We used intracortical microstimulation to investigate the lateral premotor cortex and neighboring areas in 14 hemispheres of owl monkeys, focusing on the somatotopic distribution of evoked movements, thresholds for forelimb movements, and the relative representation of proximal and distal forelimb movements. We elicited movements from the dorsal and ventral premotor areas (PMD, PMV), the caudal and rostral divisions of primary motor cortex (Mlc, Mlr), the frontal eye field (FEF), the dorsal oculomotor area (OMD; area 8b), the supplementary motor area (SMA), and somatosensory cortex (areas 3a and 3b). Area PMD was composed of architectonically distinguishable caudal and rostral subdivisions (PMDc, PMDr). Stimulation of PMD elicited movements of the hindlimb, forelimb, neck and upper trunk, face, and eyes. Hindlimb and forelimb movements were represented in the caudalmost part of PMDc. Face, neck, and eye movements were represented in the lateral and rostral parts of PMDc and in PMDr. Stimulation of PMV elicited forelimb and orofacial movements, but not hindlimb movements. Both proximal and distal forelimb movements were elicited from PMDc and PMV, although PMD stimulation elicited mainly shoulder and elbow movements, while PMV stimulation evoked primarily wrist and digit movements. Distal movements were evoked more frequently from PMV than from Mlr or Mlc. Across cases, the median forelimb thresholds for PMDc and PMV were 60 and 36 μA, respectively, values that differ significantly from each other and from the value of 11 μA obtained for Mlr. Our observations indicate that premotor cortex is much more responsive to electrical stimulation than commonly thought, and contains a large territory from which eye movements can be elicited. These results suggest that in humans, much of the electrically excitable cortex located on the precentral gyrus, including cortex sometimes considered part of the frontal eye field, is probably homologous to the premotor cortex of nonhuman primates. © 1996 Wiley-Liss, Inc.

Movement representation in the dorsal and ventral premotor areas of owl monkeys: a microstimulation study.

DNA from 80 Duchenne (DMD) and 15 Becker (BMD) index patients was analyzed with 12 genomic probes and the total cDNA. Deletions were detected in 24 DMD (30%) and 10 BMD patients (67%) by genomic probes alone, mostly p20, pXJ, and/or pERT87. All deletions were confirmed by cDNA probes, and an additional 29 DMD deletions were detected, resulting in a total of 63/95 deletions (66%). The majority of the deletions are localized between kb 6.7 and 9.7 of the cDNA; a smaller group, between kb 0.5 and 3.5. Of the deletions, 90% are detected by the three cDNA probes 1–2a, 7, and 8. This can be applied to strategies for carrier detection and prenatal diagnosis. The order of 13 exon-containing HindIII fragments in the region between probes 7 and 9–10, where most of the deletions are found, could be defined. The deletion patterns in DMD and BMD patients are different and well in accordance with the “reading frame theory” of Monaco and coworkers. Thus our findings indicate that a DMD or BMD phenotype may be predicted according to the breakpoint position and the number of deleted exons.

Molecular deletion patterns in Duchenne and Becker type muscular dystrophy

Recent advances in molecular genetics have led to the isolation of the gene defective in patients with Duchenne and Becker's muscular dystrophy and the characterization of its protein product, dystrophin. In this communication, the developments culminating in the identification of the Duchenne muscular dystrophy locus are reviewed. The practical applications of this research and pitfalls that limit prenatal diagnosis and carrier detection are discussed.

Molecular biology of Duchenne and Becker's muscular dystrophy: clinical applications.

INTRODUCTION : Mutations in the gene encoding the free radical scavenging enzyme CuZn-superoxide dismutase have been associated with amyotrophic lateral sclerosis (ALS). Ninety-eight mutations have been found worldwide in patients with ALS, all but one showing a dominant pattern of inheritance. The exception is the D90A mutation which in Finland, northern Norway and northern Sweden exists with an allele frequency of 1-2.5% and is in these regions associated with ALS inherited as a recessive trait. METHODS AND RESULTS : In this study we searched for the D90A CuZn-SOD mutation in different ethnic populations of the Russian Federation and found the D90A mutation not only in locations close to the Scandinavian peninsula but also in remote populations in Asia. CONCLUSION : The finding makes the D90A mutation the most prevalent CuZn-SOD mutation globally and has implications for interpreting the recent reports of D90A-heterozygous ALS patients in North America and Europe.

Recently there have been considerable advances made in the treatment of multiple sclerosis. For the first time since its initial clinical description in the 1800s, there are now available several medications which unequivocally exert favourable clinical effects through the lowering of the biological activity of the human illness. The therapeutic efficacy of IFN-β preparations seems particularly well established in this regard on the basis of five large, independent, trials of this agent. These trials have demonstrated remarkably consistent reductions in both attack rates and disability levels using a combination of clinical and magnetic resonance imaging outcome measures.
