▪ Abstract Plant responses to salinity stress are reviewed with emphasis on molecular mechanisms of signal transduction and on the physiological consequences of altered gene expression that affect biochemical reactions downstream of stress sensing. We make extensive use of comparisons with model organisms, halophytic plants, and yeast, which provide a paradigm for many responses to salinity exhibited by stress-sensitive plants. Among biochemical responses, we emphasize osmolyte biosynthesis and function, water flux control, and membrane transport of ions for maintenance and re-establishment of homeostasis. The advances in understanding the effectiveness of stress responses, and distinctions between pathology and adaptive advantage, are increasingly based on transgenic plant and mutant analyses, in particular the analysis of Arabidopsis mutants defective in elements of stress signal transduction pathways. In this paper, we have reviewed how the hydraulic design of trees influences the movement of water from roots to leaves. The hydraulic architecture of trees can limit their water relations, gas exchange throughout the crown of trees, the distribution of trees over different habitats and, perhaps, even the maximum height that a particular species can achieve. Parameters of particular importance include: (1) the vulnerability of stems to drought-induced cavitation events because cavitation reduces the hydraulic conductance of stems, (2) the leaf specific conductivity-of stems because it determines the pressure gradients and most negative water potentials needed to sustain evaporation from leaves, (3) the water storage capacity of tissues because this might determine the ability of trees to survive long drought periods. All of these parameters are determined by the structure and function of anatomical components of trees. Some of the ecological and physiological trade-offs of specific structures are discussed.

Publisher Summary This chapter discusses a search for physiological causes and ecological consequence in reference with the variation in growth rate between higher plants. When grown under optimum conditions, plant species from fertile, productive habitats tend to have inherently higher relative growth rates (RGR) than species from less favorable environments. Under these conditions, fast-growing species produce relatively more leaf area and less root mass, which greatly contributes to their larger carbon gain per unit plant weight. Fast-growing species also have higher respiration rates per unit organ weight, due to demands of a higher RGR and higher rate of nutrient uptake. Fast-growing species have a greater capacity to acquire nutrients, which is likely to be a consequence, rather than the cause, of their higher RGR. This chapter analyses variations in morphological, physiological, chemical, and allocation characteristics underlying variation in RGR, to arrive at an appraisal of its ecological significance. The lower specific leaf area (SLA) of slow-growing species is because of the relatively high concentration of cell-wall material and quantitative secondary compounds, which may protect against detrimental abiotic and biotic factors. This chapter concludes that it is likely that there are trade-offs between growth potential and performance under adverse conditions, however, the current ecophysiological information explaining variation in RGR is too limited to support this contention quantitatively.

Inherent Variation in Growth Rate Between Higher Plants: A Search for Physiological Causes and Ecological Consequences

The Green Revolution boosted crop yields in developing nations by introducing dwarf genotypes of wheat and rice capable of responding to fertilisation without lodging. We now need a second Green Revolution, to improve the yield of crops grown in infertile soils by farmers with little access to fertiliser, who represent the majority of third-world farmers. Just as the Green Revolution was based on crops responsive to high soil fertility, the second Green Revolution will be based on crops tolerant of low soil fertility. Substantial genetic variation in the productivity of crops in infertile soil has been known for over a century. In recent years we have developed a better understanding of the traits responsible for this variation. Root architecture is critically important by determining soil exploration and therefore nutrient acquisition. Architectural traits under genetic control include basal-root gravitropism, adventitious-root formation and lateral branching. Architectural traits that enhance topsoil foraging are important for acquisition of phosphorus from infertile soils. Genetic variation in the length and density of root hairs is important for the acquisition of immobile nutrients such as phosphorus and potassium. Genetic variation in root cortical aerenchyma formation and secondary development (‘root etiolation’) are important in reducing the metabolic costs of root growth and soil exploration. Genetic variation in rhizosphere modification through the efflux of protons, organic acids and enzymes is important for the mobilisation of nutrients such as phosphorus and transition metals, and the avoidance of aluminum toxicity. Manipulation of ion transporters may be useful for improving the acquisition of nitrate and for enhancing salt tolerance. With the noteworthy exceptions of rhizosphere modification and ion transporters, most of these traits are under complex genetic control. Genetic variation in these traits is associated with substantial yield gains in low-fertility soils, as illustrated by the case of phosphorus efficiency in bean and soybean. In breeding crops for low-fertility soils, selection for specific root traits through direct phenotypic evaluation or molecular markers is likely to be more productive than conventional field screening. Crop genotypes with greater yield in infertile soils will substantially improve the productivity and sustainability of low-input agroecosystems, and in high-input agroecosystems will reduce the environmental impacts of intensive fertilisation. Although the development of crops with reduced fertiliser requirements has been successful in the few cases it has been attempted, the global scientific effort devoted to this enterprise is small, especially considering the magnitude of the humanitarian, environmental and economic benefits being forgone. Population growth, ongoing soil degradation and increasing costs of chemical fertiliser will make the second Green Revolution a priority for plant biology in the 21st century.

As may be judged from recent reviews,1' 2 the tonoplast (the vacuolar membrane) and the ectoplast (the external cytoplasmic membrane, or plasmalemma) have received far less attention than the other cytoplasmic membrane systems-the endoplasmic reticulum and the dictyosomes, or Golgi bodies-and the various cell organelles. A definite membranous boundary between the cytoplasm and the vacuole is frequently identifiable in electron micrographs and, since it occupies the position where the vacuolar membrane is expected to be, the bounding membrane is generally interpreted as the tonoplast.' This paper deals with such a membrane, and the latter is referred to as the tonoplast. Investigators of phloem search for evidence useful in answering the question as to whether a tonoplast exists in the mature sieve element. This question, as mentioned before,3 is a critical part of a larger one regarding the role of the sieveelement protoplast in translocation of organic solutes. In the following we examine the published conclusions on the tonoplast in sieve elements based on electron-microscope studies and supply a series of electron micrographs comparing the tonoplasts of sieve elements with those of nucleate cells in the phloem of Cucurbita maxima Duchesne. The material was prepared for electron microscopy according to one of the methods outlined in a previous paper:3 killing a 4-inch-long petiole in situ on the plant by injection of a 4 per cent solution of potassium permanganate into its central cavity, excising relevant parts, dehydrating with ethyl alcohol, and embedding in methacrylate. Critique of Literature and New Observations.-The meager information on the ultrastructure of the tonoplast' suggests that this membrane may be single or double, 70 to 100 A in thickness. In certain orientations it is said to resemble a ribbon. Statements that the tonoplast gives the impression-of being continuous around the vacuole;' that some observations suggest lack of continuity,' and that the tonoplast may be ruptured in fixation2 indicate considerable uncertainty about the integrity of the tonoplast as revealed by electron microscopy. In our preparations the tonoplast often showed discontinuities; some of these appeared as ruptures (Figs. 1 and 2) when compared with the normal discontinuities in the nuclear envelope (Fig. 2).
