TL;DR: It is demonstrated that a single glycol nucleic acid or 2′–5′-RNA modification can substantially reduce small interfering RNA (siRNA) seed-mediated binding to off-target transcripts while maintaining on-target activity in siRNAs with established hepatotoxicity.
