This book describes the present state of knowledge regarding the impact of cyanobacteria on health through the use of water. It considers aspects of risk management and details the information needed for protecting drinking water sources and recreational water bodies from the health hazards caused by cyanobacteria and their toxins. It also outlines the state of knowledge regarding the principal considerations in the design of programmes and studies for monitoring water resources and supplies and describes the approaches and procedures used.
The development of this publication was guided by the recommendations of several expert meetings concerning drinking water (Geneva, December 1995; Bad Elster, June
1996) and recreational water (Bad Elster, June 1996; St Helier, May 1997). An expert meeting in Bad Elster, April 1997, critically reviewed the literature concerning the toxicity of cyanotoxins and developed the scope and content of this book. A draft manuscript was reviewed at an editorial meeting in November 1997, and a further draft was
reviewed by the working group responsible for updating the Guidelines for Drinkingwater Quality in March 1998.

Toxic cyanobacteria in water: a guide to their public health consequences, monitoring and management.

Increasingly, harmful algal blooms (HABs) are being reported worldwide due to several factors, primarily eutrophication, climate change and more scientific monitoring. All but cyanobacteria toxin poisonings (CTPs) are mainly a marine occurrence. CTPs occur in fresh (lakes, ponds, rivers and reservoirs) and brackish (seas, estuaries, and lakes) waters throughout the world. Organisms responsible include an estimated 40 genera but the main ones are Anabaena, Aphanizomenon, Cylindrospermopsis, Lyngbya, Microcystis, Nostoc, and Oscillatoria (Planktothrix). Cyanobacteria toxins (cyanotoxins) include cytotoxins and biotoxins with biotoxins being responsible for acute lethal, acute, chronic and sub-chronic poisonings of wild/domestic animals and humans. The biotoxins include the neurotoxins; ana-toxin-a, anatoxin-a(s) and saxitoxins plus the hepatotoxins; microcystins, nodularins and cylindrospermopsins. Health Effects of Toxin-Producing Cyanobacteria: “The CyanoHABs”

Cyanobacteria possess many adaptations to develop population maxima or “blooms” in lakes and reservoirs. A potential consequence of freshwater blooms of many cyanobacterial species is the production of potent toxins, including the cyclic hepatotoxins, microcystins (MCs). Approximately 70 MC variants have been isolated. Their toxicity to humans and other animals is well studied, because of public health concerns. This review focuses instead on the production and degradation of MCs in freshwater environments and their effects on aquatic organisms. Genetic research has revealed the existence of MC-related genes, yet the expression of these genes seems to be regulated by complex mechanisms and is influenced by environmental factors. In natural water bodies, the species composition of cyanobacterial communities and the ratio of toxic to nontoxic species and strains are largely responsible for total toxin production. Hepatotoxic Cyanobacteria: A Review of the Biological Importance of Microcystins in Freshwater Environments

Microcystis andNodularia species produce cyclic hepta- and pentapeptides, microcystins and nodularin, respectively, both containing the same unusual C20 amino acid, abbreviated Adda. Biosynthesis of nodularin fromNodularia and especially of Adda employs a pathway similar to that employed byMicrocystis for producting microcystins. Nearly 30 new microcystins have been isolated in our laboratory from cyanobacteria species and their structures assigned, largely employing tandem FAB mass spectrometry (FABMS/CID/MS). Acyclic peptides, some of them presumed precursors of nodularin and microcystins, have now been isolated and characterized. The numerous analogs identified or synthesized allow the identification of important parameters in a structure-activity relationship study.

Structure and biosynthesis of toxins from blue-green algae (cyanobacteria)

Cyanobacteria (blue-green algae) produce toxins that may present a hazard for drinking water safety. These toxins (microcystins, nodularins, saxitoxins, anatoxin-a, anatoxin-a(s), cylindrospermopsin) are structurally diverse and their effects range from liver damage, including liver cancer, to neurotoxicity. The occurrence of cyanobacteria and their toxins in water bodies used for the production of drinking water poses a technical challenge for water utility managers. With respect to their removal in water treatment procedures, of the more than 60 microcystin congeners, microcystin-LR (L, L-leucine; R, L-arginine) is the best studied cyanobacterial toxin, whereas information for the other toxins is largely lacking. In response to the growing concern about nonlethal acute and chronic effects of microcystins, the World Health Organization has recently set a new provisional guideline value for microcystin-LR of 1.0 microg/L drinking water. This will lead to further efforts by water suppliers to develop effective treatment procedures to remove these toxins. Of the water treatment procedures discussed in this review, chlorination, possibly micro-/ultrafiltration, but especially ozonation are the most effective in destroying cyanobacteria and in removing microcystins. However, these treatments may not be sufficient during bloom situations or when a high organic load is present, and toxin levels should therefore be monitored during the water treatment process. In order to perform an adequate human risk assessment of microcystin exposure via drinking water, the issue of water treatment byproducts will have to be addressed in the future.
