Iron-Catalyzed Oxidative Coupling of Indoline-2-ones with Aminobenzamides via Dual C-H Functionalization.

Synthesis of new derivatives of 2-amino-4H-pyrans is reported via three-component reaction of α-ketoesters, active methylenes, and OH-acids in the presence of catalytic amount of piperidine at room temperature with high to excellent yields. The structure of synthesized compounds was characterized by FTIR, 1H, 13C NMR spectroscopy, and mass spectrometry. Simplicity of procedure, mild reaction conditions, short reaction time, and easy separation of the products make this an interesting alternative to other reported approaches. Also, their antibacterial and antioxidant activities were evaluated against Staphylococcus aureus and Bacillus subtilis as Gram-positive bacteria and Escherichia coli and Pseudomonas aeruginosa as Gram-negative bacteria, as well as the radical scavenger DPPH. Among these compounds, 4l including CO2Et substituent and 6-methyl-2H-pyran-2-one moiety showed the highest antioxidant and antibacterial activities.

The invention relates to the technical field of organic compound synthesis, in particular to an intermediate of a compound with anti-cancer activity as well as a preparation method and application ofthe intermediate. The structural formula of the intermediate of the compound with the anti-cancer activity is shown in the specification, wherein R1 is selected from H and R2 is selected from hydroxyland diazonium salt groups; and n is selected from 1, 2 and 3. The invention provides the intermediate of the compound with anticancer activity, a central skeleton can be constructed through one-stepcascade reaction, the raw materials are cheap and easy to obtain, the catalyst cost is low, and the preparation method has the advantages of good selectivity, high effective conversion rate and mild reaction conditions. The intermediate can be used for preparing the compound with anti-cancer activity, and when the intermediate is used for preparing the compound with anti-cancer activity, the totalyield of the whole synthetic route is 55.3% and is increased by more than two times compared with the yield of the existing synthetic route.
