This article reviews the polyploidy events that have long been demonstrated to play a role in the evolution of Anura, while also discussing the importance of epigenetic control of gene expression and diversity. Findings on Brazilian autopolyploid anurans, mainly of the genus Odontophrynus, obtained in previous studies on their cytogenetics, chromatin ultrastructure, and molecular gene regulation are discussed here. Our data on genome duplication and on epigenetic events were analyzed here regarding phylogenetic trees, including the classic 2R model for vertebrate evolution and the growing evidence of similar epigenetic mechanisms in animal and allopolyploid plants. We propose that polyploidy and epigenetic events led to rapid Anura diversity and speciation. Also, recent advances in molecular studies in other organisms led us to revisit some controversial models of evolution.

Polyploidy and epigenetic events in the evolution of Anura.

A new species of frog of the genus Odontophrynus is described from grassland of the subtropical regions of Argentina, Brazil, and Paraguay based on morphological, osteological, bioacoustical, cytogenetic, and molecular evidence. The new species is characterized by having medium size; rostral and eye-nostril glandular warts well developed; several long glandular ridges, longitudinally oriented, on dorsum; several medium-sized, nearly rounded, dark-brown dorsal blotches; light mid-dorsal stripe continuous and well defined or restricted to the head and the urostyle regions; and mid-dorsal stripe, interorbital, and dorsolateral bands with a moderately whitish highlight, light brown, or reddish coloration. Additionally, the new species has a skull with nasals slightly separated both from each other and frontoparietals, and frontoparietal fontanelle not exposed. The advertisement call consists of a single multipulsed note with a dominant frequency of 820–1,121 Hz and a pulse rate of 99.3–140.7 pulses/s. The chromosomal complement is diploid with 2N = 2X = 22 chromosomes and nucleolar organizer regions on the interstitial regions of the short arm of chromosome pair 4. The phylogenetic analysis allowed us to recover the new species belonging to the O. americanus species group with genetic distances from its congeners ranging from 2.11 to 4.82% in a partial sequence of 16S ribosomal RNA gene. The new species is geographically isolated from the other currently known diploid species of the group, but in some localities is found in sympatry or syntopy with the tetraploid O. americanus.

A New Burrowing Frog of the Odontophrynus americanus Species Group (Anura, Odontophrynidae) from Subtropical Regions of Argentina, Brazil, and Paraguay

Pore-linked filaments were visualized in spreads of anuran spermatocyte nuclei using transmission electron microscope. We used Odontophrynus diplo and tetraploid species having the tetraploid frogs reduced metabolic activities. The filaments with 20-40 nm width are connected to a ring component of the nuclear pore complex with 90-120 nm and extend up to 1µm (or more) into the nucleus. The filaments are curved and connect single or neighboring pores. The intranuclear filaments are associated with chromatin fibers and related to RNP particles of 20-25 nm and spheroidal structures of 0.5µm, with variations. The aggregates of several neighboring pores with the filaments are more commonly observed in 4n nuclei. We concluded that the intranuclear filaments may correspond to the fibrillar network described in Xenopus oocyte nucleus being probably related to RNA transport. The molecular basis of this RNA remains elusive. Nevertheless, the morphological aspects of the spheroidal structures indicate they could correspond to nucleolar chromatin or to nucleolus-derived structures. We also speculate whether the complex aggregates of neighboring pores with intranuclear filaments may correspond to pore clustering previously described in these tetraploid animals using freeze-etching experiments.
