Abstract: A library of 50,000 recombinants representative of the human X chromosome has been constructed. Human X chromosomes were physically separated using a fluorescence-activated cell sorter. The DNA was purified from the chromosomes, digested to completion with the restriction enzyme EcoRI and cloned into the phage lambda gtWES.lambda B. The X-derived nature of the recombinants was confirmed by hybridization to rodent/human cell line DNA containing only the human X chromosome. Such libraries will be particularly useful for the investigation of genetic diseases such as Duchenne muscular dystrophy, where the basic defect has not been elucidated, and of neoplasia, where several specific chromosomal anomalies, particularly for the leukaemias, have been identified.

Abstract: A library of cloned cDNAs representative of lymphocyte total poly(A)+ RNA was screened with total DNA probes at high clone density. 10% of the recombinants showed the presence of sequences which are repeated in the genome. Further analysis of six such isolated cDNA clones indicated that they contain different families of repetitive sequences with reiteration frequencies of between 150 and 45,000 copies per haploid genomes. Five of the six clones were found to contain single copy sequences as well as a repetitive sequence. cDNA clones containing repetitive sequences have been found to be derived from high, intermediate and low abundance classes of lymphocyte poly(A)+ RNA.

Abstract: A so-called '5-S mononucleosome' fraction was isolated from nuclei of Physarum polycephalum by digestion with micrococcal nuclease and subsequent fractionation by gel filtration. This fraction had electrophoretic and sedimentation properties which were similar to DNA of approximately 140 base pairs in length. It is shown that lysis of the nuclei activates a protease which is resistant to phenyl-methyl sulphonyl fluoride, o-phenanthroline and parachloromercuri benzene sulphonate and which subsequently degrades the nucleosomes.
