Current Strategies for Site-Directed RNA editing using ADARs
TL;DR: Though the precise and efficient re-direction of ADAR activity still remains a challenge, the systems that are being developed lay the foundation for SDRE as a powerful tool for transient genome editing.
Journal ArticleDOI
Highly Efficient Knockout of a Squid Pigmentation Gene
Karen Crawford,Karen Crawford,Juan Felipe Diaz Quiroz,Kristen M. Koenig,Namrata Ahuja,Caroline B. Albertin,Joshua J. C. Rosenthal +6 more
TL;DR: This paper demonstrates efficient gene knockout in the squid Doryteuthis pealeii using CRISPR-Cas9 and represents a critical advancement toward making squid genetically tractable.
Journal ArticleDOI
Spatially regulated editing of genetic information within a neuron.
Isabel C. Vallecillo-Viejo,Noa Liscovitch-Brauer,Juan Felipe Diaz Quiroz,Maria Fernanda Montiel-Gonzalez,Sonya E. Nemes,Kavita J. Rangan,Simon R. Levinson,Eli Eisenberg,Joshua J. C. Rosenthal +8 more
TL;DR: It is shown that ADAR2 (adenosine deaminase that acts on RNA), an RNA editing enzyme, is expressed outside of the nucleus in squid neurons, indicating that within a neuron RNA editing can recode genetic information in a region-specific manner.
Book ChapterDOI
In vitro and in cellula site-directed RNA editing using the λNDD-BoxB system.
TL;DR: A detailed protocol for in vitro and in cellula editing assays using the λNDD-BoxB system, one of the first systems developed for site-directed RNA editing, is presented in this paper.
