Complete nucleotide sequence and genome organization of a Chinese isolate of Tobacco vein distorting virus.

Tobacco bushy top disease is caused by a complex of the viruses tobacco bushy top virus (TBTV, a member of the genus Umbravirus) and tobacco vein distorting virus (TVDV, a member of the genus Polerovirus), which acts as a helper virus encapsidating the TBTV genomic RNA. RNA from purified virions is separated as five bands. The two largest (6.0 and 4.2 kb) were shown by Northern blot analysis to be the genomic RNAs of TVDV and TBTV, respectively. A band of about 3 kb was cloned and sequenced and shown to be the RNA of a previously undescribed virus with two open reading frames (ORFs), the second of which is an RNA-dependent RNA polymerase (RdRp) and is probably expressed by readthrough of the ORF1a stop codon. BLAST and phylogenetic analyses of the RdRp show that it is related to two RNAs previously reported in association with the poleroviruses Beet western yellows virus and Carrot red leaf virus. These three RNAs appear to represent species of a new genus of plant viruses dependent upon a helper polerovirus for their transmission.

Molecular identification and phylogenetic analysis of a viral RNA associated with the Chinese tobacco bushy top disease complex

Panax notoginseng is normally cultivated in acidic red soil with high soluble Al, however, the distribution pattern of Al in P. notoginseng is unknown. The aim of this study is to investigate the Al distribution in different organs of P. notoginseng grown in different acid soil, in order to get a deeper insight into the Al tolerance mechanism in this species. P. notoginseng was sampled from different sites and subjected to Al determination. Al tolerance and accumulation were investigated by exposing the seedlings to different Al concentrations in hydroponic solution. The expression of pectin methylesterase genes was determined by quantitative RT-PCR. The soil pH ranged from 4.68 to 6.13. The total Al concentration in soil was 76.7–168.5 g·kg−1 and active Al was 1.34–5.01 g·kg−1. Al concentration differed with organs of P. notoginseng, following the order: rootlet > rhizome > main root > leaf and stem. Furthermore, Al concentration in the cell wall, organelle and cytoplasmic supernatant of P. notoginseng was approximately 15:2:1 and most Al in the cell wall was bound to pectin component. Al treatment significantly increased the content of pectin and enhanced the expression of PME3, PME7, and PME40 of P. notoginseng root. P. notoginseng was able to grow under high active Al condition and accumulated high Al in its underground parts. The pectin component in the cell wall was the major binding site of Al of P. notoginseng root.

Distribution pattern of aluminum in Panax notoginseng, a native medicinal plant adapted to acidic red soils

The strain ZK7 of Pochonia chlamydosporia var. chlamydosporia and IPC of Paecilomyces lilacinus are highly effective in the biological control against root-knot nematodes infecting tobacco. When applied, they require a specific monitoring method to evaluate the colonization and dispersal in soil. In this work, the randomly amplified polymorphic DNA (RAPD) technique was used to differentiate between the two individual strains and 95 other isolates, including isolates of the same species and common soil fungi. This approach allowed the selection of specific fragments of 1.2 kb (Vc1200) and 2.0 kb (Vc2000) specific for ZK7, 1.4 kb (P1400) and 0.85 kb (P850) specific for IPC, using the random Primers OPL-02, OPD-05, OPD-05 and OPC-11, respectively. These fragments were cloned, sequenced, and used to design sequence-characterized amplification region (SCAR) primers specific for the two strains. In classical polymerase chain reaction (PCR), with serial dilution of ZK7 and IPC pure culture DNAs template, the detection limits of these oligonucleotide SCAR-PCR primers were found to be 10, 1000, 500, 100 pg, respectively. In the dot blotting, digoxigenin (DIG)-labeled amplicons from these four primers specifically recognized the corresponding fragments in the DNAs template of these two strains. The detection limit of these amplicons were 0.2, 0.2, 0.5, 0.5 μg, respectively.
