The present invention is drawn to the production of fertile transformed wheat plants. The method involved subjecting wheat tissues to high velocity microprojectile bombardment, selecting for transformed cells, and regenerating stably transformed fertile plants from the transformed cells.

Methods for stable transformation of wheat

Only modest progress has been made in the molecular genetic improvement of wheat following the production of the first transgenic plants in 1992, made possible by the development of efficient, long-term regenerable embryogenic cultures derived from immature embryos and use of the biolistics method for the direct delivery of DNA into regenerable cells. Transgenic lines expressing genes that confer resistance to environmentally friendly non-selective herbicides, and pests and pathogens have been produced, in addition to lines with improved bread-making and nutritional qualities; some of these are ready for commercial production. Reduction of losses caused by weeds, pests and pathogens in such plants not only indirectly increases available arable land and fresh water supplies, but also conserves energy and natural resources. Nevertheless, the work carried out thus far can be considered only the beginning, as many difficult tasks lie ahead and much remains to be done. The challenge now is to produce higher-yielding varieties that are more nutritious, and are resistant or tolerant to a wide variety of biotic as well as abiotic stresses (especially drought, salinity, heavy metal toxicity) that currently cause substantial losses in productivity. How well we will meet this challenge for wheat, and indeed for other cereal and non-cereal crops, will depend largely on establishing collaborative partnerships between breeders, molecular biologists, biotechnologists and industry, and on how effectively they make use of the knowledge and insights gained from basic studies in plant biology and genetics, the sequencing of plant/cereal genomes, the discovery of synteny in cereals, and the availability of DNA-based markers and increasingly detailed chromosomal maps.

Molecular genetic improvement of cereals: transgenic wheat (Triticum aestivum L.)

Beginning in 1986, plants have been regenerated from protoplasts of all of the important cereal species, including wheat, rice, maize, and barley, and grasses such as sugarcane. In addition, somatic hybrids/cybrids as well as transgenic plants with introduced useful agronomic traits have been obtained in several instances. This rapid and impressive progress in the genetic manipulation of cereals has been made possible by two critical technical advances during the past decade: the establishment of embryogenic suspension cultures as a source of totipotent protoplasts and the direct delivery of DNA into protoplasts for genetic transformation

Factors Affecting Induction, Somatic Embryogenesis and Plant Regeneration of Callus from Cultured Immature Inflorescences of Sugarcane

This review focuses on the improvement of wheat (Triticum aestivum L.) via tissue and cell culture and its use in gene transfer techniques. Success of the latter critically depends on the ability to regenerate plants from cells or tissues cultured in vitro. Hence, we have devoted attention to the attempts made so far in obtaining regenerants from diverse explants. Although it is known that immature embryos are the best source for initiating morphogenic cultures, basic information related to the process of differentiation can also be gained by studying less responding tissues. The opportunity provided by anther and microspore culture in wheat improvement and the progress made is also presented. In Vitro Culture of Wheat and Genetic Transformation — Retrospect and Prospect

A friable and fast-growing type of callus was isolated from a long term shoot-competent cell culture of wheat. The suspension cultures established from this callus consisted of small, densely cytoplasmic cells which divided more rapidly but with a lower plant regeneration frequency than the original culture. A high yield of protoplasts was released from suspension cells (2 to 3×107 protoplasts per ml packed cell volume) when treated with enzyme mixtures. The isolated protoplasts divided at a relatively high frequency (20% to 50%) in both liquid and agarose-solidified KM8p medium. Up to 0.21% of the dividing protoplasts continued to divide and form micro-calli. Sixty-eight plants were regenerated from micro-calli, and among the 30 plants which were transplanted to the greenhouse, 3 have survived.
