Abstract: Several recent reportsI4 furnish a sound basis for the concept of functional interdependence of Sertoli cells and germ cells during mammalian spermatogenesis. The question that arises, however, as a corollary, namely whether stagedependent bidirectional transfer of information molecules such as proteins occurs, has not been addressed so far. An earlier report from this laboratory based on an autoradiographic studyS revealed characteristic regionalization of protein synthesis in the Sertoli cell cytoplasm in specific relation to defined germ cell associations during ontogeny of spermatogenesis in the colony-bred rat. Furthermore, analysis by single dimensional gel electrophoresis6 of the peptide profiles of Sertoli cells and of germ cells as a function of ontogeny showed the following: stage-dependence of the profiles; the presence of peptides unique to a given stage of ontogeny; and the cyclic occurrence, during ontogeny, of certain peptides. These data are consistent with the concept of intercellular communication. Parallel ultrastructural studies have provided supportive evidence.' In a further effort to document cell-to-cell communication, pulse labeling followed by two-dimensional gel electrophoretic analysis of labeled proteins was performed.
