Immobilized cells : a review of recent literature

The use of monoliths in automotive emission control systems has dominated the applications of monoliths. However, other applications have also become of interest due to unique features of monoliths. Several excellent reviews on emission control [1–3] and catalytic oxidations [4] have been published during the last few years. Deluca and Campbell [5] have reviewed monoliths used as catalyst supports. This paper reviews flow characterization, heat and mass transfer, mathematical modeling, properties of monolithic supports, and nonautomobile applications of monoliths.

Monolithic Catalysts for Nonautomobile Applications

For a wide range of purposes, primary cell cultures and/or cell lines have been prepared from most tissues and organs of a small fraction of the estimated 20,000 species of bony fish. These cell cultures usually have been maintained with mammmalian sera. For many applications their usefulness would be enhanced by a more piscine and defined environment. However, the piscine equivalents of mammalian polypeptide growth and differentiation factors are largely unknown and are unlikely ever to be available commercially. In the future they might be obtained from the medium in which fish cells have been grown. Therefore, by being a potential source of fish polypeptide growth and differentiation factors, a cell line from a fish organ might be utilized as a Rossetta stone to decipher thein vitro proliferation and differentiation of other cells from this or other organs from the same or different species.

Technology and uses of cell cultures from the tissues and organs of bony fish

Respiration rates in Spodoptera frugiperda (Sf-9) cell bioreactor cultures were successfully measured on-line using two methods: The O(2) uptake rate (OUR) was determined using gas phase pO(2) values imposed by a dissolved oxygen controller and the CO(2) evolution rate (CER) was measured using an infrared detector. The measurement methods were accurate, reliable, and relatively inexpensive. The CER was routinely determined in bioreactor cultures used for the production of several recombinant proteins. Simple linear relationships between viable cell densities and both OUR and CER in exponentially growing cultures were used to predict viable cell density. Respiration measurements were also used to follow the progress of baculoviral infections in Sf-9 cultures. Infection led to increases in volumetric and per-cell respiration rates. The relationships between respiration and several other culture parameters, including viable cell density, cell protein, cell volume, glucose consumption, lactate production, viral titer, and recombinant beta-galactosidase accumulation, were examined. The extent of the increase in CER following infection and the time postinfection at which maximum CER was attained were negatively correlated with the multiplicity of infection (MOI) at multiplicities below the level required to infect all the cells in a culture. Delays in the respiration peak related to the MOI employed were correlated with delays in the peak in recombinant protein accumulation. DO levels in the range 5-100% did not exert any major effects on viable cell densities, CER, or product titer in cultures infected with a baculovirus expressing recombinant beta-galactosidase.

A ceramic matrix having a high degree of surface area per unit volume is shown to have significant utility in the large scale culture of animal cells. The surfaces of the ceramic provide for the adhesion and growth of a wide variety of cells to densities equal to or greater than obtained with other methods such as roller bottles or microcarriers. Utilizing an automated system controlling pH and dissolved oxygen, scale-up from 0.9 m2 to 18.5 m2 of surface area was accomplished with no losses in efficiency of surface utilization. The density of Vero cells after 7–8 days culture under standard conditions averaged 6.6 × 105 cells/cm2 for each size of ceramic. Methods providing continuous monitoring of the culture through analysis of the cellular oxygen consumption rate and complete cell harvesting are described.

Ceramic Matrix for Large Scale Animal Cell Culture

A novel immobilized cell composite used in a novel apparatus/method for cell culture. The composite comprises: (a) a high surface area monolithic support having a multiplicity of mutually parallel channels passing therethrough, the channels having walls formed of a medium-insoluble, non-toxic, composition and the support having at least about 20 channels per square inch of cross-sectional area; and (b) a population of plant or animal tissue cells anchored to the porous channel walls.
