Urban greenness and plant species are key factors in shaping air microbiomes and reducing airborne pathogens.

The benefits of regular exercise include improved physical and mental health. The school gym is a particular micro-environment where students perform intensive physical training. The question is if there is an increased risk of microbiological contamination. This preliminary work studied the exposure of students to bacterial aerosol (BA) in a highschool gym located in an urban area of Southern Poland. A sampling of BA was undertaken with an Andersen six-stage impactor (ANDI). BA was identified using API (analytical profile index) tests. The BA concentrations were expressed as Colony Forming Units (CFU) per cubic metre of air. The results showed that before gym classes (BGC), the concentration of BA was 4.20 × 102 ± 49.19 CFU/m3, while during gym classes (DGC), the level of BA more than doubled (8.75 × 102 ± 121.39 CFU/m3). There was also an increase in the respirable fraction of BA (particles less than 3.3 µm). Before the start of the sports activities, respirable fraction accounted for 30% of the BA, while during physical education classes, this share increased to over 80%. Identification of BA species showed that the dominant group of bacteria in the indoor air of the gym BGC was Gram-positive rods (61%) and for DGC it was Gram-positive cocci (81%). We detected that one bacteria strain (Corynebacterium striatum) was classified into risk group 2 (RG2) according to Directive 2000/54/EC. Additionally, multi-antibiotic resistance (MAR) showed that among the isolated airborne bacteria, the highest antibiotic resistance was demonstrated by Staphylococcus epidermis (isolated DGC) and Pseudomonas sp. (isolated BGC). The quantitative and qualitative information on microbiological air quality (MIAQ) in the school gym indicates that the actions to improve indoor physical activity spaces are recommended.

This study aims to identify the recent trends of research on the topic through a systematic literature review following the preferred reporting items for systematic reviews and meta-analyses (PRISMA) methodology. A total of 171 articles published in the period 2015–2020 were selected and analyzed. Results show that there is a worldwide growing research interest in this subject, dispersed in a wide variety of scientific journals. A textometric analysis using the IRaMuTeQ software revealed four clusters of topics in the sampled articles: physicochemical pollutants, design and management of infrastructures, environmental control measures, and microbiological contamination. The studies focus mainly on hospital facilities, but there is also research interest in primary care centers and dental clinics. The majority of the analyzed articles (85%) report experimental data, with the most frequently measured parameters being related to environmental quality (temperature and relative humidity), microbiological load, CO2 and particulate matter. Non-compliance with the WHO guidelines for indoor air quality is frequently reported. This study provides an overview of the recent literature on this topic, identifying promising lines of research to improve indoor air quality in healthcare facilities.

Microorganisms existing in airborne fine particulate matter (PM2.5) have key implications in biogeochemical cycling and human health. In this study, PM2.5 samples, collected in the typical basin cities of Xi’an and Linfen, China, were analyzed through high-throughput sequencing to understand microbial seasonal variation characteristics and ecological functions. For bacteria, the highest richness and diversity were identified in autumn. The bacterial phyla were dominated by Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes. Metabolism was the most abundant pathway, with the highest relative abundance found in autumn. Pathogenic bacteria (Pseudomonas, Acinetobacter, Serratia, and Delftia) were positively correlated with most disease-related pathways. Besides, C cycling dominated in spring and summer, while N cycling dominated in autumn and winter. The relative abundance of S cycling was highest during winter in Linfen. For fungi, the highest richness was found in summer. Basidiomycota and Ascomycota mainly constituted the fungal phyla. Moreover, temperature (T) and sulfur dioxide (SO2) in Xi’an, and T, SO2, and nitrogen dioxide (NO2) in Linfen were the key factors affecting microbial community structures, which were associated with different pollution characteristics in Xi’an and Linfen. Overall, these results provide an important reference for the research into airborne microbial seasonal variations, along with their ecological functions and health impacts.

Seasonal Variation Characteristics of Bacteria and Fungi in PM2.5 in Typical Basin Cities of Xi’an and Linfen, China

Bacterial contamination of air may have human health implications by the transmission of potential human pathogens. Therefore, assessment of air bacterial abundance and composition in different built environment is essential. Jawaharlal Nehru University health centre (UHC) is a primary healthcare setting providing need-based medication to university students. Using active air sampling method, we collected eight air samples from the indoor and outdoor area of UHC across four different seasons. The total genomic DNA was extracted from the air samples and subjected to 16S rRNA gene-based next-generation sequencing. We performed the taxonomic classification along with comparative analysis of air bacterial communities. This study revealed that Proteobacteria, Actinobacteria, Bacteroidetes and Firmicutes are the dominant phyla in the sampled air. Overall, the air bacterial composition in our studied samples was comparatively simple; only ten taxonomic families accounting for ~75% of the total sequences determined. We also observed ESKAPE pathogens in the air metagenomes in a low percentage (4.42%), which were dominated by Pseudomonas, Acinetobacter and Staphylococcus. Proteobacteria, Actinobacteria and Firmicutes showed significant correlation with PM2.5. We suggest that routine air monitoring and microbiological survey is essential for air quality standards and potential human pathogens detection in health care settings. It is the first report from India to uncover the temporal dynamics of air bacterial communities in UHC using Illumina MiSeq (PE300) sequencing and Quantitative Insights into Microbial Ecology (QIIME).

Temporal Dynamics of Air Bacterial Communities in a University Health Centre Using Illumina MiSeq Sequencing

Staphylococci species are the major constituents of infectious bioaerosols, particularly methicillin-resistant Staphylococci (MRS) have serious health impacts. Here, the bacterial burden was quantified, especially prevalence of MRS in bioaerosols collected from indoors of Dr. B.R. Ambedkar Central Library (DBRACL) and Central Laboratory Animal Resources (CLAR) of Jawaharlal Nehru University, New Delhi, India. Air samplings from DBRACL and CLAR were done using the settle plate method and SKC biosampler, respectively. This study showed a maximum 6757 CFU/m2/hr of bacterial load in the DBRACL reading room, while unacceptable bacterial loads (> 1000 CFU/m3 of air) at different sites of CLAR. Further, at both the sampling sites the predominance of coagulase negative Staphylococci (CNS) was observed. A total 22 and 35 Staphylococci isolates were isolated from DBRACL and CLAR bioaerosols, respectively. Majority (16/22) of the Staphylococcal isolates from DBRACL belonged to human-associated Staphylococci where S. haemolyticus (5/22) was the most dominating species. However, in CLAR facility centre, animal-associated Staphylococci (19/35) were dominating, where S. xylosus (12/35) was the most dominating species. Further, antibiotic sensitivity tests revealed 41% MRS and 73% multidrug resistant (MDR) among airborne Staphylococci from DBRACL indoor bioaerosols. Similarly, in CLAR facility, approximately, 66% Staphylococci isolates were methicillin resistant, out of which 2 isolates showed high MIC value ≥ 16 μg/mL. Further, we confirmed the presence of 49% multidrug resistant Staphylococci in the indoor air of CLAR facility. This study suggested that the exposure of workers and students in CLAR to such a high concentration of drug-resistant Staphylococci should not be undermined, as these bacterial concentrations are the direct representative of inhalable particulate matter (PM2.5) as per collection procedure. Simultaneously, passive sampling from DBRACL assessed the risks due to microbial contamination in particle agglomerates, which may deposit on the crucial surfaces such as wounds/ cuts or on the frequently used items.
