Effects of two estradiol esters (benzoate and cypionate) on the induction of synchronized ovulations in Bos indicus cows submitted to a timed artificial insemination protocol

This experiment was designed to test the hypothesis that delayed insemination of nonestrous cows would increase pregnancy rates when using sex-sorted semen in conjunction with fixed-time artificial insemination (FTAI). Estrus was synchronized for 656 suckled beef cows with the 7-d CO-Synch + controlled internal drug release (CIDR) protocol (100 μg GnRH + CIDR [1.38 g progesterone] on d 0, 25 mg PGF2α at CIDR removal on d 7, and 100 μg GnRH on d 10, 66 h after CIDR removal). Estrus detection aids (Estrotect) were applied at PGF2α and CIDR removal on d 7, and estrous expression was recorded at GnRH on d 10. Cows were assigned to 1 of 3 treatments: 1) FTAI (concurrent with GnRH, 66 h after CIDR removal) with conventional semen regardless of estrous expression, 2) FTAI with sex-sorted semen regardless of estrous expression, or 3) FTAI with sex-sorted semen for cows having expressed estrus and delayed AI 20 h after final GnRH for cows failing to express estrus. A treatment × estrous expression interaction was found (P < 0.0001). Higher pregnancy rates (P < 0.0001) were achieved with conventional semen (Treatment 1; 77%) than with sex-sorted semen (Treatments 2 and 3; 51 and 42%, respectively) among cows that expressed estrus. However, among cows that failed to express estrus, delayed insemination with sex-sorted semen yielded higher (P < 0.0001) pregnancy rates than with sex-sorted semen at the standard time (Treatments 2 and 3; 3 versus 36%, respectively). Furthermore, among cows that failed to express estrus, FTAI pregnancy rates when using sex-sorted semen at the delayed time (36%) were comparable (P = 0.9) to those achieved using conventional semen at the standard time (Treatment 1; 37%). These results indicate that delaying AI of nonestrous cows by 20 h from the standard FTAI improves pregnancy rates when sex-sorted semen is used with FTAI.

There is speculation that beef bull semen quality is inferior to that of dairy bulls although few scientific studies are available in the literature. The aim of this study was to evaluate sperm quality in beef bull semen and to determine which parameters could be indicative of fertility after insemination. Sperm quality, assessed by computer assisted sperm motility analysis and flow cytometric evaluation of membrane integrity, levels of reactive oxygen species, mitochondrial membrane potential, acrosome status and DNA fragmentation index, was evaluated in beef and dairy bull semen. For beef bulls, normal morphology (r = 0.62, P < 0.05) and WOBBLE (r = 0.57, P < 0.05) were significantly correlated with 56-day non-return rate, whereas sperm quality was not significantly correlated with the fertility index score for dairy bulls. Membrane integrity (46 ± 8.0% versus 40 ± 11%, P < 0.05), normal morphology (87 ± 6% versus 76 ± 8%; P < 0.05), and high respiratory activity (52 ± 13 versus 12 ± 4%; P < 0.001) were higher for dairy bulls than for beef bulls. The DNA fragmentation index was lower for dairy bull spermatozoa than beef (3.8 ± 1.1% versus 6.1 ± 2.9%; P < 0.01), whereas some sperm kinematics were higher. Multivariate analysis indicated that type of bull (beef versus dairy) had an impact on sperm quality. Different assays of sperm quality may be needed for appropriate analysis of beef and dairy bull semen. These finding could be important for cattle breeding stations when evaluating semen quality.

Effective use of SexedULTRA™ sex-sorted semen for timed artificial insemination of beef heifers

Three experiments were designed to evaluate methods to optimize the use of sex-sorted sperm in timed AI (TAI) programs for suckled beef cows. In all 3 experiments, suckled Bos indicus cows were synchronized using an intravaginal progesterone (P4) device during 8 d and a 2.0-mg injection of intramuscular estradiol benzoate (EB) at device insertion. The females received PG and eCG (300 IU) at P4 device removal and 1.0 mg of EB 24 h later. The cows were inseminated 60 to 64 h after P4 device withdrawal. All cows had their ovaries scanned by transrectal ultrasound at TAI to indentify and to measure the largest follicle (LF) present. In Exp. 1, a total of 853 cows had their LF classified as <9 mm or ≥9 mm at the time of TAI; these cows were then randomly assigned to 4 groups according to their LF diameter (<9 mm or ≥9 mm) and the type of sperm used (sex-sorted or non-sex-sorted). There was an interaction (P = 0.02) between the type of sperm and LF diameter beginning at TAI[non-sex-sorted ≥9 mm = 58.9%a (126/214); non-sex-sorted <9 mm = 49.5%b (106/214);sex-sorted ≥9 mm = 56.8%ab (134/236); and sex-sorted <9 mm = 31.2%c (59/189), a≠b≠c = P < 0.05]. In Exp. 2, suckled cows (n = 491) were classified immediately before TAI as having displayed estrus or not (estrus or no estrus) between P4 device removal and TAI. These cows were randomly assigned to 4 groups according to the occurrence of estrus and the type of sperm (sex-sorted or non-sex-sorted). There were effects of the occurrence of estrus (P = 0.0003) and the type of sperm (P = 0.05) on pregnancy per AI [P/AI; no estrus, non-sex-sorted = 43.6% (27/62); estrus, non-sex-sorted = 58.5%; (107/183); no estrus, sex-sorted = 33.9% (21/62), and estrus, sex-sorted = 50.0% (92/184)]; however, no interaction between the occurrence of estrus and type of sperm was observed (P = 0.87). In Exp. 3, a total of 200 suckled cows presenting LF ≥9 mm at TAI were randomly assigned to receive sex-sorted sperm deposited into the uterine body (n = 100) or into the uterine horn ipsilateral to the recorded LF (n = 100). No effect of deeper AI on P/AI was found (P = 0.57). Therefore, the LF diameter at TAI and the occurrence of estrus can be used as selection criteria to identify cows with greater odds of pregnancy to receive sex-sorted sperm in TAI programs. In addition, performing TAI with sex-sorted sperm deeper into the uterus did not alter the pregnancy results.
