PhysioBank, PhysioToolkit, and PhysioNet: components of a new research resource for complex physiologic signals.

According to classical concepts of physiologic control, healthy systems are self-regulated to reduce variability and maintain physiologic constancy. Contrary to the predictions of homeostasis, however, the output of a wide variety of systems, such as the normal human heartbeat, fluctuates in a complex manner, even under resting conditions. Scaling techniques adapted from statistical physics reveal the presence of long-range, power-law correlations, as part of multifractal cascades operating over a wide range of time scales. These scaling properties suggest that the nonlinear regulatory systems are operating far from equilibrium, and that maintaining constancy is not the goal of physiologic control. In contrast, for subjects at high risk of sudden death (including those with heart failure), fractal organization, along with certain nonlinear interactions, breaks down. Application of fractal analysis may provide new approaches to assessing cardiac risk and forecasting sudden cardiac death, as well as to monitoring the aging process. Similar approaches show promise in assessing other regulatory systems, such as human gait control in health and disease. Elucidating the fractal and nonlinear mechanisms involved in physiologic control and complex signaling networks is emerging as a major challenge in the postgenomic era.

Fractal dynamics in physiology: Alterations with disease and aging

There is evidence that physiological signals under healthy conditions may have a fractal temporal structure. Here we investigate the possibility that time series generated by certain physiological control systems may be members of a special class of complex processes, termed multifractal, which require a large number of exponents to characterize their scaling properties. We report on evidence for multifractality in a biological dynamical system, the healthy human heartbeat, and show that the multifractal character and nonlinear properties of the healthy heart rate are encoded in the Fourier phases. We uncover a loss of multifractality for a life-threatening condition, congestive heart failure.

Multifractality in human heartbeat dynamics

The human brain spontaneously generates neural oscillations with a large variability in frequency, amplitude, duration, and recurrence. Little, however, is known about the long-term spatiotemporal structure of the complex patterns of ongoing activity. A central unresolved issue is whether fluctuations in oscillatory activity reflect a memory of the dynamics of the system for more than a few seconds. We investigated the temporal correlations of network oscillations in the normal human brain at time scales ranging from a few seconds to several minutes. Ongoing activity during eyes-open and eyes-closed conditions was recorded with simultaneous magnetoencephalography and electroencephalography. Here we show that amplitude fluctuations of 10 and 20 Hz oscillations are correlated over thousands of oscillation cycles. Our analyses also indicated that these amplitude fluctuations obey power-law scaling behavior. The scaling exponents were highly invariant across subjects. We propose that the large variability, the long-range correlations, and the power-law scaling behavior of spontaneous oscillations find a unifying explanation within the theory of self-organized criticality, which offers a general mechanism for the emergence of correlations and complex dynamics in stochastic multiunit systems. The demonstrated scaling laws pose novel quantitative constraints on computational models of network oscillations. We argue that critical-state dynamics of spontaneous oscillations may lend neural networks capable of quick reorganization during processing demands.

Long-Range Temporal Correlations and Scaling Behavior in Human Brain Oscillations

Detrended fluctuation analysis ~DFA! is a scaling analysis method used to quantify long-range power-law correlations in signals. Many physical and biological signals are ‘‘noisy,’’ heterogeneous, and exhibit different types of nonstationarities, which can affect the correlation properties of these signals. We systematically study the effects of three types of nonstationarities often encountered in real data. Specifically, we consider nonstationary sequences formed in three ways: ~i! stitching together segments of data obtained from discontinuous experimental recordings, or removing some noisy and unreliable parts from continuous recordings and stitching together the remaining parts—a ‘‘cutting’’ procedure commonly used in preparing data prior to signal analysis; ~ii! adding to a signal with known correlations a tunable concentration of random outliers or spikes with different amplitudes; and ~iii! generating a signal comprised of segments with different properties—e.g., different standard deviations or different correlation exponents. We compare the difference between the scaling results obtained for stationary correlated signals and correlated signals with these three types of nonstationarities. We find that introducing nonstationarities to stationary correlated signals leads to the appearance of crossovers in the scaling behavior and we study how the characteristics of these crossovers depend on ~a! the fraction and size of the parts cut out from the signal, ~b! the concentration of spikes and their amplitudes ~c! the proportion between segments with different standard deviations or different correlations and ~d! the correlation properties of the stationary signal. We show how to develop strategies for preprocessing ‘‘raw’’ data prior to analysis, which will minimize the effects of nonstationarities on the scaling properties of the data, and how to interpret the results of DFA for complex signals with different local characteristics.

Effect of nonstationarities on detrended fluctuation analysis.

We propose a general approach to the question of how biological rhythms spontaneously self-regulate, based on the concept of "stochastic feedback". We illustrate this approach by considering at a coarse-grained level the neuroautonomic regulation of the heart rate. The model generates complex dynamics and successfully acounts for key characteristics of cardiac variability, including the l/f power spectrum, the functional form and scaling of the distribution of variations, and correlations in the Fourier phases indicating nonlinear dynamics.

Stochastic feedback and the regulation of biological rhythms

We study the depinning transition for models representative of each of the two universality classes of interface roughening with quenched disorder. For one of the universality classes, the roughness exponent changes value at the transition, while the dynamical exponent remains unchanged. We also find that the prefactor of the width scales with the driving force. We propose several scaling relations connecting the values of the exponents on both sides of the transition, and discuss some experimental results in light of these findings.
