The present invention encompasses a solid dose delivery vehicle for ballistic administration of a bioactive material to subcutaneous and intradermal tissue, the delivery vehicle being sized and shaped for penetrating the epidermis. The delivery vehicle further comprises a stabilizing polyol glass loaded with the bioactive material and capable of releasing the bioactive material in situ. The present invention further includes methods of making and using the solid dose delivery vehicle of the invention.

Systemic delivery of parathyroid hormone to a mammalian host is accomplished by inhalation through the mouth of a dispersion of an N-terminal fragment of PTH. It has been found that such respiratory delivery of the PTH fragment provides a pulsatile concentration profile of the PTH in the host's serum. PTH fragment compositions include dry powder formulations having the PTH present in a dry bulking powder, liquid solutions or suspensions suitable for nebulization, and aerosol propellants suitable for use in a metered dose inhaler.

Pulmonary delivery of active fragments of parathyroid hormone

Poly-3-hydroxybutyric acid is separated from bacterial cells by drying a finely divided stream or spray of an aqueous suspension of the cells with a gas heated to above 100°C and then extracting the PHB, preferably after a lipid extraction step with a solvent such as methanol or acetone, with a PHB- solvent such as a partially halogenated hydrocarbon such as 1,2-dichloroethane, chloroform or dichloromethane. 1,2-Dichloroethane may also be used to extract PHB directly from an aqueous cell suspension without an intermediate drying step. In this case, for some bacteria, the suspension is preferably subjected to a cell disruption step, eg milling, prior to contact with the 1,2-dichloroethane.
