One of the most important branches of genetic engineering is the expression of recombinant proteins using biological expression systems. Nowadays, different expression systems are used for the production of recombinant proteins including bacteria, yeasts, molds, mammals, plants, and insects. Yeast expression systems such as Saccharomyces cerevisiae (S. cerevisiae) and Pichia pastoris (P. pastoris) are more popular. P. pastoris expression system is one of the most popular and standard tools for the production of recombinant protein in molecular biology. Overall, the benefits of protein production by P. pastoris system include appropriate folding (in the endoplasmic reticulum) and secretion (by Kex2 as signal peptidase) of recombinant proteins to the external environment of the cell. Moreover, in the P. pastoris expression system due to its limited production of endogenous secretory proteins, the purification of recombinant protein is easy. It is also considered a unique host for the expression of subunit vaccines which could significantly affect the growing market of medical biotechnology. Although P. pastoris expression systems are impressive and easy to use with well-defined process protocols, some degree of process optimization is required to achieve maximum production of the target proteins. Methanol and sorbitol concentration, Mut forms, temperature and incubation time have to be adjusted to obtain optimal conditions, which might vary among different strains and externally expressed protein. Eventually, optimal conditions for the production of a recombinant protein in P. pastoris expression system differ according to the target protein.

Pichia pastoris: A highly successful expression system for optimal synthesis of heterologous proteins

Medicinal plants are part and parcel of human society to combat diseases from the dawn of civilization. Terminalia chebula Retz. (Fam. Combretaceae), is called the ‘King of Medicine’ in Tibet and is always listed at the top of the list of ‘Ayurvedic Materia Medica’ because of its extraordinary power of healing. The whole plant possesses high medicinal value and traditionally used for the treatment of various ailments for human beings. Some of the folklore people used this plant in the treatment of asthma, sore throat, vomiting, hiccough, diarrhea, dysentery, bleeding piles, ulcers, gout, heart and bladder diseases. The plant has been demonstrated to possess multiple pharmacological and medicinal activities, such as antioxidant, antimicrobial, antidiabetic, hepatoprotective, anti-inflammatory, antimutagenic, antiproliferative, radioprotective, cardioprotective, antiarthritic, anticaries, gastrointestinal motility and wound healing activity. But no systematic updated information on the therapeutic effectiveness of Terminalia chebula, a popular herbal remedy in India and South-East Asia has so far been reported. This review highlights an updated information particularly on the phytochemistry and various pharmacological and medicinal properties of Terminalia chebula Retz. and some of its isolated compounds, along with their safety evaluation. This may provide incentive for proper evaluation of the plant as medicinal agent against the human diseases and also to bridge the lacunae in the existing literature and future scope which may offer immense opportunity for researchers engaged in validation of the traditional claims and development of safe and effective botanical medicine.

The development of Terminalia chebula Retz. (Combretaceae) in clinical research

Vaccination is the most successful application of immunological principles to human health. Vaccine efficacy needs to be reviewed from time to time and its safety is an overriding consideration. DNA vaccines offer simple yet effective means of inducing broad-based immunity. These vaccines work by allowing the expression of the microbial antigen inside host cells that take up the plasmid. These vaccines function by generating the desired antigen inside the cells, with the advantage that this may facilitate presentation through the major histocompatibility complex. This review article is based on a literature survey and it describes the working and designing strategies of DNA vaccines. Advantages and disadvantages for this type of vaccines have also been explained, together with applications of DNA vaccines. DNA vaccines against cancer, tuberculosis, Edwardsiella tarda, HIV, anthrax, influenza, malaria, dengue, typhoid and other diseases were explored.

DNA vaccines: roles against diseases.

Ethno pharmacological relevance: Traditional medicinal plants are practiced worldwide for treatment of arthritis especially in developing countries where resources are meagre. This review presents the plants profiles inhabiting throughout the world regarding their traditional usage by various tribes/ethnic groups for treatment of arthritis.
Materials and methods: Bibliographic investigation was carried out by analysing classical text books and peer reviewed papers, consulting worldwide accepted scientific databases from the last six decades. Plants/their parts/extracts/polyherbal formulations/toxicity studies for arthritis have been included in the review article. The profiles presented also include information about the scientific name, family, dose, methodology along with mechanism of action and toxicity profile. Research status of twenty potential plant species has been discussed. Further, geographical distribution of research, plants distribution according to families has been given in graphical form.
Results: 485 plant species belonging to 100 families, traditionally used in arthritis are used. Among 100 plant families, malvaceae constitute 16, leguminasae 7, fabaceae 13, euphorbiaceae 7, compositae 20, araceae 7, solanaceae 12, liliaceae 9, apocynaceae, lauraceae and rubiaceae 10, and remaining in lesser proportion. It was observed in our study that majority of researches are carried mainly in developing countries like India, China, Korea and Nigeria.
Conclusion: This review clearly indicates that list of medicinal plants presented in this review might be useful to researchers as well as practioners. This review can be useful for preliminary screening of potential anti- arthritis plants. Further toxicity profile given in the review can be useful for the researchers for finding the safe dose.

Medicinal plants with potential anti-arthritic activity:

The production of proteins in appropriate quantity and quality is an essential requirement of the present time. There appears to be a progressive increase in the application of mammalian cells for proteins production. Expression systems utilizing mammalian cells for recombinant proteins are able to introduce proper protein folding, post-translational modifications, and product assembly, which are important for complete biological activity. This review article is totally based on literature survey. In this article much emphasis has been done on the mammalian expression system. The author focused on different mammalian cell lines that express the gene. The different vector systems that transfer the gene into mammalian cells like plasmid based expression vectors, adenovirus vectors, vaccinia vectors, retroviral vector and baculovirus as vectors were explored. The processes for the transfer of gene into mammalian cells were also reviewed. Application and limitations of mammalian expression system were also focused. The purpose of research in writing this article is to create awareness in researchers, starting their career in gene expression related to mammalian cells. The principal result and major conclusion of this article is to make available the molecular technologies, expression system and applications of gene expression in mammalian cell lines.

Gene expression in Mammalian cells and its applications.

Here we report the biochemical composition and antibacterial activities of the leaves and flowers of four Lantana camara (Verbanaceae) plants with yellow, lavender, red, and white flowers. Three out of the four plants showed almost similar carbohydrates and lipid compositions. The levels of carbohydrates (mg/g dry weight) in the flowers were comparatively higher than in the leaves and the lipids content was relatively higher in the leaves except L. camara lavender and white. In lavender L. camara the amount of the total carbohydrates was very low. Proteins extracted from the leaves and flowers were subjected to electrophoresis. Electrophoresis of leaf proteins revealed similarity among L. camara yellow, red, and white flowers while that of flowers proteins showed similarity between L. camara yellow, lavender, red, and white. Extracts of leaves and flowers obtained with ethyl acetate were studied for antibacterial activities. Leaf and flower ethyl acetate extracts exhibited considerable antibacterial activities against the bacteria used where the value of zone of inhibition ranged from 10-21 and 9-15 mm, respectively. For flower and leaf ethyl acetate extracts, the zone of inhibition (mm) measured ranged from 10-21 and 9-15 mm, respectively. Thus, the study of the biochemical parameters viz., lipids, carbohydrates and proteins has revealed similarity among the four different L camara. However, antibacterial activities vary from one L. camara to the other and also according to the type of tissue used.

Biochemical compositions and antibacterial activities of Lantana camara plants with yellow, lavender, red and white flowers

Typhoid is endemic and in most developing countries remains a public health problem. Due to an increasing resistance to antibiotics and the limited scope of the vaccine the requirement is to explore the efficacy of natural plant products in the treatment of this disease. In this study we have evaluated the aqueous extract of Terminalia chebula in connection with the oxidative stress generated in Swiss albino mice by Salmonella typhimurium. Mice pretreated through the oral route with the water extract of T. chebula at a dose of 500 mg/kg (T500) body wt for a period of 30 d exhibited a full protection against 1X10 5 Colony forming units (CFU) of S. typhimurium injected intraperitonially. Mice pretreated with T500 for a period of 30 d followed with 50000 CFU of S. typhimurium showed a decrease in Xanthine oxidase activity by 31% and an increase in both glutathione peroxidase and glutathione reductase activity by 25% as compared to the infected saline treated control. The reduction in the oxidative stress indicated the effectiveness of the drug against S. typhimurium which can also be used against typhoid.
