In addition to the bacteria in the rumen there are many larger (5–250/mm long) organisms which at various times have been designated protozoa Of these the ‘ovals’ (Quin’s and Eadie’s) are now known to be large bacteria (Orpin, 1976) and the ‘flagellates’ Neocallimastix frontalis, Piromonas communis and Sphaeromonas communis are the zoospores of phycomycete fungi (Orpin, 1977a, b) There are genuine flagellates in the rumen, eg Trichomonas spp, Monoceromonas sp and Chilomastix sp, but almost nothing is known about their metabolism (Jensen and Hammon, 1964) The largest, most obvious and most important protozoa are the ciliates, of which there are two groups both in the subclass Trichostomatia The so called ‘holotrich’ protozoa belong to the order Vestibuliferida and the entodiniomorphs to the order Entodiniomorphida, suborder Entodiniomorphina and family Ophryoscolecidae As the properties and metabolism of these two protozoal groups are different, they will be considered separately below

The Rumen Protozoa

The subkingdom Protozoa now inclues over 65,000 named species, of which over half are fossil and approximately 10,000 are parasitic. Among living species, this includes approximately 250 parasitic and 11,300 free-living sarcodines (of which approximately 4,600 are foraminiferids); approximately 1,8000 parasitic and 5,100 free-living flagellates; approximately 5,600 parasitic "Sporozoa" (including Apicomplexa, Microspora, Myxospora, and Ascetospora); and approximately 2,5000 parasitic and 4,700 free-living ciliates. There are undoubtedly thousands more still unnamed. Seven phyla of PROTOZOA are accepted in this classification--SARCOMASTIGOPHORA, LABYRINTHOMORPHA, APICOMPLEXA, MICROSPORA, ASCETOSPORA, MYXOSPORA, and CILIOPHORA. Diagnoses are given for these and for all higher taxa through suborders, and reporesentative genera of each are named. The present scheme is a considerable revision of the Society's 1964 classification, which was prepared at a time when perhaps 48,000 species had been named. It has been necessitated by the acquisition of a great deal of nex taxonomic information, much of it through electron microscopy. It is hoped that the present classification incorporatesmost of the major changes that will be made for some time, and that it will be used for many years by both protozoologist and non-protozoologists.

A Newly Revised Classification of the Protozoa

are those who consider questions in science which have no unequivocal , experimentally determined answer scarcely worth discussing. Such feeling, along with conservatism, may have been responsible for the long and almost unchallenged dominance of the system of two kingdoms-plants and animals-in the broad classification of organisms. The unchallenged position of these kingdoms has ended, however; alternative systems are being widely considered (1-18) and are appearing in many introductory biology texts (19-24). My purpose in this article is to discuss the merits of two classifications which depart from the traditional two kingdoms, the systems of Copeland (1-3) and Whittaker (4, 5). Two-Kingdom System Man is terrestrial, and he sees around him two major groups of organisms of very different adaptation to nutrition on land-the photosynthetic, rooted, higher plants, and the food-ingesting, motile, higher animals. So distinct in way of life, direction of evolution, and kind of body organization are these groups that a concept of dichotomy-plants versus animals-is almost inescapable if they are considered by themselves. The two groups became the nuclei around which concepts of the plant and animal kingdoms were developed by early naturalists. The kingdoms have been part of the formal classification of living things since Linnaeus (25). Mosses, liverworts, and macroscopic algae are clearly plants in their photo-synthetic and nonmotile way of life, and (though the photosynthetic process itself was not understood by early naturalists) these forms were grouped 150 with the higher land plants. The higher fungi on land are nonmotile, and their apparently \"rooted\" manner of growth suggested the plants. It thus seemed reasonable to assign fungi to the plant kingdom, and some students believed that they had evolved from algae. The wealth of unicellular life discovered by microscopists offered greater difficulty. Some forms were motile and ingested food, however, and were naturally regarded as one-celled animals or proto-zoans. Others were nonmotile and photosynthetic, hence one-celled plants. There remained a wide range of uni-cellular forms in which nonmotility and flagellate or pseudopodial motility, and ingestive, photosynthetic, and absorp-tive nutrition, were combined in various ways which were neither clearly plant-like nor animal-like. In a number of cases plant-like and animal-like unicells were connected by a series of closely related intergrading forms within the same major taxon. New concepts of kingdoms of organisms

Isospora-type oocysts were excreted by cats following the ingestion of Toxoplasma fromn infected mice. Oocysts appeared 3 to 5 days after cyst. were ingested and 8 to 10 days after trophozoites were ingested, and also 21 to 24 days after the administration of infective fecal suspensions from cats. A close quanititative and biologic correlation between oocysts and Toxoplasma infectivity of the feces was observed which could not be separated by density gradient centrifugation and filtration methods. Toxoplasma is an intestinal coccidian of cats which is fecally spread. It has evolved to multiply in brain and muscle and in other species, making it possible for carnivorism to become another means of transmission.

Toxoplasma gondii in Cats: Fecal Stages Identified as Coccidian Oocysts

Coccidian oocysts resembling those of Isospora bigemina were excreted by cats fed Toxoplasma. In order to identify these oocysts with Toxoplasma infectivity a number of critical comparisons were made. The appearance of oocysts and Toxoplasma infectivity was simultaneous in the feces of 23 of 24 adult cats, 3-5 days after feeding of Toxoplasma cysts; in the feces of 4 out of 9 cats, 7-10 days after feeding of trophozoites; and in 8 out of 17 cats, 20-24 days after feeding of cat feces containing oocysts. Oocysts and infectivity were present in similar numbers, and they disappeared simultaneously from the feces of cats. Oocysts and infectivity were also observed simultaneously in the feces of 9 kittens, 1-2 days old, fed Toxoplasma cysts. Oocysts could not be separated from infectivity by filtration, by continuous particle electrophoresis, or by density gradient centrifugation. Excystation of oocysts was followed by an increase in titer of Toxoplasma infectivity. Unsporulated oocysts in fresh cat feces were noninfectious to mice, but oocyst sporulation was associated quantitatively with the development of infectivity at different temperatures and conditions of oxygenation. Maximum oocyst sporulation at 48 hr correlated with the development of maximum Toxoplasma infectivity. 1 and 2% sulfuric acid, and 2.5% potassium dichromate were found to be the best preservatives for sporulation of oocysts and for the development of Toxoplasma infectivity. Low sporulation rates in 0.1% formalin, 20% ethanol, and in water were associated with low infectivity in these reagents. Neither Toxoplasma infectivity nor oocysts developed in 0.3% formalin, 1% ammonium hydroxide, or 1% iodine in 20% ethanol. Oocysts, sporocysts, and sporozoites were stained specifically with Toxoplasma antibody in the indirect fluorescent antibody test. Typical coccidian stages, schizonts, and male and female gametocytes were found in the epithelium of the small intestine of kittens fed Toxoplasma cysts. The classification of T. gondii is discussed in relation to that of other isosporan coccidia of cats and dogs. The term "Toxoplasma oocyst" is introduced and Toxoplasma is classified in the family Toxoplasmidae of the suborder Eimeriina. The species Isospora bigemina is restricted to dogs, and I. cati to cats. I. felis and so-called I. rivolta from cats were noninfectious to dogs, and did not confer immunity to subsequent infection with I. canis and I. rivolta from dogs.

The toxoplasma gondii oocyst from cat feces

SYNOPSIS. A classification of the phylum Protozoa at supra-familial levels is given with definitions or descriptions of all of the involved taxa, some 140 in number. The scheme represents the result of the cooperative efforts of an international group of specialists and consultants who have‘been studying the overall problem for the past several years. Innovations of a nomenclatural nature have been held to a minimum, aside from the use of a system of uniform endings, but several major shifts of taxonomic significance have been introduced at all levels treated, including the subphyla. These revisions are explained in appropriately placed footnotes.
