The mitochondrial cyt b gene plays a serious role in investigating untruthful meat species. This study aimed to authenticate the species of poultry products (Escallop, Nugget, Steak, and Sausage) depending on cyt b gene by using universal cyt b primer. DNA was isolated, and then a band of 359 bp of a mitochondrial cytochrome b gene was produced during the PCR amplification. The PCR products were exposed to Hinf1 and Rsa I restriction enzymes. The restricted fragments produced by restriction fragment length polymorphism technique (RFLP), were run by agarose gel electrophoresis. Results showed that all products had a similar band except sausage product does not follow the rule and showed mislabeling product by the REs, Two bands were yielded by HinfI RE for all products (114 and 245) bp with the differentiated sausage among other products based on the fake product (63 and 296) bp, while digestion by Rsa I produced three bands for escallop, nugget, and steak, (63, 100, 196), but only two bands for sausage was generated (148 and 211). As result, the study offered that analyzing meat products to detect the origin species via a PCR-RFLP technique by using these restriction enzymes can give reliable results. In short sausage is considered as fraud products because the results showed different bands as compared with poultry meat.

Identification and differentiation of poultry meat and products using pcr-rflp technique

This study was conducted at the University of Duhok, College of Agricultural Engineering Sciences, Animal Production Department. , to estimate the effect of different programs of adding the phytogenic (Optifeed, Oleobiotec and Veo premium) which are nutritional plant materials that have ability to improve the productive performance of broiler, and that in the phases of broiler feeding: starter, grower , finisher and for the whole period of the experiment, as well as, carcass characteristics and economic efficiency at 35 days of age. 1440 one-day old chicks were distributed to five groups with six replicates for each group (48 chicks/replicate). Adding dietary phytogenic materials significantly (P<0.05) improved broiler performance at grower, finisher and during whole experimental period (35) days compared to control. Regarding the carcass characteristics and internal organs, all treatments significantly (P<0.01) increased dressing percentage compared to control group (basal diet), while all other carcass characteristics and internal organs did not significantly affected by phytogenic dietary supplementation. Concerning economic efficiency, all phytogenic dietary supplementation resulted in improved profit compared to the basal diet.

Effect of different feed additives of optifeed, oleobiotec and veo premium on the productive performance of broilers under heat stress conditions in dohuk governorate

The quality and quantity of the extracted DNA are two key aspects for a successful PCR (Polymerase Chain Reaction) amplification. Moreover, reduction in time and cost required for DNA extraction are also two considerable factors, in cases when large number of samples are to be analyzed within a limited time-frame and budget. Accordingly, the aim of this study was to compare and optimize performance of five different DNA extraction methods by boiling meat tissues from cattle, buffalo, sheep, goat, chicken, camel, horse and dog in PBS (Phosphate Buffer Saline), distilled water, alkaline lysis buffers 1, 2 or 3. The results indicated that the boiling of meat and its products in alkaline lysis buffers was a good method to extract crude DNA. The optimized crude DNA extraction protocol was coupled with PCR-RFLP (Restriction Fragment Length Polymorphism) analysis for meat species differentiation. This developed workflow was tested on fifty-three commercial beef and mutton samples, out of which three samples were found to be adulterated. In conclusion, the rapid crude DNA extraction protocol has led to the development of a direct PCR-RFLP workflow that is simple, time-saving and cost-effective for PCR-based identification of different meat species.

Mitochondrial cytochrome b gene plays a serious role in studying adulteration of meat species. This research study designed to distinguish the raw meat species of sheep, goat, cattle and donkey using RFLP-PCR technique of a universal cyt b gene 359bp. Ten indigenous samples were collected from each animal in different parts of the body. All the samples were processed for DNA isolation and amplified with a Polymerase chain reaction (PCR). The amplicons were cleavage with HinfI and RsaI restriction enzymes, digestion of PCR product resulted in production of specific characterization bands for each species then analyzed by agarose electrophoresis. HinfI RE created three fragments for sheep, goat and cattle, with some similarities in a few bands between them, while yielded two bands for donkey. RsaI RE produced two bands for all species with different length except sheep and goats have the same length. Thus, results recommend that the RFLP-PCR technique with HinfI and RsaI play an important role to detect the animal meat species, since it is a fast, simple and easily handle method for identification of animal species.

Molecular detection of raw meat for some animal species using rflp-pcr technique

The study aimed to identify the species origin of imported buffalo meat from three countries Ukraine, Brazil and India to Erbil using PCR-RFLP technique. The pair of universal cyt b primer was designed to amplify a 359 bp DNA fragment in PCR amplification. Then the amplified fragments were digested with Hinf1 and Rsa I restriction enzymes, achieving a characteristics banding pattern in a 2% agarose which produced evidence to identify origin meat species. The results presented that digestion of samples with the Hinf1 RE, produced two bands in each (Ukraine and Brazil), (58 and 301) bp while it was showed different bands in Indian buffalo meat (85, 274) bp. On the other hand the outcomes in the Rsa I RE were two bands in Ukraine and Brazil (156, 203) bp and two bands were obtained in Indian buffalo meat (106, 253) bp. The results realized that the Indian buffalo meat species was not acceptable and showed mislabeling products. Thus, the obtained results recommend that the PCR-RFLP technique with HinfI and Rsa I restriction enzymes play an important role to detect the origin meat species, since it is a fast, simple and easily handle technique for detection of meat species.
