencies to identify and prioritize their advance preparation to effectively mitigate such attacks. RRECAP also demonstrated the relative utility and limitations of the ability of medical resources to handle large numbers of patients if the attack is not recognized and mitigated rapidly; and exercise results showed that proper advance preparation would reduce these deficiencies to adequately handle all

The Chilean Altiplano ecosystem is conserved free from contaminants and pollutants because of the absence of major local human activities such as agriculture or other industries. We studied the effects of paraoxon on proliferation and apoptosis of testicular cells during active spermatogenesis in Dugesia gonocephala collected from a pristine river (Guacollo) in the Altiplano region nearby Visviri town, Chile.
