Grass carp is an important commercial fish widely cultivated in China. A wide range of temperatures, particularly extremely low temperatures, have dramatic effects on the aquaculture of this teleost. However, relatively few studies have characterized the molecular responses of grass carp exposed to acute cooling in natural environment. Here, we investigated the transcriptome profiles of the grass carp brain in response to cooling. Through regulation pattern analyses, we identified 2,513 differentially expressed genes (DEGs) that responded to moderate cold stress (12°C), while 99 DEGs were induced by severe low temperature (4°C).The pathway analyses revealed that the DEGs sensitive to moderate cold were largely enriched in steroid biosynthesis, spliceosome, translation, protein metabolism, phagosome, gap junction and estrogen signaling pathways. Additionally, we discerned genes most likely involved in low temperature tolerance, of which the MAPK signaling pathway was dominantly enriched. Further examination and characterization of the candidate genes may help to elucidate the mechanisms underpinning extreme plasticity to severe cold stress in grass carp.

Egg storage duration and breeder age are probably interacting to influence egg quality, hatchability, and hatchling quality. To evaluate this interaction, the impact of breeder age (31, 42, 66 weeks) and storage duration (2, 5, 12, 19 days) was investigated on broiler breeder eggs (Arbor Acres). Thick albumen diameter and pH increased, and yolk dry matter decreased between 2 and 19 days of storage. With the increase of breeder age from 31 to 66 weeks, albumen height, percentage and dry matter and shell percentage decreased and the egg weight and yolk percentage, dry matter and diameter increased. Prolonged egg storage increased the yolk pH in all breeder ages, but earlier and steeper in the oldest breeders. Prolonged egg storage resulted in a lower hatchability of set and fertile eggs due to a higher percentage of embryonic mortality. Early mortality increased earlier and steeper with prolonged egg storage in the oldest compared to younger breeders. Between 5 and 19 days of storage, yolk free body mass, liver and proventriculus + gizzard percentages decreased, as well as hatchling length and yolk efficiency (yolk absorption per initial yolk weight). The latter effects were most pronounced in the younger than in the older breeders. Therefore, eggs are preferably stored shorter than 7 d, but if long storage (≥12 days) cannot be avoided, we recommend to store eggs of older breeders when egg quality and hatchability are most important. In case hatchling quality is most important, it would be better to store eggs of younger breeders (31 weeks) for a prolonged period.

Interactions between egg storage duration and breeder age on selected egg quality, hatching results, and chicken quality

This study aimed to investigate the protective value of Eucommia ulmoides extract (EUE) on chicks under cold stress. A total of 21 compounds were identified in EUE using mass spectrometry (LC-MS). Ninety chicks were divided into a control group (CS) fed a basal diet and an experimental group supplemented with EUE, exposed to 10 ± 1 °C for 8 h per day. Results showed, compared with the CS group, the body weights (BW) (p < 0.01) and average daily gains ADG (p < 0.05) of the EUE group were increased throughout the study period. Chicks fed EUE had higher AFI (0–7 d, p < 0.001) and lower feed-to-gain ratios (F/G) (0–15 d, p < 0.001). EUE increased the activities of superoxide dismutase (SOD) (15 d, p < 0.05) and glutathione peroxidase (GSH-Px) (7 d, p < 0.05), whereas it decreased malondialdehyde (MDA) (15 d, p < 0.01). The contents of IgA (7 d, p < 0.05), IgG (7 d; 15 d, p < 0.01), and IgM (15 d, p < 0. 001) were higher in the EUE group. Dietary EUE could also reduce chick organ damage. Overall, EUE as a natural feed additive can improve the growth performance, antioxidant capacity, and immune level, and reduce the organ damage of cold-stressed chicks.

Dietary Eucommia ulmoides Extract Alleviates the Effect of Cold Stress on Chick Growth Performance, Antioxidant and Immune Ability.

Temperature tolerance restricts the distribution of a species. However, the molecular and cellular mechanisms that set the thermal tolerance limits of an organism are poorly understood. Here, we report on the function of dual-specificity phosphatase 1 (DUSP1) in thermal tolerance regulation. Notably, we found that dusp1-/- zebrafish grew normally but survived within a narrowed temperature range. The higher susceptibility of these mutant fish to both cold and heat challenges was attributed to accelerated cell death caused by aggravated mitochondrial dysfunction and over-production of reactive oxygen species in the gills. The DUSP1-MAPK-DRP1 axis was identified as a key pathway regulating these processes in both fish and human cells. These observations suggest that DUSP1 may play a role in maintaining mitochondrial integrity and redox homeostasis. We therefore propose that maintenance of cellular redox homeostasis may be a key mechanism for coping with cellular thermal stress and that the interplay between signaling pathways regulating redox homeostasis in the most thermosensitive tissue (i.e., gills) may play an important role in setting the thermal tolerance limit of zebrafish.
